Western science has been strong in measuring details of biological systems such as gene expression levels and metabolite concentrations, and has generally followed a bottom up approach with regard to explaining biological phenomena. Chinese medicine in contrast has evolved as a top down approach in which body and mind is seen as a whole, a phenomenological approach based on the organization and dynamics of symptom patterns. Western and Chinese perspectives are developing towards a ‘middle out' approach. Chinese medicine diagnosis, we will argue, allows bridging the gap between biologists and psychologists and offers new opportunities for the development of health monitoring tools and health promotion *** this paper, health, resilience and stress concepts are explored from an integrated systems biology perspective. This is followed by a discussion about measuring aspects of health and resilience. Then the issue of how to integrate different types of data is discussed. Semi-quantitative modeling appears to be very suitable and looks promising for building integrated health models. The challenge for the future lies in applying those models in developing personalized health monitoring and advice systems that will tempt people to lead more healthy lives.

Extensive studies have been performed to describe the phenotypic changes occurring during malignant transformation of the ***,the cell types and associated changes that contribute to the development of prostate diseases and cancer remain elusive,largely due to the heterogeneous composition of prostatic ***,we conduct a comprehensive evaluation of four human prostate tissues by singlecell RNA sequencing(sc RNA-seq)to analyze their cellular *** identify 18 clusters of cell types,each with distinct gene expression profiles and unique features;of these,one cluster of epithelial cells(Ep)is found to be associated with immune *** addition,we characterize a special cluster of fibroblasts and aberrant signaling changes associated with prostate cancer(PCa).Moreover,we provide insights into the epithelial changes that occur during the cellular senescence and *** results expand our understanding of the unique functional associations between the diverse prostatic cell types and the contributions of specific cell clusters to the malignant transformation of prostate tissues and PCa development.

The 6th Annual Meeting of the United States Chinese Anti-Cancer Association(USCACA) was held in conjunction with the 50 th Annual Meeting of American Society of Clinical Oncology(ASCO) on May 30, 2014 in Chicago, Illinois, the United States of America. With a focus on personalized medicine, the conference featured novel approaches to investigate genomic aberrations in cancer cells and innovative clinical trial designs to expedite cancer drug development in biomarker-defined patient populations. A panel discussion further provided in-depth advice on advancing development of personalized cancer medicines in China. The conference also summarized USCACA key initiatives and accomplishments, including two awards designated to recognize young investigators from China for their achievements and to support their training in the United States. As an effort to promote international collaboration, USCACA will team up with Chinese Society of Clinical Oncology(CSCO) to host a joint session on "Breakthrough Cancer medicines" at the upcoming CSCO Annual Meeting on September 20 th, 2014 in Xiamen, China.

Unprecedented progress has been made in genomic personalized medicine in the last several years, allowing for more individualized healthcare assessments and recommendations than ever before. However, most of this progress in prostate cancer （PCa） care has focused on developing and selecting therapies for late-stage disease. To address this issue of limited focus, we propose a model for incorporating genomic-based personalized medicine into all levels of PCa care, from prevention and screening to diagnosis, and ultimately to the treatment of both early-stage and late-stage cancers. We have termed this strategy the ＂Pyramid Model＂ of personalized cancer care. In this perspective paper, our objective is to demonstrate the potential application of the Pyramid Model to PCa care. This proactive and comprehensive personalized cancer care approach has the potential to achieve three important medical goals： reducing mortality, improving quality of life and decreasing both individual and societal healthcare costs.

The two-dimensional quantum dots (2D-QDs) have been developed significantly in the past decades. The 2D-QDs could be used in bioimaging, biosensing, drug/gene delivery, and photodynamic/photothermal therapy. The potential applications in biology receive increasing attention, which makes them the novel and emerging candidates in biomaterial research fields. In this context, we discuss a variety of 2D-QDs with different physical and chemical properties. We focuse on the latest synthesis progress and recent applications in biotechnological, and biomedical applications of the 2D-QDs and we also evaluate the challenges and prospects in this field.

Dual specificity tyrosine phosphorylation regulated kinase 1 A(DYRK1 A) is an evolutionarily conserved protein kinase belonging to the CMGC kinase family, which is closely related to Down syndrome(DS)and Alzheimer’s disease(AD). In recent years, not only the treatment of diabetes, but also the treatment of cancer gradually focuses on targeting DYRK1 A. Therefore, a series of DYRK1 A inhibitors have been developed to treat relevant diseases and clarify their treatment mechanism furtherly. DYRK1 A inhibitors are mainly divided into natural products and synthetic compounds. Among them, harmine is an excellent DYRK1 A inhibitor. Therefore, the synthetic DYRK1 A inhibitors are mainly based on harmine, which greatly enriches the structure and quantity of DYRK1 A inhibitors. The interaction between the inhibitors and the DYRK1 A protein has a guiding significance in predicting the activity of the inhibitors, and plays an irreplaceable role in the design of the compounds. This paper mainly reviews DYRK1 A inhibitors found in recent years and their structure-activity relationship, looking forward to providing a theoretical basis for the development of DYRK1 A inhibitors.

One of the quintessential challenges in cancer treatment is drug *** mechanisms of drug resistance have been described to date,and new modes of drug resistance continue to be *** phenomenon of cancer drug resistance is now widespread,with approximately 90% of cancer-related deaths associated with drug *** significant advances in the drug discovery process,the emergence of innate and acquired mechanisms of drug resistance has impeded the progress in cancer ***,understanding the mechanisms of drug resistance and the various pathways involved is integral to treatment *** the present review,I discuss the different mechanisms of drug resistance in cancer cells,including DNA damage repair,epithelial to mesenchymal transition,inhibition of cell death,alteration of drug targets,inactivation of drugs,deregulation of cellular energetics,immune evasion,tumor-promoting inflammation,genome instability,and other contributing epigenetic ***,I highlight available treatment options and conclude with future directions.
