The endoplasmic reticulum(ER)membrane protein complex(EMC)is responsible for monitoring the biogenesis and synthetic quality of membrane proteins with tail-anchored or multiple transmembrane *** EMC subunit EMC6 is one of the core members of EMC and forms an enclosed hydrophilic vestibule in cooperation with *** studies demonstrating that deletion of EMC3 led to rhodopsin mislocalization in rod photoreceptors of mice,the precise mechanism leading to the failure of rhodopsin trafficking remains ***,we generated the first rod photoreceptor-specific knockout of Emc6(RKO)and cone photoreceptor-specific knockout of Emc6(CKO)mouse *** of Emc6 in rod photoreceptors led to progressive shortening of outer segments(OS),impaired visual function,mislocalization and reduced expression of rhodopsin,and increased gliosis in rod *** addition,CKO mice displayed the progressive death of cone photoreceptors and abnormal localization of cone opsin ***,proteomics analysis of the RKO mouse retina illustrated that several cilium-related proteins,particularly anoctamin-2(ANO2)and transmembrane protein 67(TMEM67),were significantly down-regulated prior to OS *** rod photoreceptor cilia and mislocalized membrane disc proteins were evident in RKO *** data revealed that in addition to monitoring the synthesis of rhodopsin-dominated membrane disc proteins,EMC6 also impacted rod photoreceptors'ciliogenesis by regulating the synthesis of membrane proteins associated with cilia,contributing to the mislocalization of membrane disc proteins.

Parasitic plants and their hosts communicate through haustorial *** deficiency is a common stress for plants,yet little is known about whether and how host plants and parasites communicate during adaptation to such nutrient *** this study,we used transcriptomics and proteomics to analyze how soybean(Glycine max)and its parasitizing dodder(Cuscuta australis)respond to nitrate and phosphate deficiency(-N and-P).After-N and-P treatment,the soybean and dodder plants exhibited substantial changes of transcriptome and proteome,although soybean plants showed very few transcriptional responses to-P and dodder did not show any transcriptional changes to either-N ***,large-scale interplant transport of mRNAs and proteins was *** the mobile mRNAs only comprised at most 0.2%of the transcriptomes,the foreign mobile proteins could reach 6.8%of the total proteins,suggesting that proteins may be the major forms of interplant ***,the interplant mobility of macromolecules was specifically affected by the nutrient regimes and the transport of these macromolecules was very likely independently *** study provides new insight into the communication between host plants and parasites under stress conditions.

Background： An important product of mevalonate pathway is downstream synthesis of isoprenoid unitsthat has long been implicated in development and progression of tumor. It has been speculated thatinhibition of protein prenylation might be therapeutically beneficial. The objective of current study wasto evaluate antitumor potential of a novel therapeutic combination of mevalonate pathway inhibitors, FTI-277 and alendronate. We also examined differentially expressed proteins in response to treatment usingproteomics approach.

Bioactive peptides have various excellent biological activities and serve as functional foods to prevent chronic diseases in the human body. This article investigated the regulatory effect of egg white peptides (EPs) on the intestine barrier of young mice with colitis. The results showed that the intake of EPs could protect the intestines from inflammation damage. Besides， the expression of tight junction (TJ) proteins and mucin is upregulated. Markedly， the intake of EPs can increase the concentration of amino acids in the serum of young mice， which is crucial for nutritional supplementation during intestinal inflammation process. proteomics analysis implied that EPs can regulate protein expression in the intestine， involving multiple inflammatory pathways including PI3K-AKT and MAPK signaling pathway. This demonstrated the health benefits of bioactive peptides and provides a theoretical basis for the development of animal derived proteins as functional foods.

Screen biomarkers of human lung squamous carcinoma by serological proteome analysis of HTB-182

Carcinogenesis of lung squamous carcinoma is a complex process involving multiple events and steps. At present, there are very few special lung cancer molecular markers for an 'early-stage' diagnosis and prognosis evaluation. To identify tumor-associated antigens, serological proteome analysis (SERPA) of human lung squamous carcinoma cell line HTB-182 are performed. We characterized sixteen differentially expressed proteins which react with lung squamous carcinoma patient sera while not react with control sera. Some of these candidate lung squamous carcinoma-associated antigens were metabolic enzymes, such as triosephosphatase isomerase (TPIS). Some proteins were involved in the regulation of cell cycle and signal transduction. The results will provide scientific foundation for screening the molecular biomarkers used to diagnose and treat lung squamous carcinoma, as well as to elevate the patient's prognosis and provide new clue for the research of lung squamous carcinogenic mechanism. Thus, SERPA represents a valuable approach for the identification of differentially expressed proteins, which might be used as markers for the diagnosis and prognosis of lung squamous carcinoma.

