Truttmann MC et al.[1] recently reported that AMPylation of heat shock protein 70 (HSP70) family of chaperones participates in altering the aggregation properties and maintaining protein homeostasis (proteostasis), thereby playing a vital role in the development of neurodegenerative diseases (NDs). NDs are commonly manifested by protein aggregates, which exert harmful effects on proteostasis. Interestingly, it has been observed that AMPylation of heat shock proteins (HSPs) can maintain proteostasis by inhibiting the formation of protein aggregates. As previous studies only indicate that HSPs could regulate proteostasis, such a novel discovery further demonstrates the involvement of HSP70 AMPylation in the regulation of protein aggregation and the maintenance of proteostasis. Therefore, AMPylation can be considered to possess a therapeutic potential to target certain physiological processes related to proteostasis, such as age-related diseases.

Aims:Apelin is the endogenous ligand for the G protein coupled receptor *** promotes vascular smooth muscle cell proliferation and monocyte adhesion to endothelial *** our previous research,we found that apelin-13 promotes H9c2 rat cardiomyocytes hypertrophy by PI3K-Akt-ERK1/2-p70S6K and PI3K-autophagy *** objective of this study was to explore the role of apelin-13 on the regulation of ER stress and autophagy,And then to investigate the association of ER stress and autophagy with the improvement of cell diameter,cell volume aftergiving *** and results:Fuorescence microscope analysis exhibited that apelin-13 increases the production of ROS in a dose dependent *** microscope observation reflected that apelin-13（1μmol/L）activate the endoplasmic reticulum *** Blot results show that apelin-13 promotes the expression of NOX4,Bip and CHOP in dose and time dependent *** sh RNA,NOX4 inhibitor dibenziodolium chloride（DPI）block the increase of Bip,CHOP induced by apelin-13（1μmol/L）.Apelin-13promotes the expression of LC3-II/I,beclin 1and reduce the expression of p62 in dose and time dependent *** sh RNA,NOX4 inhibitor DPI,endoplasmic reticulum stress inhibitor Salubrinal（10μmol/L）,Bip si RNA and CHOP si RNA all can attenuate the overexpression of LC3-II/I,beclin 1 and cripple the decrease of p62 induced by apelin-13（1μmol/L）;Scepter TM Handheld Automated Cell Counter data explain that endoplasmic reticulum stress inhibitor Salubrinal（10μmol/L）,Bip si RNA,CHOP si RNA or autophagy inhibitor 3MA（5mmol/L）all weaken the increase of cell diameter,volume and intracellular protein content induced by apelin-13（2μmol/L）;Endoplasmic reticulum stress revulsant Tunicamycin（1mg/L）enhanced the increase of cell diameter,volume and intracellular protein content induced by apelin-13（2μmol/L）.Conclusion:These findings show that ER stress-autophagy involved in the H9c2 rat cardiomyocytes hypertrophy induced by apelin-13.

Apelin, which is the endogenous ligand of the orphan G protein-coupled receptor APJ, is isolated from bovine stomach. Both Apelin and APJ receptor are widely distributed in various tissues such as heart, brain, limbs, retina and liver. Recent researches indicate that the Apelin/APJ system plays an important role in pathological angiogenesis. Angiogenesis is a progress of new blood branches developing from preexisting vessels via sprouting. New blood branches supply blood and nutrient to damaged regions in various diseases. In this paper, we review the role of Apelin/APJ system in pathological angiogenesis. The Apelin/APJ system promotes angiogenesis in myocardial infarction, ischemic stroke, critical limb ischemia, tumor, retinal angiogenesis diseases, cirrhosis, obesity, diabetes and other related diseases. Furthermore, we illustrate the mechanism of pathological angiogenesis induced by Apelin/APJ system. In conclusion, the Apelin/APJ system would be a promising therapeutic target for angiogenesis-related diseases.

Elabela,a new endogenous ligand for APJ receptor,has been reported to play crucial roles in inducing cardiogenesis, facilitating vasculogenesis and regulating apoptosis during embryonic development. Furthermore, Elabela is also capable of enhancing cardiac contractility, promoting vasodilatory effects, mediating fluid homeostasis in the circulatory system and reducing food intake in the central nervous system. A recent research clarifies that the Elabela/APJ pathway limits Sox32 levels to up-regulate the proportion of Pou5 f3–Nanog complexes in ventrolateral endodermal cells, which may activate the BMP signaling to induce bone formation. Collectively, Elabela is biologically functional in both embryos and adult organisms. Therefore,Elabela may serve as a potential therapeutic peptide for dysontogenesis, cardiac repair/regeneration, blood-pressure control and kidney diseases.
