Autophagy is a conserved intracellular degradation system that plays a dual role in cell death;thus,therapies targeting autophagy in cancer are somewhat *** is a new form of regulated cell death featured with the iron-dependent accumulation of lethal lipid *** pathway is morphologically,biochemically and genetically distinct from other forms of cell *** studies have revealed crosstalk between autophagy and ferroptosis at the molecular *** this review,we summarize the mechanisms of ferroptosis and autophagy,and more importantly,their roles in the drug resistance of *** connections between ferroptosis and autophagy have been revealed,and a strong causal relationship exists wherein one process controls the other and can be utilized as potential therapeutic targets for *** elucidation of when and how to modulate their crosstalk using therapeutic strategies depends on an understanding of the fine-tuned switch between ferroptosis and autophagy,and approaches designed to manipulate the intensity of autophagy might be the key.

Objective:This study aimed to develop a new polyethylene glycol(PEG)ylatedβ-elemene liposome(PEG-Lipo-β-E)and evaluate its characterization,pharmacokinetics,antitumor effects and safety in vitro and in ***:The liposomes were prepared by ethanol injection and high-pressure micro-jet *** of the liposomes was conducted,and drug content,entrapment efficiency(EE),in vitro release and stability were studied by ultra-fast liquid chromatography(UFLC)and a liquid surface *** was drawn from rats to establish the pharmacokinetic *** anticancer effect was evaluated in a KU-19-19 bladder cancer xenograft *** analyses were performed to evaluate ***:The PEG-Lipo-β-E showed good stability and was characterized as 83.31±0.181 nm in size,0.279±0.004 in polydispersity index(PDI),-21.4±1.06 mV in zeta potential,6.65±0.02 in pH,5.024±0.107 mg/mL inβ-elemene(β-E)content,and 95.53±1.712%in average *** Fourier transform infrared spectroscopy(FTIR)and differential scanning calorimetry(DSC)indicated the formation of PEG-Lipo-β-*** to elemene injection,PEG-Lipo-β-E demonstrated a 1.75-fold decrease in clearance,a 1.62-fold increase in half-life,and a 1.76-fold increase in area under the concentration-time curves(AUCs)from 0 hour to 1.5 hours(Panticancer effect in *** analyses showed that there was no evidence of toxicity to the heart,kidney,liver,lung or ***:The present study demonstrates PEG-Lipo-β-E as a new formulation with ease of preparation,high EE,good stability,improved bioavailability and antitumor effects.

5-Fluorouracil(5-FU)is known as a first-line chemotherapeutic agent against colorectal cancer(CRC),but drug resistance occurs frequently and significantly limits its clinical *** previous study showed that the protocadherin 17(PCDH17)gene was frequently methylated and functioned as a tumor suppressor in ***,the relationship between PCDH17 and 5-FU resistance in CRC remains ***,we revealed that PCDH17 was more highly expressed in 5-FU-sensitive CRC tissues than in 5-FU-resistant CRC tissues,and high expression of PCDH17 was correlated with high BECN1 ***,this expression profile contributed to superior prognosis and increased survival in CRC *** PCDH17 expression augmented the 5-FU sensitivity of CRC in vitro and in vivo by promoting apoptosis and autophagic cell ***,autophagy played a dominant role in PCDH17-induced cell death,as an autophagy inhibitor blocked cell death to a greater extent than the pancaspase inhibitor ***17 inhibition by siRNA decreased the autophagy response and 5-FU ***,we showed that c-Jun NH2-terminal kinase(JNK)activation was a key determinant in PCDH17-induced *** compound SP600125,an inhibitor of JNK,suppressed autophagy and 5-FU-induced cell death in PCDH17-reexpressing CRC *** together,our findings suggest for the first time that PCDH17 increases the sensitivity of CRC to 5-FU treatment by inducing apoptosis and JNK-dependent autophagic cell ***17 may be a potential prognostic marker for predicting 5-FU sensitivity in CRC patients.