Streptococcus suis serotype 2(*** 2)is a zoonotic pathogen that clinically causes severe swine and human infections(such as meningitis,endocarditis,and septicemia).In order to cause widespread diseases in different organs,*** 2 must colonize the host,break the blood barrier,and cause exaggerated *** the last few years,most studies have focused on a single virulence factor and its influences on the *** vesicles(MVs)can be actively secreted into the extracellular environment contributing to bacteria-host ***-negative bacteria-derived outer membrane vesicles(OMVs)were recently shown to activate host Caspase-11-mediated non-canonical inflammasome pathway via deliverance of OMV-bound lipopolysaccharide(LPS),causing host cell ***,little is known about the effect of the MVs from *** 2(Gram-positive bacteria without LPS)on cell ***,we investigated the molecular mechanism by which *** 2 MVs participate in endothelial cell *** this study,we used proteomics,electron scanning microscopy,fluorescence microscope,Western blotting,and bioassays,to investigate the MVs secreted by *** ***,we demonstrated that *** 2 secreted MVs with an average diameter of 72.04 nm,and 200 proteins in MVs were ***,we showed that MVs were transported to cells via mainly dynamin-dependent *** *** 2 MVs activated NLRP3/Caspase-1/GSDMD canonical inflammasome signaling pathway,resulting in cell pyroptosis,but it did not activate the Caspase-4/-5 *** importantly,endothelial cells produce large amounts of reactive oxygen species(ROS)and lost their mitochondrial membrane potential under induction by *** 2 *** results in this study suggest for the first time that MVs from *** 2 were internalized by endothelial cells via mainly dynamin-dependent endocytosis and might promote NLRP3/Caspase-1/GSDMD pathway by mitochondrial damage,which produced mtDNA and ROS under induction,

The characterization of protein lactylation in relation to cardiac metabolic reprogramming in neonatal mouse hearts

In mammals,the neonatal heart can regenerate upon injury within a short time after birth,while adults lose this *** reprogramming has been demonstrated to be critical for cardiomyocyte proliferation in the neonatal ***,we reveal that cardiac metabolic reprogramming could be regulated by altering global protein *** performing 4D label-free proteomics and lysine lactylation(Kla)omics analyses in mouse hearts at postnatal days 1,5,and 7,2297 Kla sites from 980 proteins are identified,among which 1262 Kla sites from 409 proteins are *** clustering analysis reveals that the proteins with altered Kla sites are mainly involved in metabolic *** expression and Kla levels of proteins in glycolysis show a positive correlation while a negative correlation in fatty acid ***,we verify the Kla levels of several differentially modified proteins,including ACAT1,ACADL,ACADVL,PFKM,PKM,and ***,our study reports a comprehensive Kla map in the neonatal mouse heart,which will help to understand the regulatory network of metabolic reprogramming and cardiac regeneration.

Mass spectrometry based proteomics profiling of human monocytes

Human monocyte is an important cell type which is involved in various complex human diseases. To better understand the biology of human monocytes and facilitate further studies, we developed the first comprehensive proteome knowledge base specifically for human monocytes by integrating both in vivo and in vitro datasets. The top 2000 expressed genes from in vitro datasets and 779 genes from in vivo experiments were integrated into this study. Altogether, a total of 2237 unique monocyte-expressed genes were cataloged. Biological functions of these monocyte-expressed genes were annotated and classified via Gene Ontology （GO） analysis. Furthermore, by extracting the overlapped genes from in vivo and in vitro datasets, a core gene list including 541 unique genes was generated. Based on the core gene list, further gene-disease associations, pathway and network analyses were performed. Data analyses based on multiple bioinformatics tools produced a large body of biologically meaningful information, and revealed a number of genes such as SAMHDI, G6PD, GPD2 and EN01, which have been reported to be related to immune response, blood biology, bone remodeling, and cancer respectively. As a unique resource, this study can serve as a reference map for future in-depth research on monocytes biology and monocyte-involved human diseases.
