Colorectal cancer(CRC)is one of the most deadly cancers in the world with few reliable biomarkers that have been selected into clinical guidelines for prognosis of CRC *** this study,mRNA microarray datasets GSE113513,GSE21510,GSE44076,and GSE32323 were obtained from the Gene Expression Omnibus(GEO)and analyzed with bioinformatics to identify hub genes in CRC *** expressed genes(DEGs)were analyzed using the GEO2 R *** ontology(GO)and KEGG analyses were performed through the DAVID *** database and Cytoscape software were used to construct a protein-protein interaction(PPI)network and identify key modules and hub *** analyses of the DEGs were performed on GEPIA *** Connectivity Map database was used to screen potential drugs.A total of 865 DEGs were identified,including 374 upregulated and 491 downregulated *** DEGs were mainly associated with metabolic pathways,pathways in cancer,cell cycle and so *** PPI network was identified with 863 nodes and 5817 *** analysis revealed that HMMR,PAICS,ETFDH,and SCG2 were significantly associated with overall survival of CRC *** blebbistatin and sulconazole were identified as candidate *** conclusion,our study found four hub genes involved in CRC,which may provide novel potential biomarkers for CRC prognosis,and two potential candidate drugs for CRC.

Background and Aims:Hepatitis B virus(HBV)biomark-ers have been used for a better categorization of patients,even though the lack of simple algorithms and the impact of genotypes limit their *** aim was to assess the usefulness of noninvasive markers for the identification of HBV inactive carriers(ICs)in a single-point evaluation and to design a predictive model for their ***-ods:This retrospective-prospective study included 343 consecutive HBeAg-negative ***,analytical,and virological data were collected,and a liver biopsy was performed if *** were classified at the end of follow-up as ICs,chronic hepatitis B and gray zone.A pre-dictive model was constructed,and validated by 1000-boot-strap ***:After 39 months of follow-up,298 subjects were ICs,36 were chronic hepatitis B CHB,and nine were gray ***-nine(25.9%)individuals re-quired a liver *** HBV DNA hazard ratio(HR)6.0,p<0.001),HBV core-related antigen(HBcrAg)(HR 6.5,p<0.001),and elastography(HR 4.6,p<0.001)were inde-pendently associated with the IC *** ACE score(HBV DNA,HBcrAg,elastography),obtained by bootstrapping,yielded an area under the receiver operating characteris-tics(AUROC)of 0.925(95%CI:0.880-0.970,p<0.001)for identification of *** AUROC for genotype D was 0.95,0.96 for A,0.90 for E,and 0.88 for H/*** ACE score of<1 had a positive predictive value of 99.5%,and a score≤12 points had a diagnostic accuracy of 93.8%.Conclusions:Low baseline HBV DNA,HBcrAg,and liver stiffness were in-dependently associated with the IC phase.A score including those variables identified ICs at a single-point evaluation,and might be applied to implement less intensive follow-up strategies.

背景:由于较高的远处转移率,胰体尾癌是致死率最高的肿瘤之一。本研究旨在评估脾血管浸润对于胰体尾癌切除术患者的预后价值。方法:在这项回顾性研究中,我们收集了454例胰体尾癌患者的临床病理资料,通过单因素和多因素分析明确本组患者无进展生存(PFS)和总体生存(OS)的预后影响因素,重点关注脾动/静脉浸润的预后价值。结果:单因素分析结果显示,肿瘤较大、非导管内乳头状黏液瘤(non-IPMN)相关的腺癌、低分化、pT3期、淋巴结转移、脉管浸润、神经浸润、病理学脾静脉浸润、影像学脾静脉浸润者PFS和OS均显著缩短(均P<0.05)。多因素分析证实,non-IPMN相关腺瘤、pT3期、pN1-2期及术后辅助化疗是PFS和OS的独立危险因素;此外,肿瘤较大和影像学脾静脉浸润同时也是PFS的独立预后因素。结论:应当就如何检测和报告胰体尾癌手术标本的脾血管状态,形成一个统一的规范。

