Background： Glucocorticoids （GCs） are involved in the control of appetite in birds and mammals. The effect of GCs on feed intake in birds depends on their dietary energy level. But the regulation mechanism of GCs on appetite is still unclear in chickens facing to different energy level. An experiment was conducted to investigate the effect of dexamethasone （DEX） on hypothalamic expression of appetite-related peptides in chickens fed high/low fat diet and under fasting/feeding condition. Results： An interaction between DEX injection and dietary energy level was found on hypothalamic corticotropin- releasing hormone （CRH） gene expression in fasted chickens （P 〈 0.05）. The chickens, given a DEX injection and a low fat diet treatment, had the highest CRH mRNA levels than any of the fasted chickens given treatments （P 〈 0.05）. Under fasting conditions, the DEX treatment significantly increased hypothalamic neuropeptide Y （NPY） and GC receptors mRNA levels （P 〈 0.05）. Under re-feeding conditions, DEX treatment significantly decreased hypothalamic expression levels of NPY and agouti-related peptide （AgRP） but significantly increased the level of hypothalamic CRH expression （P 〈 0.05）. Conclusion： A regulatory network formed by NPY, AgRP and CRH is associated with the appetite-control by GCs. The result suggests that the regulation of GCs on orexigenic neuropeptides expression is dependent at least partially on dietary energy level and feeding state.

Isolation and characterization of H7N9 viruses from live poultry markets--Implication of the source of current H7N9 infection in humans

On March 31, 2013, the National Health and Family Planning Commission announced that human infections with a previously undescribed influenza A (H7N9) virus had occurred in Shanghai and Anhui Province, China. To investigate the possible origins of the H7N9 viruses causing these human infections, we collected 970 samples, including drinking water, soil, and cloacal and tracheal swabs of poultry from live poultry markets and poultry farms in Shanghai and Anhui Province. Twenty samples were positive for the H7N9 influenza virus. Notably, all 20 viruses were isolated from samples collected from live poultry markets in Shanghai. Phylogenetic analyses showed that the six internal genes of these novel human H7N9 viruses were derived from avian H9N2 viruses, but the ancestor of their HA and NA genes is uncertain. When we examined the phylogenetic relationship between the H7N9 isolates from live poultry markets and the viruses that caused the human infections, we found that they shared high homology across all eight gene segments. We thus identified the direct avian origin of the H7N9 influenza viruses that caused the human infections. Importantly, we observed that the H7N9 viruses isolated from humans had acquired critical mutations that made them more "human-like". It is therefore imperative to take strong measures to control the spread of H7N9 viruses in birds and humans to prevent further threats to human health.

L-Arginine (L-Arg), the precursor of nitric oxide (NO), plays an important role in muscle function. Fasttwitchglycolytic fibres are more susceptible to age-related atrophy than slow-twitch oxidative *** effect of L-Arg/NO on protein metabolism of fast- and slow-twitch muscle fibres was evaluated inchickens. In Exp. 1, 48 chicks at 1 day old were divided into 4 groups of 12 birds and subjected to 4treatments: basal diet without supplementation or supplemented with 1% L-Arg, and water supplementedwith or without L-nitro-arginine methyl ester (L-NAME, 18.5 mM). In Exp. 2, 48 chicks weredivided into 4 groups of 12 birds fed with the basal diet and subjected to the following treatments: tapwater (control), tap water supplemented with L-NAME (18.5 mM), or molsidomine (MS, 0.1 mM), or18.5 mM L-NAME t 0.1 mM MS (NAMS). The regulatory effect of L-Arg/NO was further investigatedin vitro with myoblasts obtained from chicken embryo pectoralis major (PM) and biceps femoris (BF).In vivo, dietary L-Arg supplementation increased breast (t14.94%, P < 0.05) and thigh muscle mass(t23.40%, P < 0.05);whereas, MS treatment had no detectable influence. However, L-NAME treatmentblocked the beneficial influence of L-Arg on muscle development. L-Arg decreased (P < 0.05) proteinsynthesis rate, phosphorylated mTOR and ribosomal protein S6 kinase beta-1 (p70S6K) levels in breastmuscle, which was recovered by L-NAME treatment. In vitro, L-Arg or sodium nitroprusside (SNP)reduced protein synthesis rate, suppressed phosphorylated mTOR/p70S6K and decreased atrogin-1 andmuscle RING finger 1 (MuRF1) in myoblasts from PM muscle (P < 0.05). L-NAME abolished the inhibitoryeffect of L-Arg on protein synthesis and the mTOR/p70S6K pathway. However, myoblasts from BF muscleshowed the weak influence. The aim of present investigation is to study the effect of single- and co-infection with REV and ALV-J on T lymphocytes bioactivities and histopathology in broiler chickens. The bioactivities of blood and spleen T lymphocytes including lymphoproliferation responses, cytotoxicitic responses, and histopathology of spleen were detected in broiler chickens singly- or co-infected with REV and ALV-J at different days post inoculation and the virus expressions in spleen of infected broiler chickens were detected with immunofluorescence assay （IFA）. The results indicated that blood and spleen T lymphocytes proliferation responses and cytotoxicity in broilers infected with REV or/and ALV-J were inhibited in the whole observed period compared with controls. In the co-infected chickens they were highly inhibited than in the single-infected. The histopathology of spleen in infected chickens at 17 and 37 d post inoculation （dpi） indicated that cell interium increased, the numbers of lymphocytes decreased, and the regrowth were destroyed or decreased, especially more significantly at 17 than at 37 dpi. The different numbers of virus were detected in spleen lymphocytes in REV- infected and/or ALV-J-infected chickens. In the spleen of co-infected chicken, both REV and ALV-J were detected and the total numbers of viruses were more than in chickens singly-infected with REV or ALV-J. Thus, the co-effect of REV and ALV-J caused more immunosuppression on T lymphocytes bioactivities in broiler chickens than single-effect of ALV-J or REV, which contributed to the sever histopathology and the product of tumor cells. This study will be helpful for understanding the effect of co-infection with many viruses and control them in poultry.

Prevalence, genetic diversity, and antimicrobial susceptibility profiles of Staphylococcus aureus isolated from bovine mastitis in Zhejiang Province, China

This study was conducted to determine genetic diversity and antimicrobial susceptibility profiles of Staphylococcus aureus recovered from bovine mastiffs in Zhejiang Province, China. Out of 3178 quarter milk samples from 846 lactating cows, among which 459 cows （54.3%） were found HMT positive, 890 quarters （28%） were found having subclinical mastitis. From 75 representative S. aureus isolates, 16 distinct types were identified by pulsed-field gel electrophoresis （PFGE）. Four major PFGE types （A, B, C, and D） accounted for 82.7% of all isolates, and type A （41.3%） was observed in multiple herds across the studied areas. Each region was found to have a predominant type： Hangzhou type A （64.1%）, Ningbo type C （34.5%） and type B （23.1%）, Jinhua type D （53.3%）, and Taizhou type C （62.5%）. Results of antimicrobial susceptibility tests showed that 90.7% of the isolates were resistant to at least one antimicrobial. Resistance to penicillin and ampicillin （77.3%）, tetracycline （60.0%）, or erythromycin （48.0%） was observed. The bacteria resistant to multiple antibiotics such as penicillin, ampicillin, tetracycline, and erythromycin were commonly found. The information obtained from this study is useful for designing specific control programs for bovine S. aureus mastitis in this region.
