Seasonal Changes in Phytoplankton Biomass and Dominant Species in the Changjiang River Estuary and Adjacent Seas:General Trends Based on Field Survey Data 1959-2009

The characteristics of seasonal variation in phytoplankton biomass and dominant species in the Changjiang River Estuary and adjacent seas were discussed based on field investigation data from 1959 to 2009. The field data from 1981 to 2004 showed that the Chlorophyll-a concentration in surface seawater was between 0.4 and 8.5 ktg dm-3. The seasonal changes generally presented a bimodal trend, with the biomass peaks occurring in May and August, and Chlorophyll-a concentration was the lowest in winter. Seasonal biomass changes were mainly controlled by temperature and nutrient levels. From the end of autumn to the next early spring, phytoplankton biomass was mainly influenced by temperature, and in other seasons, nutrient level （including the nutrient supply from the terrestrial runoffs） was the major influence factor. Field investigation data from 1959 to 2009 demonstrated that dia- toms were the main phytoplankton in this area, and Skeletonerna costatum, Pseudo-nitzschia pungens, Coscinodiscus oculus-iridis, Thalassinoema nitzschioides, Paralia sulcata, Chaetoceros lorenzianus, Chaetoceros curvisetus, and Prorocentrum donghaiense Lu were common dominant species. The seasonal variations in major dominant phytoplankton species presented the following trends： 1） Skeletonema （mainly S. costatum） was dominant throughout the year; and 2） seasonal succession trends were Coscinodiscus （spring） →Chaetoceros （summer and autumn） → Coscinodiscus （winter）. The annual dominance of S. costatum was attributed to its environmental eurytopicity and long standing time in surface waters. The seasonal succession of Coscinodiscus and Chaetoceros was associated with the seasonal variation in water stability and nutrient level in this area. Genetic Parameters of Body Length and Response to Selection for Growth Across Four Generations of Artemia sinica

To investigate the genetic components of growth in the brine shrimp Artemia sinica, we estimated the genetic parameters of body length and the response to selection using a fully pedigreed population of A. sinica. The base population was generated from four wild founder populations. We tested 4160 offspring in 360 families over four generations for growth and survival performance. Across four generations, we produced full-and half-sib families with nested mating, where two dams were mated to the same sire. Individual body length was measured for each nauplius at day 20 post-hatching. Heritability of body length was estimated across four generations with the restricted maximum likelihood method. The heritability of body length in A. sinica was low(0.14 ± 0.05), and the common environmental effect was 0.14 ± 0.02. We estimated the response to selection for body length by calculating the difference in the mean breeding values between different generations. The accumulated genetic gain in body length was 278.94 μm after three generations of selection. This low response to selection was probably caused by the low heritability of body length, small sample size, and the low selection intensity(50%). The results suggest that A. sinica selective breeding programs must be changed to generate any substantial, sustainable genetic increases in body length. We suggest that optimal genetic gains could be achieved by introducing wild strains into the nuclear breeding population to increase genetic variation, and by increasing the size of the breeding population to allow for increased selection intensity.

Estimating genetic parameters with molecular relatedness and pedigree reconstruction for growth traits in early mixed breeding of juvenile turbot

An introduced turbot population was used to establish families and to estimate genetic parameters of the offspring. However, there is a lack of pedigree information, and common environmental effects can be introduced when each full-sib family is raised in a single tank. Therefore, in the genetic evaluation, SSRs(simple sequence repeats) were used to reconstruct the pedigree and to calculate molecular relatedness between individuals, and the early mixed-family culture model was used to remove the impact of the common environmental effects. After100 d of early mixed culture, twenty SSRs were used to cluster 20 families and to calculate paired molecular relationships(n=880). Additive genetic matrices were constructed using molecular relatedness(MR) and pedigree reconstruction(PR) and were then applied to the same animal model to estimate genetic parameters. Based on PR, the heritabilities for body weight and body length were 0.214±0.124 and 0.117±0.141, and based on MR they were 0.101±0.031 and 0.102±0.034, respectively. Cross validation showed that the accuracies of the estimated breeding values based on MR(body weight and body length of 0.717±0.045 and 0.629±0.141, respectively) were higher than those of PR(body weight and body length of 0.692±0.052 and 0.615±0.060, respectively). The MR method ensure availability of all genotyped selection candidates, thereby improving the accuracy of the breeding value estimation.

Effect of lamellarization on the microstructure and mechanical properties of marine 10Ni5CrMoV steel

Multistage heat treatment involving quenching(Q),lamellarizing(L),and tempering(T)is applied to marine 10Ni5CrMoV *** microstructure and mechanical properties were studied by multiscale characterizations,and the kinetics of reverse austenite transformation,strain hardening behavior,and toughening mechanism were further *** lamellarized specimens possess low yield strength but high toughness,especially cryogenic *** leads to the development of film-like reversed austenite at the martensite block and lath boundaries,refining the martensite structure and lowering the equivalent grain *** analysis of austenite reversion based on the JMAK model shows that the isothermal transformation is dominated by the growth of reversed austenite,and the maximum transformation of reversed austenite is reached at the peak temperature(750℃).The strain hardening behavior based on the modified Crussard-Jaoul analysis indicates that the reversed austenite obtained from lamellarization reduces the proportion of martensite,significantly hindering crack propagation via martensitic transformation during the *** a consequence,the QLT specimens exhibit high machinability and low yield *** with the QT specimen,the ductile-brittle transition temperature of the QLT specimens decreases from-116 to-130℃due to the low equivalent grain size and reversed austenite,which increases the cleavage force required for crack propagation and absorbs the energy of external load,*** work provides an idea to improve the cryogenic toughness of marine 10Ni5CrMoV steel and lays a theoretical foundation for its industrial application and comprehensive performance improvement.

