Transforming growth factor-beta 1 enhances discharge activity of cortical neurons

Transforming growth factor-beta 1(TGF-β1)has been extensively studied for its pleiotropic effects on central nervous system *** neuroprotective or neurotoxic effects of TGF-β1 in specific brain areas may depend on the pathological process and cell types ***-gated sodium channels(VGSCs)are essential ion channels for the generation of action potentials in neurons,and are involved in various neuroexcitation-related ***,the effects of TGF-β1 on the functional properties of VGSCs and firing properties in cortical neurons remain *** this study,we investigated the effects of TGF-β1 on VGSC function and firing properties in primary cortical neurons from *** found that TGF-β1 increased VGSC current density in a dose-and time-dependent manner,which was attributable to the upregulation of Nav1.3 *** VGSC current density and Nav1.3 expression were significantly abolished by preincubation with inhibitors of mitogen-activated protein kinase kinase(PD98059),p38 mitogen-activated protein kinase(SB203580),and Jun NH2-terminal kinase 1/2 inhibitor(SP600125).Interestingly,TGF-β1 significantly increased the firing threshold of action potentials but did not change their firing rate in cortical *** findings suggest that TGF-β1 can increase Nav1.3 expression through activation of the ERK1/2-JNK-MAPK pathway,which leads to a decrease in the firing threshold of action potentials in cortical neurons under pathological ***,this contributes to the occurrence and progression of neuroexcitatory-related diseases of the central nervous system.

In this review, we summarize the major fundamental advances in immunological research reported in 2011. The highlights focus on the improved understanding of key questions in basic immunology, including the initiation and activation of innate responses as well as mechanisms for the development and function of various T-cell subsets. The research includes the identification of novel cytosolic RNA and DNA sensors as well as the identification of the novel regulators of the Toll-like receptor （TLR） and retinoic acid-inducible gene I （RIG-I）-Iike receptor （RLR） signaling pathway. Moreover, remarkable advances have been made in the developmental and functional properties of innate lymphoid cells （I LCs）. Helper T cells and regulatory T （Treg） cells play indispensable roles in orchestrating adaptive immunity. There have been exciting discoveries regarding the regulatory mechanisms of the development of distinct T-cell subsets, particularly Th17 cells and Treg cells. The emerging roles of microRNAs （miRNAs） in T cell immunity are discussed, as is the recent identification of a novel T-cell subset referred to as follicular regulatory T （TFR） cells.

The immune system has been attracting increasing attention in the field of chronic neurological disorders in the central nervous system(CNS).Autoreactive T cells targeting CNS antigens play a crucial role in the development of various autoimmune diseases,such as multiple sclerosis(MS)and neuromyelitis optica spectrum disorder(NMOSD).Moreover,T cells are now recognized as a pivotal contributor to the pathology of neurodegenerative disorders,including Alzheimer's disease(AD),Parkinson's disease(PD),and multiple system atrophy.

Selection and characterization of the novel anti-human PD-1 FV78 antibody from a targeted epitope mammalian cell-displayed antibody library

Currently,display-based methods are well established and widely used in antibody engineering for affinity maturation and structural stability *** obtained a novel anti-human programmed death 1(PD-1)antibody using computer-aided design and a mammalian cell display technology *** used computer-aided modeling and distance geometry methods to predict and assign the key residues that contributed to the binding of human PD-L1 to ***,we analyzed the sequence of nivolumab(an anti-human PD-1 antibody,referred to as MIL75 in the article)to determine the template for antibody design and library *** identified a series of potential substitutions on the obtained template and constructed a virtual epitope-targeted antibody library based on the physicochemical properties and each possible location of the assigned key *** virtual antibody libraries were displayed on the surface of mammalian cells as the antigen-binding fragments of full-length immunoglobulin ***,we used flow cytometry and sequencing approaches to sort and screen the ***,we obtained a novel anti-human PD-1 antibody named ***78 competitively recognized the PD-1 epitopes that interacted with MIL75 and possessed an affinity comparable to *** results implied that FV78 possessed equivalent bioactivity in vitro and in vivo compared with MIL75,which highlighted the probability and prospect of FV78 becoming a new potential antibody therapy.

Natural evolution of hepatitis C virus infection in hemodialysis Tunisian patients and CTLA-4 SNP's

AIM: To analyze the polymorphisms of CTLA-4 gene involved in the response against hepatitis C virus(HCV) ***: We recruited 500 hemodialysed patients from several hemodialysis centers, all HCV-antibody positive, spread over different regions of Tunisia, as part of a national survey in 2008 conducted in the laboratory of immunology at the Charles Nicolle hospital Tunisia, classified into two groups G1(PCR+) and G2(PCR-) according to the presence or absence of viral RNA. Of these patients, 307 were followed prospectively on a viral molecular level over a period from 2002 to 2008, divided into two groups based on the persistence and viral clearance. PCR-RFLP was performed for the analysis of SNPs(+49) A/G and(+6230) G/A CTLA-4 for these 500 patients and 358 healthy ***: Analysis of clinical and virological charac-teristics of our cohort suggests a nosocomial infection in our hemodialysed patients with transfusion history as a primary risk factor and a predominance of genotype 1b. The haplotype analysis revealed an increase of frequencies of GG(+49)/(CT60) CTLA-4 in the entire patients group compared to controls(P = 0.0036 and OR = 1.42; 95%CI: 1.12-1.79, respectively). This haplotype is therefore associated with susceptibility to HCV infection. CONCLUSION: Our study suggests a possible role of CTLA-4 polymorphisms in the outcome of HCV infection in the Tunisian hemodialysed population.

IMMUNOSUPPRESSIVE ACTION OF QINGHAOSU

Qinghaosu （青蒿素）, isolated and purified from the Chinese herb Artemisia annua Linn（黄花蒿）, and identified as a sesquiterpene with a peroxide bridge and lactone structure,is a highly potent and non-toxic new antimalaria drug. This paper reports the immunosup-pressive action of its water soluble derivative （hemisuccinate Na, QHS）. The remarkable sup-pression by QHS of the in vitroHTdR incorporation by mitogen-stimulated mouse spleencells and human peripheral lymphocytes, as well as the spontaneous incorporation by mousethymocytes and blood cells from some leukemia patients is presented and its characteristics aredescribed. The in vivo effect as shown by quantitative PFC is studied and the differencebetween the present in vitro and in vivo effects is investigated. The possible mechanism ofinhibition and discrepancy in effects are discussed.

Membrane molecules in induction of apoptosis of thymocytes by mouse thymic dendritic cells which express Fas ligands

Apoptosis of thymocytes is involved in the negative selection of thymus, but it remains unclear how the cell death of thymocytes is regulated by the thymic stromal cells. By using immunohistochemistry, TdT mediated dUTP nick end labeling (TUNEL) and flow cytometry methods, it was found that Fas ligand was expressed in the thymic medulla and a mouse thymic medullary type dendritic cell line (MTSC4). The DNA fragmentation and TUNEL positive staining of thymocytes were detected in 6 h of cocultures with MTSC4. 97% of the thymocytes which bound to MTSC4 were Fas antigen positive cells. The DNA fragmentation of thymocytes induced by MTSC4 was inhibited by the addition of 20 mmol/L N acetyl galactosamin monosaccharide and the pretreatment of a monoclonal antibody (PF 18 3) which recognized a putative antigen on MTSC4. These results suggested that the mechanisms of induction of cell death by stromal cells may include the interactions of multiple cell surface molecules, in addition to the Fas/Fas ligand system.
