Currently,colorectal cancer(CRC)represents the third most common malignancy and the second most deadly cancer worldwide,with a higher incidence in developed *** other solid tumors,CRC is a heterogeneous genomic disease in which various alterations,such as point mutations,genomic rearrangements,gene fusions or chromosomal copy number alterations,can contribute to the disease ***,because of its orderly natural history,easily accessible onset location and high lifetime incidence,CRC is ideally suited for preventive intervention,but the many screening efforts of the last decades have been compromised by performance limitations and low penetrance of the standard screening *** advent of next-generation sequencing(NGS)has both facilitated the identification of previously unrecognized CRC features such as its relationship with gut microbial pathogens and revolutionized the speed and throughput of cataloguing CRC-related genomic ***,in this review,we summarized the several diagnostic tools used for CRC screening in the past and the present,focusing on recent NGS approaches and their revolutionary role in the identification of novel genomic CRC characteristics,the advancement of understanding the CRC carcinogenesis and the screening of clinically actionable targets for personalized medicine.

Objective:To evaluate the protective effect of DNA vaccine with the gene encoding 55kDa antigen fragment of Pneumocystis carinii(***) against *** in ***:The fragment of the antigen within p55(p55-582) was *** recombinant plasmid was constructed based on the eukaryotic expression vector pcDNA3.1(+).BALB/c mice were used as experimental models to examine the immunogenicity of pcDNA3.1(+)-*** and RTPCR were used to evaluate the role of this kind of DNA ***:The results of western blot indicated that the recombinant DNA[pcDNA3.1(+)-p55-582]could be expressed correctly and had antigenicity in transfected COS-7 *** and RT-PCR showed that pcDNA3.1(+)- p55-582 elicited antibody production,stimulated lymphocyte proliferation and provided partial protection by reducing the *** ***:The data demonstrate that pcDNA3.1(+)-p55-582 might be potent vaccination that can afford the partial protection for the immunized animals.

Objective:To identify whether Canova medication changes TNF-α and IL-10 serum levels in mice infected with Trypanosoma cruzi Y ***:Animals were divided into five groups:non-treated infected animals(I); benznidazole-treated infected animals(Bz; 100 mg/kg body weight,single daily dose by gavage); Canova medication(CM) treated infected animals(CM;0.2 mL/animal,single daily dose by gavage); benznidazole- and Canova medication–treated infected animals with the above-mentioned dose(Bz+CM);and non-infected animals(C).TNF-α and IL-10 levels were determined in serum aliquots after 4,7,10,13,and 29 days of *** ELISA technique was employed with R&D System *** ***:A high increase in TNF-α and IL-10 levels occurred in the infected and CM-treated groups within the treatment employed on the 10 th day after infection,coupled with a IL-10 decrease on the 13 th day after infection when compared with the other experimental ***:CM may change the balance between plasma cytokine levels(TNF-α and IL-10) in mice infected with Y strain ***,with important consequences leading towards a more severe infection.

The majority of our knowledge of avian energetics is based on studies of birds from temperate and high latitudes. Using the largest existing sample of wild-caught Old World tropical species, we showed that birds from Southern Vietnam had lower basal metabolic rate （BMR） than temperate species. The strongest dissimilarity between tropical and temperate species was the low scaling exponent in the allometric relation between BMR and body mass in tropical birds （the regression slope was 0.573）. The passerine migrants to temperate and high latitudes had higher BMR than tropical sedentary passerines. Body mass alone accounted for 93% of the variation in BMR （body mass ranged from 5 to 252 g）. Contrary to some other studies, we did not find evidence besides the above mentioned that phylogeny, taxonomy, behavior, or ecology have a significant influence on BMR variation among tropical birds.

Effect of organic mineral supplementation in reducing oxidative stress in Holstein calves during short‑term heat stress and recovery conditions

Over 2 billion people worldwide are estimated to have nonalcoholic fatty liver disease(NAFLD),defined by excess hepatic ***,progression into nonalcoholic steatohepatitis(NASH)is characterized by the onset of liver inflammation following exacerbated steatosis.1 This suggests that reducing liver inflammation(*** lifestyle interventions involving exercise training)may come secondary to a reduction of ***,both the metabolic and inflammatory processes involved in NAFLD are under circadian control and could respond differently to exercise at different times of day.2 To investigate the time-of-day–dependent effect of exercise training on NAFLD amelioration in the early disease stages we trained high-fat high-cholesterol(HFHC)-fed APOE*3-Leiden cholesteryl ester transfer protein(CETP)mice during their early or late active *** mouse model was chosen because of its humanized lipid metabolism and its ability to develop all hallmarks of human NAFLD upon HFHC feeding.

