The aim of study was to detect H5N1 virus in wild geese in Qinghai Province in 2012. The work was provided according to WHO and OIE guidelines. In 2012, we collected 532 samples from wild geese of two species: Bar-headed Goose (Anser indicus) and Graylag Goose (Anser anser). We analyzed samples by chicken embryo inoculation and PCR. No avian influenza viruses were isolated. History of HPAI H5N1 shows obvious importance of Central Asian region in its spreading. The outbreaks of the H5N1 Highly Pathogenic Avian Influenza (HPAI H5N1) were reported in wild birds at the Qinghai Lake since 2005. This area seems to be key point for H5N1 avian influenza surveillance in wild birds. We did not find viruses although H5N1 cases in poultry were reported from 5 provinces of China in 2012. Annual surveillance is required for early AIV detection in this region.

Largemouth Bass Micropterus salmoides is an economically important freshwater fishcultured in *** new emerging viral diseases evoked by iridovirus and rhabdovirus havecaused great economic losses in *** better exploring the pathogenesis of LargemouthBass virus(LMBV),a ranavirus isolate,we investigated the underlying mechanism involved inviral replication and virus induced cell death in *** fluorescence microscopy and flowcytometry,we found that LMBV infection in EPC cells induced typical apoptosis,indicated as theappearance of apoptotic bodies,cytochrome C release and MMP ***,caspaseactivity detection revealed caspase-3,8 and 9 were activated with the infection time,suggestedthat not only mitochondrion-mediated,but also death receptor-mediated apoptosis were involvedin LMBV *** treatment with different inhibitors,viral gene transcription and virusproduction were significantly suppressed by the disruption of proteasome and PI3K ***,both proteasome inhibitor MG132 and PI3K inhibitor LY294002 enhanced virusinduced *** addition,LY294002 also affected the cell cycle alteration induced by ***,we speculated that proteasome system and PI3K pathway played crucial rolesin virus replication by regulated apoptosis evoked by LMBV *** together,our resultsprovided new insights into the molecular mechanism of ranavirus infection.

As a key link between innate and adaptive immune responses,the interferon(IFN)system is the first line of defense against viral ***,and in particular,IFN-α,has been used as an effective therapeutic agent for virus infection in the clinic However,different subtypes of IFN-αdemonstrate distinct antiviral ***,it is important to identify IFN-αsubtypes with high antiviral activity to develop genetically engineered antiviral *** this study,we cloned the genes forl 3 IFN-αsubtypes from peripheral blood lymphocytes of the *** homologies of the 13 mink IFN-αgenes were 93.6-99.3%and 88.8-98.4%at the nucleotide and amino acid sequence levels,*** contrast to human and canine IFN-αsubtypes,most mink IFN-αsubtypes contained two N-glycosylation *** expressed and purified 13mink IFN-αsubtypes in Escherichia *** cytopathic effect inhibition assayshowed that all the 13 recombinant mink IFN-αsubtypes inhibited the propagation of VSV in WISH cells,with IFN-α2 and IFN-α12 demonstrating the highest ***,recombinant mink IFN-α2 and IFN-α12 significantly suppressed the propagation of canine distemper virus in Vera cells,with IFN-α2 demonstrating higher *** summary,we identified mink IFN-α2subtype as a promising prototype for the development of effective antiviral drugs.

Emerging infectious diseases represent a serious threat for human public health worldwide.[1,2]The 2019 novel coronavirus(2019-nCoV)caused a pneumonia outbreak,which is spreading around the country and has affected 32 provinces and regions of China as of January 27,2020.[3,4]Countries outside China,including Japan,the United States,Thailand,and South Korea,have also reported cases imported from other countries.[5]With the joint efforts of Chinese scientists,health workers,and related departments,the pathogen causing this epidemic was quickly identified as a new type of coronavirus,10 days after the first official *** confirming the pathogen,specific detection methods were rapidly developed,with improvement in etiological *** of January 22,2020,it has been confirmed that the new coronavirus came from wild bats and belonged to group 2b of the beta coronavirus,which includes severe acute respiratory syndrome-associated coronavirus(SARS-CoV).[6]Although 2019-nCoV and SARS-CoV belong to the same sub-group of beta coronaviruses,the similarity at the genome level is only 80%,[7,8]meaning that the new virus is genetically different from SARS-CoV[Supplementary Figure 1A,http://***/CM9/A209].Rapid discovery of the causative agent and development of diagnostic reagents demonstrated that technology has greatly improved in the 17 years since the SARS ***,no effective anti-viral medication or vaccines are available for this new virus,and many of its aspects remain to be *** to the SARS outbreak,this outbreak also occurred during the spring festival,the most important of the Chinese traditional festivals,when 3 billion people travel throughout the country.[9]This unexpectedly provides beneficial conditions for the transmission of this highly infectious disease and correspondingly poses great challenges for the prevention and control of the outbreak.