Numerical Simulation of Dynamic Response of Supple Nets

A simulation method based on the lumped mass model is proposed for determining the dynamic behavior of nets exposed to a uniform current. Every mesh bar is modeled by a linear bar clement. The lumped mass point is set at the ends of each element. The net can be simulated by a discretized model consisting of many point masses and elements without mass. 3D shapes and the distribution of tensions of the net at different moments can be found from time integration of a set of motion equations with a computer program. Two nets are simulated according to reference experiments. Calculated results are in accordance with experimental results. The method is applicable and can be applied to improving design of, and research into other flexible structures, such as net cages.

Cloning of catalase gene and antioxidant genes in Scophthalmus maximus response to metalloprotease of Vibrio anguillarum stress

Metalloproteases represent a class of extracellular proteases found in Vibrio anguillarum that can generate toxic and pathogenic eff ects in turbot(Scophthalmus maximus).The toxicological eff ect partly results from oxidative damage due to the production of excessive reactive oxygen species(ROS).Catalase(CAT),superoxide dismutase(SOD),and glutathione peroxidase(GPx)are major antioxidant enzymes induced by various oxidative stresses and can scavenge peroxides generated in *** evaluate the eff ects of metalloprotease-induced ROS on the antioxidation defense mechanism of *** head kidney cells,the cDNA of CAT gene(designated as SmCAT)was cloned and *** comprises a 1584-bp coding sequence that encodes a protein containing 527 amino acids with a poly(A)*** analysis revealed an active site signature sequence,a heme-ligand signature sequence,and three catalytic amino acid *** deduced SmCAT amino acid sequence shares a sequence similarity of 66.1%-92.4%with those of other *** analysis revealed that SmCAT is classifi ed with CAT of other fi *** real-time PCR analysis showed that SmCAT was extensively expressed in all tested tissues,especially in *** expression of SmCAT,SmMnSOD,and SmGPx were inhibited signifi cantly in head kidney cells treated with metalloprotease from 12 to 24 *** 6 to 24 h metalloprotease-treated groups compared to that of the untreated group,it was found that the production of ROS was markedly increased,and the mitochondrial membrane potential was decreased *** 33342 staining revealed the presence of apoptotic bodies when the cells were incubated with 8.0 or 40.0μg/mL metalloprotease for 12 and 24 ***,the toxic eff ects of metalloprotease are associated with the down-regulation of antioxidant enzyme expression and increased ROS levels,which trigger the activation of apoptosis in the head kidney cells of *** fi ndings provide a better under

Experimental Study on Diesel Spray and Flame Asymmetry Characteristics of Dual-Orifice Nozzles for a Medium-Speed Engine

Transcriptomic Profiling of the Immune Response to Crowding Stress in Juvenile Turbot (Scophthalmus maximus)

In this study,juvenile turbot Scophthalmus maximus were vaccinated with attenuated Edwardsiella tarda(EIBAV1)and reared at two different densities,low density(LD),(5.25±0.02)kg m−2,as control group and high density(HD),(20.53±0.05)kg m−2,as experimental *** density was considered as the *** weeks after vaccination,the transcriptomes of spleen and head kidney from the turbot in two groups were analyzed with RNA-Seq technology.A total of 447 million reads were assembled into 41136 genes with an average length of 1274 bp and a N50 size of 2295 bp.A comparison of gene expression between HD and LD groups revealed 1155 differentially expressed genes(DEGs).Enrichment and pathway analysis of the 10 immune-related DEGs showed the centrality of toll-like receptor signaling pathway,cytosolic DNA-sensing pathway and platelet activation in the host immune *** 5 overexpressed inflammatory cytokines and 5 downregulated signal-regulated cytokines genes are covered by these immune-related *** was inferred that cells suffer damage and the immune response is restrained in turbot under crowding stress.

Analysis of DNA methylation in different tissues of Fenneropenaeus chinensis from the wild population and Huanghai No. 1

DNA methylation plays an important role in the regulation of gene expression during biological development and tissue differentiation in eukaryotes. A methylation sensitive amplification polymorphism（MSAP） including digestion, pre-selective amplification and selective amplification was optimized to compare the levels of DNA cytosine methylation at CCGG sites in muscle, gill and hemocyte from the wild populations and the selective breeding of Huanghai No. 1 of Fenneropenaeus chinensis, respectively. Significant differences in cytosine methylation levels among three tissues in two populations were detected. The average DNA methylation ratios in muscle, gill and hemocyte of the wild population were 23.1%, 22.3% and 19.7%, while those were 21.4%, 19.6%,and 18.9% in Huanghai No. 1, respectively. The DNA methylation levels of gill from the two populations were highly significant（P〈0.01）, the difference of muscle was significant（P〈0.05）, while in hemocyte, there were no significant differences（P〉0.05）. DNA polymorphic methylation of gill and hemocyte between the wild population and Huanghai No. 1 varies to some extent, while those of muscle kept in a balanced degree. Furthermore,polymorphic methylation was associated with demethylation and methylation of CCGG loci.