Background:Traditional Chinese medicine(TCM)syndrome,also named syndrome,are comprehensive and integral analyses of clinical information which helps to guide different individualized treatment ***:Thirty healthy controls and 80 colorectal cancer(CRC)patients(including 33 Spleen Qi Deficiency syndrome,23 Dampness Heat syndrome,17 Blood Stasis syndrome and 7 other syndrome)were enrolled into this *** mRNAs were extracted from peripheral blood mononuclear *** gene expression for CRC patients with different TCM syndrome was determined by microarray and ***:Spleen Qi Deficiency,Dampness Heat and Blood Stasis were the most common syndromes in CRC *** is a significant difference was found in mRNA expression levels(especially for PIK3CA,STAT3,SOX9 and KDM5C)among Spleen Qi Deficiency,Dampness Heat and Blood Stasis syndrome *** higher mRNA levels of JNK1,TP53,MLH1,MSH6,PMS2,SOCS3,TCF7L2,FAM123B,PSAP,FBXW7,SALL4 and the lower expression of inflammatory cytokine IL-6 were found in Spleen Qi Deficiency group but not other syndrome *** higher mRNA levels of KRAS,MUC16,EGFR,GRASP65,PIK3CA,MAPK7,CD24,STAT3,SLC11A1,Bcl-2,TXNDC17 and some inflammatory cytokines(IL-6,IL-23,TNF-a,CXCR4)were found in Dampness Heat group but not other syndrome *** Stasis syndrome showed higher expression of SOX9,MLH1,MSH6,KDM5C,PCDH11X,PSAP and SALL4,and lower mRNA levels of PIK3CA,CD24,STAT3,CXCR4,TXNDC17 and *** CRC patients with Dampness Heat syndrome might have a poor prognosis than other syndrome ***:The identification of syndrome conditions had different impacts on CRC prognosis,and which might be related with different mRNA expression *** oncogenes and pro-inflammatory cytokines were highly expressed in Dampness Heat group but not other syndrome types,suggesting that the CRC patients with Dampness Heat syndrome might have a poor *** results prelimitarily uncovered the molecular basis of

Ferroptosis,a novel form of programmed cell death,is characterized by iron-dependent lipid peroxidation and has been shown to be involved in multiple diseases,including *** ferroptosis in cancer cells may be a potential strategy for cancer ***,ferroptosis-inducing drugs are attracting more attention for cancer ***,we showed that erianin,a natural product isolated from Dendrobium chrysotoxum Lindl,exerted its anticancer activity by inducing cell death and inhibiting cell migration in lung cancer ***,we demonstrated for the first time that erianin induced ferroptotic cell death in lung cancer cells,which was accompanied by ROS accumulation,lipid peroxidation,and GSH *** ferroptosis inhibitors Fer-1 and Lip-1 but not Z-VAD-FMK,CQ,or necrostatin-1 rescued erianin-induced cell death,indicating that ferroptosis contributed to erianin-induced cell ***,we demonstrated that Ca^(2+)/CaM signaling was a critical mediator of erianin-induced ferroptosis and that blockade of this signaling significantly rescued cell death induced by erianin treatment by suppressing *** together,our data suggest that the natural product erianin exerts its anticancer effects by inducing Ca^(2+)/CaMdependent ferroptosis and inhibiting cell migration,and erianin will hopefully serve as a prospective compound for lung cancer treatment.

Objective:Malignant tumors greatly endanger and affect the quality of human *** antitumor biotherapy strategies,DNA vaccines hold promise in part because of their unique *** this study,an effective broad-spectrum antitumor DNA vaccine expressing human survivin T34A dominant-negative mutant fused with humanβ-defensin-2(HBD2)was ***:The expression profiles of genes of interest were examined using western blotting,reverse transcription-polymerase chain reaction(RT-PCR),and enzyme-linked immunosorbent assay(ELISA)in *** immune function of the fusion gene vaccine(FGV)was assessed in BALB/c mice,which included detection of serum antibody,spleen lymphocyte proliferation,interferon-gamma(IFN-γ)secretion,and lactate dehydrogenase(LDH)*** vivo antitumor effects of FGV were examined in a mouse breast cancer(4T1)model,whereas in vitro effects were assessed using tumor cells derived from different ***-3activity in tumor cells was also assessed after vaccine ***:The FGV triggered humoral as well as cellular immune responses against *** exhibited more potent inhibition of tumor growth as well as prolonged the survival of immunized mice compared to mice immunized with only either survivin T34A or HBD2 *** addition,FGV displayed stronger cytotoxicity against tumor cells derived from different origins compared to the other vaccines and facilitated increased caspase-3 activity in transfected tumor ***:The novel DNA vaccine consisting of a fusion of the universal tumor antigen survivin T34A mutant with molecular adjuvant HBD2generates enhanced broad-spectrum antitumor efficacy against cancers derived from various origins.