Acute and chronic hepatitis B virus(HBV) infections remain to present a major global health problem. The infection can be associated with acute symptomatic or asymptomatic hepatitis which can cause chronic inflammation of the liver and over years this can lead to cirrhosis and the development of hepatocellularcarcinomas. Currently available therapeutics for chronically infected individuals aim at reducing viral replication and to slow down or stop the progression of the disease. Therefore, novel treatment options are needed to efficiently combat and eradicate this disease. Here we provide a state of the art overview of gene therapeutic approaches to inhibit HBV replication. We discuss non-viral and viral approaches which were explored to deliver therapeutic nucleic acids aiming at reducing HBV replication. Types of delivered therapeutic nucleic acids which were studied since many years include antisense oligodeoxynucleotides and antisense RNA, ribozymes and DNAzymes, RNA interference, and external guide sequences. More recently designer nucleases gained increased attention and were exploited to destroy the HBV genome. In addition we mention other strategies to reduce HBV replication based on delivery of DNA encoding dominant negative mutants and DNA vaccination. In combination with available cell culture and animal models for HBV infection, in vitro and in vivo studies can be performed to test efficacy of gene therapeutic approaches. Recent progress but also challenges will be specified and future perspectives will be discussed. This is an exciting time to explore such approaches because recent successes of gene therapeutic strategies in the clinic to treat genetic diseases raise hope to find alternative treatment options for patients chronically infected with HBV.

Dear Editor,Sugarcane mosaic virus （SCMV） causes severe viral diseases in maize worldwide （Fuchs and Gruntzig, 1995）, resulting in significant losses in grain and forage yield in susceptible cultivars of maize and related crops. The most promising solution is to cultivate resistant varieties, which contribute to sustainable crop production. Two epistatically interacting major SCMV resistance loci （Scmvl and Scmv2） are required to confer complete resistance against SCMV in the resistant nearisogenic line F7RPJRR （the letters left of the slash refer to the genotype at Scmv2 on chromosome 3 and those on the right refer to the genotype at Scmvl on chromosome 6, with R indicating a resistance allele and S a susceptibility allele）（Xing et al., 2006）.

Strategic Engagement of Private Facilities to Increase Public-Private Mix (PPM) Contribution to Nigeria Tuberculosis Case Notification

Introduction: Finding the missing Tuberculosis (TB) cases remains the single most important priority for TB control in Nigeria. Between 66% - 92% of all cases of respiratory diseases including those with symptoms suggestive of TB are first seen byprivate health providers. Dependable, quality surveillance systems and notification are key roles in health services delivery, particularly as it is related to TB control. However, poor notification has been a challenge. This study was to assess the contribution of the public private mix (PPM) to Nigeria Tuberculosis national case notification. Methods: It was a national cross-sectional study. Data were extracted from the National database and reviewed. Private facilities were engaged in 2017 and assessed over 2018-2020. Interventions included: enrolling private practitioners (Private-For-Profit, Faith Based Organization, Private Medicine Vendors and Community Pharmacists), engaging a private standalone laboratory for Gene Xpert testing within the network of private facilities, use of Mobile App for easy screening and reporting, instituting a HUB and spoke, and incentives to private providers for participating. Each private provider had a customized approach. Trend analysis was performed using Cochran-Armitage χ2 test for linear trends. Level of significance was at a p value of Results: Total case notification increased from 104,904 cases in 2017 to 138,591 in 2020. There were 2.0% increase in 2018, 13.0% in 2019 and 15.0% in 2020 (p < 0.001). PPM contribution to case notification increased from 10,699 cases in 2017 to 12,625 in 2018, then 17,250 in 2019 and 38,865 in 2020. There were 18.0% increase in 2018, 36.6% in 2019 and 125.3% increase in 2020 (p ). Conclusion: Effective engagement of the private sector in TB control efforts in Nigeria using a variety of approaches resulting in improved TB notification is possible. Regeneration of Lo Han Kuo (Siraitia grosvenori) in vitro by Direct Organogenesis

The traditional technique for asexual propagation of Siraitia grosvenori in vitro is adopted widely by pressing the vine, which has a high risk of carrying viral diseases and limits the production of promoted cultivars. So this paper reported in vitro regeneration of S. grosvenori by testing for shoot induction from various explant sources such as leaf, petiole and stem. Several phytohormone combinations of TDZ, BA and IAA were examined for shoot regeneration and NAA or IBA for rooting. The highest shoot induction rate （100% of regeneration frequency and 15.3 shoots per explant） in leaf was obtained by incubation on MS medium supplemented with 0.5 mg·L^-1 TDZ, 2.0 mg·L^-1 BA and 0.5 mg·L^-1 IAA; unlike shoot regeneration in leaves, the most efficient bud inductions in petiole and stem explants were initiated on MS medium containing 0.2 mg·L^-1 TDZ, 2.0mg·L^-1 BA and 0.5 mg·L^-1 IAA, in addition, adventitious buds in petiole and stem explants needed to be transformed to MS medium for development; optimal root was obtained when shoots were cultured on 1/2MS medum supplemented with 0.5 mg·L^-1 NAA or 0.5 mg·L^-1 IBA.
