Intestinal anti-inflammatory activity of Ground Cherry(Physalis angulata L.)standardized CO2 phytopharmaceutical preparation

AIM To investigate the effects of Ground Cherry(Physalis angulata L.)standardized supercritical CO_2 extract in trinitrobenzenesulphonic acid(TNBS)model of rat intestinal *** The animals were divided into groups that received vehicle or *** extract(PACO_2)orally at the doses 25,50 and 100 mg/kg daily by 5 d before TNBS *** effects of PACO_2 were assessed by macroscopic analysis,biochemical determinations of the levels of myeloperoxidase(MPO),alkaline phosphatase(ALP),glutathione and cytokines(such as INF-γ,IL-1β,IL-6,IL-10 and TNF-α),gene expression evaluation(including Hsp70,heparanase,NF-κB,mitogenactivated protein kinases(Mapk)1,3,6 and 9,and the mucins genes Muc 1,2,3 and 4)and histopathological studies using optical,and electronic(transmission and scanning)*** PACO2 extract promoted a significant reduction in MPO and ALP activities,reducing oxidative stress and neutrophil *** effects were accompanied by significant reduction of colonic levels of IFN-γand IL-6 and down-regulation of heparanase,Hsp70,Mapk3,Mapk9,Muc1 and Muc2 genes expression when compared with TNBS-control *** addition,protective effects were also evidenced by reduced neutrophil infiltration,recovery of cell architecture and replacement of mucin by histopathological and ultrastructural *** Physalis angulata supercritical CO2 extract is an intestinal anti-inflammatory product that modulates oxidative stress,immune response and expression of inflammatory mediators,with potentially utility for treating inflammatory bowel disease.

WHO strategy on traditional medicine is based on security and quality of phytomedicine. Commonly adulterated affecting mostly metabolic and sexual dysfunction drugs. Control quality of those phytomedicines requires development of strategy and techniques applicable to them. Among the techniques, Reverse Phase Liquid Chromatography is the most used and has been developed in these studies to assess a protocol to characterize Gardenia aqualla leaves extract. The method consists in determining chromatographic conditions using organic and pH gradient models based on water and acetonitrile combined with pH modifiers made up of formic acid (AF) and ammonium hydroxide (NH3). Results show that extracts contain mainly acidic compounds quickly eluted by NH3 and more retained by AF. Optimal pH range for separation is 3 - 7 corresponding to 1.59 mM of NH3 and 6.55 mM of AF. In these conditions, elution of many polar compounds could be effective using a C18 based-deactivated column in a short period of time.

Activated pancreatic stellate cells(PSCs)have been widely accepted as a key precursor of excessive pancreatic fibrosis,which is a crucial hallmark of chronic pancreatitis(CP)and its formidable associated disease,pancreatic cancer(PC).Hence,anti-fibrotic therapy has been identified as a novel therapeutic strategy for treating CP and PC by targeting *** of the anti-fibrotic agents have been limited to phaseⅠ/Ⅱclinical trials involving vitamin analogs,which are abundant in medicinal plants and have proved to be promising for clinical *** use of phytomedicines,as new anti-fibrotic agents,has been applied to a variety of complementary and alternative *** aim of this review was to present a focused update on the selective new potential anti-fibrotic agents,including curcumin,resveratrol,rhein,emodin,green tea catechin derivatives,metformin,eruberin A,and ellagic acid,in combating PSC in CP and PC *** aimed to describe the mechanism(s)of the phytochemicals used,either alone or in combination,and the associated molecular *** of them were tested in PC models with similar mechanism of actions,and curcumin was tested *** research may explore the issues of bioavailability,drug design,and nano-formulation,in order to achieve successful clinical outcomes with promising activity and tolerability.

Objective: Boswellia papyrifera (Del.) Hochst and Ruta chalepensis L. are aromatic plants used in the Republic of Djibouti, both as food and medicine. The importance of essential oils of these two species in the treatment of certain pathologies such as inflammations and infections has led to an assessment of their composition in volatile organic compounds and their hepato-protective properties on rats poisoned by carbon tetrachloride (CCl4). Methods: The chemical composition of essential oils studied was determined by GC-MS. The hepato-protective properties of the essential oils of Boswellia papyrifera and Ruta chalepensis were assessed through inhibition of hepato toxicity in rats by CCl4 poisoning. The hepato-protection of essential oils was estimated by measuring biochemical and hematological parameters. Results: Essential oils of Boswellia papyrifera and Ruta chalepensis reduced the blood level of transaminases and triglycerides at the dependent dosage, and restored liver proteins (0.27 g/l) to levels similar to those rats not poisoned by CCl4. Blood levels of uric acid, urea, creatinine and HDL have also been restored to values similar to those of non-poisoned rats. The chemical composition of the essential oils studied shows that Boswellia papyrifera contains incensole acetate (43.76%) and isoincensole (18.42%), and that Ruta chalepensis contains menthyl acetate (29.8%) and piperitone (22.22%). Conclusion: The studies carried out have shown that the essential oils of Boswellia papyrifera and Ruta chalepensis have a hepato-protective potential similar to that of sylimarin (reference hepato-protective substances). The results show that the essential oils of Boswellia papyrifera and Ruta chalepensis could present good prospects in the formulation of phytomedicines for the man
