A Novel Reassortant H2N3 Influenza Virus Isolated from China

Objective To analyze the genetic composition of a novel H2N3 virus isolate identified from a duck cage swab in a live poultry market （LPM） in 2009 in Guangdong province of China. Methods PCR-positive specimens were inoculated into embryonated chicken eggs and subtyped by conventional RT-PCR. All segments of the virus A/environment/Guangdong/2/2009 were sequenced, and phylogenetic trees were constructed and analyzed. Results The genes of this virus belong to Eurasian-lineage avian viruses. The virus is a reassortant with the HA gene from an H2N2 virus and the NA gene from an H5N3 virus. The PB1, PB2, and NP genes were from an H4N6 virus, the PA was from an H3N8 virus, the M gene was from an H1N3 virus, and the NS gene was from an H10N6 virus. Conclusion market. Its A novel avian-origin reassortant H2N3 influenza virus was detected in a live poultry potential impacts and evolution should be closely monitored.

The title compound 2-（2-methoxy-4-nitrophenylcarbamoyl）phenyl-4＇-fluorobenzoate （C21H15FN206, Mr = 410.35）, a fluorine-containing derivative of salicylamide, was conve- niently synthesized through two steps and crystallized in the orthorhombic space group P21/c with a = 7.6453（15）, b = 14.323（3）, c = 17.035（3） A, V= 1865.4（6） A3, Z = 4, Dc = 1.461 Mg/m^3, 2 = 0.71073 A, μ（MoKa） = 0.115 mm-1, F（000） = 848, R = 0.0705 and wR = 0.1834 for 3267 independent refections with 1 〉 2σ（I）. X-ray analysis reveals that the dihedral angles formed between the 2-methoxy-4-nitrobenzene and benzene ring, the benzene and 4-fluorobenzene, and the 2-methoxy-4-nitrobenzene and 4-fluorobenzene ring are 3.2（4）, 69.8（3） and 72.9（2）~, respectively. Bioassay shows that the title compound has anti-parasitic activity against hydatid protoscoleces.

Dear Editor, Influenza is a contagious, acute respiratory disease caused by influenza viruses. Type A influenza virus has a broad host range and has caused substantial human morbidity and mortality. There are sixteen subtypes of influenza A virus so far, which are divided into two distinct groups： group 1 （H1, H2, H5, H6, H8, H9, Hll, H12, H13 and H16） and group 2 （H3, H4, H7, H10, H14 and H15）（Corti et al., 2011）. Seasonal H1 and H3 subtype influenza viruses are circulating in the human population.

Coxsackievirus A24 variant(CVA24v)is a major pathogen that causes continued outbreaks and pandemics of acute hemorrhagic conjunctivitis(AHC).In China,the first confirmed outbreak of CVA24v-related AHC occurred in Beijing in 1988,followed by another two significant outbreaks respectively in 1994 and 2007,which coincides with the three-stage dynamic distribution of AHC in the world after *** illustrate the genetic characteristics of CVA24v in different periods,a total of 23 strains were isolated from those three outbreaks and the whole genome of those isolations were sequenced and *** with the prototype strain,the 23 strains shared four nucleotide deletions in the 5'UTR except the 0744 strain isolated in *** at the 98th site,one nucleotide insertion was found in all the strains collected from *** 1994 to 2007,amino acid polarity in the VP1 region at the 25th and the 32nd site were *** the 3C and VP1 phylogenetic tree indicated that isolates from 1988 and 1994 belonged to Genotype III(GIII),and 2007 strains to Genotype IV(GIV).According to the Bayesian analysis based on complete genome sequence,the most recent common ancestors for the isolates in1988,1994 and 2007 were respectively estimated around October 1987,February 1993 and December *** evolutionary rate of the CVA24v was estimated to be 7.45×10^(-3) substitutions/site/*** study indicated that the early epidemic of CVA24v in Chinese mainland was the *** mutations and amino acid changes in different genotypes of CVA24v may generate intensity differences of the AHC ***24v has been evolving constantly with a relatively rapid rate.

Objective To provide a feasible and cost-effective next-generation sequencing （NGS） method for accurate identification of viral pathogens in clinical specimens, because enormous limitations impede the clinical use of common NGS, such as high cost, complicated procedures, tremendous data analysis, and high background noise in clinical samples. Methods Viruses from cell culture materials or clinical specimens were identified following an improved NGS procedure： reduction of background noise by sample preprocessing, viral enrichment by barcoded oligonucleotide （random hexamer or non-ribosomal hexanucleotide） primer-based amplification, fragmentation-free library construction and sequencing of one-tube mixtures, as well as rapid data analysis using an in-house pipeline. Results NGS data demonstrated that both barcoded primer sets were useful to simultaneously capture multiple viral pathogens in cell culture materials or clinical specimens and verified that hexanucleotide primers captured as many viral sequences as hexamers did. Moreover, direct testing of clinical specimens using this improved hexanucleotide primer-based NGS approach provided further detailed genotypes of enteroviruses causing hand, foot, and mouth disease （HFMD） and identified other potential viruses or differentiated misdiagnosis events. Conclusion The improved barcoded oligonucleotide primer-based NGS approach is simplified, time saving, cost effective, and appropriate for direct identification of viral pathogens in clinical practice.

Introduction:The severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)Omicron variant is the dominant circulating strain *** assess the importation of SARS-CoV-2 variants in the mainland of China during the Omicron epidemic,the genomic surveillance data of SARS-CoV-2 from imported coronavirus disease 2019(COVID-19)cases in the mainland of China during the first half of 2022 were ***:Sequences submitted from January to July 2022,with a collection date before June 30,2022,were *** proportions of SARS-CoV-2 variants as well as the relationships between the origin and destination of each Omicron imported case were ***:4,946 sequences of imported cases were submitted from 27 provincial-level administrative divisions(PLADs),and the median submission interval was within 1 month after *** 3,851 Omicron sequences with good quality,1 recombinant(XU)and 4 subvariants under monitoring(BA.4,BA.5,BA.2.12.1,and BA.2.13)were recorded,and 3 of them(BA.4,BA.5,and BA.2.12.1)caused local transmissions in the mainland of China later than that recorded in the *** subvariants dominated in the first half of 2022 and shifted from BA.1 to BA.2 then to BA.4 and *** percentage of BA.2 in the imported SARS-CoV-2 surveillance data was far higher than that in the Global Initiative on Sharing All Influenza Data(GISAID).The imported cases from Hong Kong Special Administrative Region,China,accounted for 32.30%of Omicron cases sampled,and 98.71%of them were ***:The Omicron variant showed the intra-Omicron evolution in the first half of 2022,and all of the Omicron subvariants were introduced into the mainland of China multiple times from multiple different locations.
