Effects of dietary forage to concentrate ratio and wildrye length on nutrient intake, digestibility, plasma metabolites, ruminal fermentation and fecal microflora of male Chinese Holstein calves

Twenty-eight male, weaned Chinese Holstein calves（（156.8±33.4） kg） were used to investigate the effects of dietary forage to concentrate ratio（F：C） and forage length on nutrient digestibility, plasma metabolites, ruminal fermentation, and fecal microflora. animals were randomly allocated to four treatments in a 2×2 factorial arrangement： whole-length forage（WL） with low F：C（50：50）; WL with high F：C（65：35）; short-length forage（SL） with high F：C（65：35）; and SL with low F：C（50：50）. Chinese wildrye was used as the only forage source in this trial. The grass in the SL treatments was chopped using a chaff cutter to achieve small particle size（-50% particles 〉19 mm）. Dry matter intake（DMI） and organic matter（OM） intake was increased by increasing both F：C（P〈0.01） and forage length（FL）（P〈0.05）, while acid detergent fiber（ADF） and neutral detergent fiber（NDF） intakes were only increased by increasing the F：C（P〈0.01）. The digestibility of NDF was increased as the FL increased（P〈0.01）, and it was also affected by interaction between F：C and FL（P〈0.05）. Cholesterol（CHO）（P〈0.01）, leptin（LP）（P〈0.05）, and growth hormone（GH）（P〈0.01） concentrations in plasma were increased as dietary F：C increased. A significant increase in plasma triglyceride（TG）（P〈0.01）, insulin（INS）（P〈0.05）, and GH（P〈0.01） levels was observed with decreasing dietary FL. Ruminal p H values of calves fed with low F：C diets were significantly lower than those in high F：C treatment（P〈0.05）. Increasing the F：C enhanced ruminal acetic acid（P〈0.05） and acetic acid/propionic acid（P〈0.01）. Fecal Lactobacillus content was significantly higher, while Escherichia coli and Salmonella contents were significantly lower in WL and high F：C groups（P〈0.05）. Lower fecal scores（higher diarrhea rate） were observed in calves fed with SL hay compared to WL hay（P〈0.05）. Glucose supplementation improves intestinal amino acid transport and muscle amino acid pool in pigs during chronic cold exposure

SFRP2 affects prenatal muscle development and is regulated by micro RNA-1/206 in pigs

Secreted frizzled-related protein 2 （SFRP2）, a member of the SFRPs family, is associated with cell growth and differentiation in myogenesis. Our previous study suggested that SFRP2 was a potential target of microRNA （miRNA）-I/206, which was considered as myomiRs. To further explore the biological function and regulation mechanisms of the SFRP2 gene in porcine skeletal muscle development, we first analyzed the sequence structure of the porcine SFRP2 gene. Subsequently, we detected its tissue distribution in adult Tongcheng pigs （a Chinese indigenous breed） and investigated its dynamic expression in developmental skeletal muscle （13 prenatal and 7 postnatal time points）in Tongcheng pigs. An interaction analysis between SFRP2 and myomiRs was also performed. The results showed that the expression pattern of the SFRP2 varied greatly across diverse tissues. It exhibited abundant expression in prenatal skeletal muscle and peaked at 55 days post coitus （E55）, and had a lower expression in postnatal skeletal muscle, indicating that the SFRP2 gene might affect porcine embryonic skeletal muscle development. Co-expression analysis revealed that the expression levels of SFRP2 correlated negatively with miRNA-1 （t=-0.570, P-value=0.009） and miRNA-206 （r=-0.546, P-value=0.013）, but positively with SFRP1 （r=0.613, P-value=0.004）. The bioinformatics analysis and dual luciferase assay verified that the SFRP2 was a putative target of miRNA-1/206 in pigs. Therefore, this study is helpful for understanding the biological function and mo- lecular regulation of the SFRP2 gene during porcine skeletal muscle development.