We aimed to develop a whole-genome sequencing(WGS)-based copy number variant(CNV)calling algorithm with the potential of replacing chromosomal microarray assay(CMA)for clinical ***-CNV is thus developed for CNV detection from WGS *** performance of this CNV calling algorithm was evaluated in a blinded manner on 31 samples and compared to the 112 CNVs reported by clinically validated CMAs for these 31 *** result showed that JAX-CNV recalled 100%of these ***,JAX-CNV identified an average of 30 CNVs per individual,representing an approximately seven-fold increase compared to calls of clinically validated *** validation of 24 randomly selected CNVs showed one false positive,i.e.,a false discovery rate(FDR)of 4.17%.A robustness test on lowercoverage data revealed a 100%sensitivity for CNVs larger than 300 kb(the current threshold for College of American Pathologists)down to 10×*** CNVs larger than 50 kb,sensitivities were 100%for coverages deeper than 20×,97%for 15×,and 95%for 10×.We developed a WGS-based CNV pipeline,including this newly developed CNV caller JAX-CNV,and found it capable of detecting CMA-reported CNVs at a sensitivity of 100%with about a FDR of 4%.We propose that JAX-CNV could be further examined in a multi-institutional study to justify the transition of first-tier genetic testing from CMAs to ***-CNV is available at https://***/TheJacksonlaboratory/JAX-CNV.

Analysis of the Abnormal Segregation of Pathogenicity in Magnaporthe grisea by Using a Genetic Cross of Oryza and Eleusine Isolates

Background: FK778 is a malononitrilamide, a class of immune suppressive compounds with anti viral featuresand experimental activity in chronic rejection, a potentially interesting combination for organ *** goal of this project was to study the tolerability, immune suppressive efficacy, and anti CMV activity ofFK778 and to assess the in vivo relevance of its previously described inhibition of de novo ***: Heart transplants were performed in rats (BN to Lewis) and treated with varying doses of FK778 orleflunomide for 28 days. At 28 days, at the time of rejection or at the death of the animal, the allograft andother vital organs were obtained for study by light microscopy and immunohistochemistry. In separateexperiments, Lewis rats were given sub-lethal irradiation, inoculated with rat CMV (Maastricht strain) andtreated with varying doses of FK778 and leflunomide. In both the transplant and CMV studies, IP uridinewas given at 250 mg/kg to cohorts or animals receiving FK778 and ***: FK778 controls acute rejection, and inhibits CMV replication at 20 mg/kg but is toxic at 25 mg/*** is manifested as anemia, changes in hepatic and intestinal histology and mortality. The toxicity butnot the immune suppressive or anti viral efficacy, is reduced significantly by exogenous uridine ***: FK778 has both immune suppressive and anti viral activities, neither of which are dependent oninhibition of pyrimidine synthesis. These, and other published observations, suggest that the antiviral activityand a considerable part of the efficacy of the malononitrilamide family of drugs is attributable to activities otherthan drug induced pyrimidine deficiency.

A genetic cross between Oryza isolate Y93-164a-1 and Eleusine isolate SA98-4 was established, and the pathogenicity of 151 F1 progeny isolates was investigated on both host plants rice and finger millet. Results showed that the segregation of pathogenicity in this genetic cross was abnormal, i.e., most of the progeny isolates were nonpathogenic on both host plants. However, no abnormal segregation was observed when middle repetitive sequence MGR586 and 31 single-copy RFLP markers from all of the chromosomes were genetically analyzed. At the same time, comparison of the chromosomal organization among two pairs of parental isolates did not find any genomic abnormity. These results suggested that the ＂abnormal＂ inheritance of pathogenicity in this cross was most likely due to the reassortment of numerous host species specificity genes but not the biased segregation of the host species specificity genes. The host species specificities in M. grisea were likely to be multigenically controlled, at least in the genetic cross involving rice pathogen and the grasses pathogen other than rice.