Background Alzheimer’s disease(AD)is a progressive multifaceted neurodegenerative disorder for which no disease-modifying treatment *** is central to the pathology progression,with evidence suggesting that microglia-released galectin-3(gal3)plays a pivotal role by amplifying neuroinflammation in ***,the possible involvement of gal3 in the disruption of neuronal network oscillations typical of AD remains *** Here,we investigated the functional implications of gal3 signaling on experimentally induced gamma oscillations ex vivo(20-80 Hz)by performing electrophysiological recordings in the hippocampal CA3 area of wild-type(WT)mice and of the 5×FAD mouse model of *** addition,the recorded slices from WT mice under acute gal3 application were analyzed with RT-qPCR to detect expression of some neuroinflammation-related genes,and amyloid-β(Aβ)plaque load was quantified by immunostaining in the CA3 area of 6-month-old 5×FAD mice with or without Gal3 knockout(KO).Results Gal3 application decreased gamma oscillation power and rhythmicity in an activity-dependent manner,which was accompanied by impairment of cellular dynamics in fast-spiking interneurons(FSNs)and pyramidal *** found that the gal3-induced disruption was mediated by the gal3 carbohydrate-recognition domain and prevented by the gal3 inhibitor TD139,which also prevented Aβ42-induced degradation of gamma ***-more,the 5×FAD mice lacking gal3(5×FAD-Gal3KO)exhibited WT-like gamma network dynamics and decreased Aβplaque *** We report for the first time that gal3 impairs neuronal network dynamics by spike-phase uncoupling of FSNs,inducing a network performance ***,our findings suggest gal3 inhibition as a potential therapeutic strategy to counteract the neuronal network instability typical of AD and other neurological disorders encompassing neuroinflammation and cognitive decline.

Incidence and histopathology of encysted progenetic metacercaria of Clinostomum complanatum(Digenea:Clinostomidae) in Channa punctatus and its development in experimental host

Objective:To study the incidence of encysted progenetic metacercariae of Clinostomum complanatum(C. complanatum) in Channa punctatus(C. punctatus), associated histopathology and the experimental infection to laboratory chicken to obtain ovigerous adult ***:Live C. punctatus were brought from local fish market of Aligarh, India, dissected and examined on a monthly basis for the presence of C. complanatumcysts. For histochemistry, infected tissue sections with attached cysts were processed for haematoxylene and eosin staining. Cysts were aseptically fed to 4 day old leghorn chicken to obtain adult worms. Mechanically excysted metacercaria and the ovigerous adult worms were stained in carmine to prepare permanent ***:One year survey for the infection of encysted progenetic metacercaria of ***. punctatusrevealed the prevalence, intensity and abundance of 24.7%, 2.27and 0.608, respectively. Histopathology showed heavy infiltration of immune cells at the site of cyst attachment and some tissue damage was also evident. Following feeding to experimental chicken, about 41.07% of the encysted metacercariae were able to excyst and migrate back to bucco-pharyngeal region where they tenaciously attached and fed on blood, and transformed into ovigerous adult worms from 62 hours onwards of post ***:The parasite is potentially pathogenic to the host, and the availability of a suitable intermediate host can be a contributing factor for the occurrence of C. complanatummetacercaria either in the excysted or encysted form, indicating loose host specificity and zoonotic potential.

Context-dependent insect predation pressure on an avian ectoparasite

Context dependence arises when ecological relationships vary with the conditions under which they are observed. Context dependence of interactions involving parasites is poorly known, even if it is key to understanding host–parasite relationships and food web dynamics. This paper investigates to which extent predation pressure on an avian ectoparasite (Carnus hemapterus) is context-dependent. Based on a predator-exclusion experiment, predation pressure on C. hemapterus pupae in the host's nest for 3 years, and its variation between habitat types are quantified. Variation in precipitation and normalized difference vegetation index (NDVI) is also explored as a likely cause of context dependency. We hypothesize that predation pressure should fluctuate with such surrogates of food availability, so that inter-annual and intra-annual differences may emerge. The number of nests with significant reduction of pupae varied widely among years ranging from 24% to 75%. However, average pupae reduction in nests where a significant reduction occurred did not vary between years. No differences in predation rates between habitat types were detected. Precipitation and NDVI varied widely between years and NDVI was consistently lower around nests on cliffs than around nests on trees and farmhouses. Parallels were found between variation in predation pressure and precipitation/NDVI at a wide scale (highest predation the driest year, and much lower the 2 rainier ones), but not at the nest scale. This paper shows clear context-dependent insect predation pressure on an ectoparasite under natural conditions, and that such interaction changes in signs rather than magnitude between years. The causes for these variations require longer-term studies and/or well-designed, large-scale experiments.

Objective:To compare histopathology and PCR based detection in diagnosis of experimentally induced toxoplasmosis of RH human strain of the parasite in murine ***:A comparison of histopathology and PCR based detection was done to diagnose experimentally induced toxoplasmosis in ten inbred swiss albino mice after intraperitoneal inoculation of 100 tachyzoites of laboratory mantained human RH strain of the *** samples from lung,liver,spleen,brain,heart and kidney were taken and processed for histopathological examination while all the samples also were subjected to PCR,using primers directed to the multicopy of SAG 3 gene,in ***:Histopathology revealed presence of tachyzoites only in liver while along with lung,liver,spleen and brain tissue yielded desired positive PCR ***:The SAG 3 based PCR is able to diagnose toxoplasmosis in those tissues which are declared negative by histopathological assay.