Nonalcoholic fatty liver disease(NAFLD) is characterized by hepatic steatosis and insulin resistance and there are currently no approved drugs for its *** of mTOR complex1(mTORCl) and subsequent impairment of the transcription factor EB(TFEB)-mediated autophagy-lysosomal pathway(ALP) are implicated in the development of ***,agents that augment hepatic TFEB transcriptional activity may have therapeutic potential against *** objective of this study was to investigate the effects of nuciferine,a major active component from lotus leaf,on NAFLD and its underlying mechanism of *** we show that nuciferine activated ALP and alleviated steatosis,insulin resistance in the livers of NAFLD mice and palmitic acid-challenged hepatocytes in a TFEB-dependent *** investigation revealed that nuciferine interacts with the Ragulator subunit hepatitis B X-interacting protein and impairs the interaction of the Ragulator complex with Rag GTPases,thereby suppressing lysosomal localization and activity of mTORC1,which activates TFEB-mediated ALP and further ameliorates hepatic steatosis and insulin *** present results indicate that nuciferine may be a potential agent for treating NAFLD and that regulation of the mTORCl-TFEB-ALP axis could represent a novel pharmacological strategy to combat NAFLD.

Secretory pore-forming proteins(PFPs) have been identified in organisms from all kingdoms of life. Our studies with the toad species Bombina maxima found an interaction network among aerolysin family PFPs(af-PFPs) and trefoil factors(TFFs). As a toad af-PFP, Bm ALP1 can be reversibly regulated between active and inactive forms, with its paralog Bm ALP3 acting as a negative regulator. Bm ALP1 interacts with Bm TFF3 to form a cellular active complex called βγ-CAT. This PFP complex is characterized by acting on endocytic pathways and forming pores on endolysosomes, including stimulating cell macropinocytosis. In addition, cell exocytosis can be induced and/or modulated in the presence of βγ-CAT. Depending on cell contexts and surroundings, these effects can facilitate the toad in material uptake and vesicular transport, while maintaining mucosal barrier function as well as immune defense. Based on experimental evidence,we hereby propose a secretory endolysosome channel(SELC) pathway conducted by a secreted PFP in cell endocytic and exocytic systems, with βγ-CAT being the first example of a SELC protein. With essential roles in cell interactions and environmental adaptations, the proposed SELC protein pathway should be conserved in other living organisms.

Vitamin K alleviates bone calcium loss caused by Salmonella Enteritidis through carboxylation of osteocalcin

Background:The present study aimed at evaluating the effect of vitamin K(VK)supplementation on bone health of laying hens challenged by Salmonella ***:A total of 8032-week-old double negative salmonella-free brown-egg laying hens were randomly assigned to 4 treatments with 20 replicates each(1 bird per replicate)according to a 2×2 factorial design with 2 dietary VK supplementation levels[0 mg/kg(VK0)vs 2 mg/kg VK(VK2)and 2 challenge treatments[Salmonella Enteritidis(SE)vs physiological saline solution(PS)].During the last 3 days of week 43 of age,birds of both VK treatments were either orally challenged with 1.0 mL suspension of 109 cfu/mL *** daily or received the same volume of ***:The laying rate,daily egg mass,tibia strength,CT,cOC and cOC/(cOC+ucOC)of VK2 treatment increased(Pinteraction(P<0.05)between SE challenge and VK levels with regard to tibia strength and serum Ca *** week 42,serum CT was positively correlated with cOC(R=0.99,P=0.009);at week 44,tibia strength was positively correlated with BMD(R=0.95,P=0.045),but negatively correlated with medullary area(R=−0.98,P=0.018).Conclusions:VK(2 mg/kg)supplementation to diets of laying hens can enhance bone strength under challenge situations with Salmonella *** area has proven to be a sensitive biomarker for bone calcium loss caused by SE infection.

Simulation of gene pyramiding in Drosophila melanogaster

Gene pyramiding has been successfully practiced in plant breeding for developing new breeds or lines in which favorable genes from several different lines were integrated. But it has not been used in animal breeding, and some theoretical investigation and simulation analysis with respect to its strategies, feasibility and efficiency are needed before it can be implemented in animals. In this study, we used four different pure lines of Drosophila melanogaster, each of which is homozygous at a specific mutant gene with a visible effect on phenotype, to simulate the gene pyramiding process and analyze the duration and population size required in different pyramiding strategies. We finally got the ideal individuals, which are homozygous at the four target genes simultaneously. This study demonstrates that gene pyramiding is feasible in animal breeding and the interaction between genes may affect the final results.
