The expression characteristics of vitellogenin(VTG) in response to B(a)p exposure in polychaete Perinereis aibuhitensis

In order to investigate the endocrine toxicity of B(a)p to marine polychaete P erinereis aibuhitensis, vitellogenin(VTG) cDNA from the P. aibuhitensis was isolated, recombinated and expressed for the first time. The full length P. aibuhitensis vitellogenin gene(PaVTG) was 5 325 bp, and encoded 1 692 amino acids. It contained the vitellogenin_N domain of unknown function(DUF1943), a von Willebrand factor type D domain, as well as a conserved KALGNAG motif. The expression of VTG gene and protein were mainly up-regulated after exposed to B(a)p at transcriptional and translational levels. PaVTG gene expression did not change significantly at day 4. At day 7 PaVTG expression was up-regulated in 0.5 μg/L and 5 μg/L B(a)p group. At day 14 PaVTG was significantly up-regulated in 0.5–10 μg/L B(a)p. The protein expression of PaVTG in 0.5 μg/L and 10 μg/L B(a)p group was up-regulated with time prolonging, but the expression in 5 μg/L and 50 μg/L B(a)p group exhibited first increased and then decreased trend. With the increasing of B(a)p concentration PaVTG mRNA and protein expression both firstly increased then decreased. In contrast to B(a)p exposure, estradiol did not induce PaVTG gene and protein expression, until late times of exposure(14 d). Overall, the results in this study indicate that PaVTG could be used as a potential indicator of the effects environmental estrogenic compounds.

Diel variation in metabolism and ammonia excretion of Marphysa sanguinea (Polychaeta: Eunicidae)

Polychaetes provide an excellent food resource for fi sh and represent the dominant zoobenthos in marine ecosystems. Diel variation in the rates of metabolism and ammonia-N excretion of Marphysa sanguinea were studied. The worms were grouped according to their wet body weight into small（S; 1.24±0.06 g）, medium（M; 4.00±0.30 g）, and large（L; 8.54±1.08 g） categories. Their weight-specifi c metabolic rates, based on aerobic respiration（ R）, were measured at 16℃（±0.2℃） and classed as either routine（ RR） or standard（ RS） rates. Both respiration types decreased with increasing body weight. Respiration was described by R = a W^b, where b was-0.400 9 and-0.532 0 for RR and RS, respectively. Diurnal changes in RS for each group was relatively flat, with a slightly increasing trend with time, but was relatively stable as a whole. RR of the diurnal variation of worms was higher than RS, but both had similar overall trends. The peak values of specifi c dynamic action（SDA）（ RSDA） in the S, M, and L groups were 2.704, 1.149, and 0.682 mg/（g·h）, respectively. The durations of SDA were 13, 6, and 6 h, respectively and the energy expenditures of SDA were 377.98, 117.34, and 74.94 J/g, respectively. These data indicate that the metabolic rates were higher in smaller individuals, which is advantageous for their rapid growth.

mRNA expression of CYP4 in marine polychaete Marphysa sanguinea in response to benzo[a]pyrene

Rapid amplification of cDNA ends (RACE) and real-time polymerase chain reaction (RT-PCR) were carried out to analyze the CYP4 gene expression in polychaete Marphysa sanguinea exposed to benzo[a]pyrene (BaP) in this study. The full length of MsCYP4 cDNA was 2 470 bp, and it encoded 512 amino acids. The deduced amino acid sequence showed 47% identity with CYP4F from frog Xenopus tropicalis and shared high homology with other known CYP4 sequences. To analyse the role of CYP4 in protecting M. sanguinea from BaP exposure, three BaP groups were established: 0.5, 5 and 50 μg/L. Polychaetes were sampled after 3, 7 and 12 d. At 0.5 μg/L, the effect of BaP on MsCYP4 gene expression increased with time prolonged. MsCYP4 gene expression curve showed Ushaped trend with time in 5 and 50 μg/L BaP groups. Therefore, MsCYP4 gene may play an important role in maintaining the balance of cellular metabolism and protecting M. sanguinea from BaP toxicity.

Microsatellite Analysis of Genetic Diversity Between Loach with Different Levels of Ploidy

We used microsatellite markers to investigate the genetic parameters of three different polyploidy populations of Misgurnus anguillicaudatus from Honghu City, Hubei province. Polyacrylamide gel electrophoresis banding patterns of diploid（2n）, triploid（3n） and tetraploid loaches（4n） were analyzed with Pop Gen software. A total of 68 alleles were obtained from seven microsatellite loci and the polymorphism information content（PIC） indices were all above 0.5. The average expected mean heterozygosity values（He） were 0.8420, 0.7186 and 0.8521; the average observed mean heterozygosity values（Ho） were 0.9674, 0.9785 and 0.8928; and the HardyWeinberg P values were 0.3078, 0.3151 and 0.3762, for diploid, triploid and tetraploid individuals, respectively. The results indicated that the three populations were highly polymorphic, with no deviations from Hardy-Weinberg equilibrium observed at all the seven microsatellite loci. This indicated a high level of genetic diversity within the populations. A cluster analysis diagram showed that the shortest genetic distance was between diploid and tetraploid loaches and they shared a close phylogenetic relationship. The triploid and tetraploid individuals had the most distant phylogenetic relationship.

Cloning of Ds MAPK Gene and Construction of Antisense Expression Vector

In order to investigate the role of MAPK gene in adaptation of Dunaliella salina to hypersaline environment, the Ds MAPK gene of D. salina was amplified by PCR. After inverted insertion of the open reading flame ofDs MAPK gene into downstream sequence of 35S promoter of plant expression vector, an antisense expression vector of Ds MAPK gene was successfully constructed and introduced into D. salina cells by LiAc/PEG-mediated method. The expression of Ds MAPK gene in transgenic D. salina was analyzed by semi-quantitative RT-PCR method. The results showed that the expression of Ds MAPK gene in D. salina was significantly inhibited at the transcriptional level. The study laid the foundation for further identification of the function of Ds MAPK gene.

Effects of temperature on keeping-alive without water and antioxidant enzyme of the rock scallop Crassadoma gigantea

Temperature was one of the key factors for the living environment of aquatic organisms, especially for shellfish. The effects of temperature was studied on the survival rate and the median lethal time （LT50） of the rock scallop Crassadoma gigantea, and the effects of temperature stress on the antioxidant enzymes of rock scallop Crassadoma gigantea were studied in order to explore the effects of temperature on keeping-alive without water and antioxidant system of the rock scallop Crassadoma gigantea. The rock scallop Crassadoma gigantea （shell length （39.7± 5） mm） at different temperatures （5℃ and 15℃）, humidity （moisture treatment, not moisturizing treatment） and oxygen （aerobic treatment, not aerobic treatment） in laboratory was studied by the survival analysis and the analysis of factors affecting the effects of the median lethal time （LTso） in order to investigate the effects of temperature on the survival rate and the median lethal time of the rock scallop Crassadoma gigantea. Results showed that low temperature, moisture and aerobic treatment can significantly improve the survival rate and the median lethal time of the rock scallop Crassadoma gigantea （P〈0.05）. The effect of temperature on the survival rate and the median lethal time of the rock scallop Crassadoma gigantea was most significant, followed by the effect of oxygen, and that of humidity was the most weakest. Interaction between temperature and humidity, temperature and oxygen significant interaction（P〉0.05）, and interaction between humidity and oxygen interaction was not significant （P〈0.05） in the interaction of three factors. It provided an important basis for the determination of the rock scallop Crassadoma gigantea in the seedling transportation time.
