Reshaping the active pocket of an aldo-keto reductase to enhance its detoxification activity towards 3-keto-deoxynivalenol

Deoxynivalenol （DON）， the most common mycotoxin in the world， poses serious health hazards to both humans and animals and causes significant economic losses to the food and feed industry. An intriguing method for DON detoxification involves enzymatic cascade catalysis by dehydrogenase （DADH） and aldo-keto reductase （AKR13B3）. However， the poor catalytic activity of AKR13B3 has limited its usefulness. Here， structure-guided steric hindrance engineering， computer-assisted protein engineering， and combinatorial mutagenesis were carried out to increase the catalytic efficiency of AKR13B3. The best mutant in this project was M28S/S65V， which outperformed wild-type AKR13B3 in terms of catalytic efficiency (kcat/Km) and specific activity by factors of 45.08 and 43.11， respectively. Kinetic parameter determination revealed that the enhanced catalytic efficiency of M28S/S65V toward 3-keto-DON was attributed to the decreased Kmand increased kcatvalues. Moreover， the engineered enzyme was applied to degrade DON in contaminated corn steep liquor， for a 90.62% removal rate. Structure-based computational analysis provided insights into the enhanced catalytic efficiency of M28S/S65V， which was attributed to the enlarged substrate-binding pocket and reshaped pocket with a favorable hydrophilic attack distance. The results provide a reference for improving the catalytic activity of an enzyme toward non-native bulky substrates and pave the way for the development of enzymes as detoxification agents to mitigate DON in the food and feed industry.

Antimicrobial peptides(AMPs)in insects are endogenous peptides that are effector components of the innate defense system of the *** may serve as antimicrobial agents because of their small molecular weight and broad-spectrum antimicrobial *** this study,we performed transcriptome analysis of cigarette beetle(Lasioderma serricorne)larvae,parasitized by the ectoparasitic wasp,Anisopteromalus *** AMP genes were significantly upregulated following *** parasitism,postulating the hypothesis that the parasitization enhanced the host's resistance against pathogenic microorganisms through the regulation of host AMP ***,3 AMP genes(LsDef1,LsDef2,and LsCole)were significantly upregulated and we studied their immune function in *** challenge and functional analysis showed that LsCole was responsible for the immune response against Gram-negative and Gram-positive bacteria,while LsDef1 and LsDef2 were involved in insect defense against Gram-positive *** recombinant LsCole exhibited antimicrobial activities against the Gram-negative bacterium Escherichia coli and the Gram-positive bacterium Staphylococcus ***2 showed an antibacterial effect against *** and LsDef2 exhibited antibiofilm activity against *** 2 AMPs disrupted cell membranes and caused leakage of *** cell *** results indicated that the 3 AMPs in *** are involved in the innate immunity of this pest *** AMPs may have potential as antimicrobial agents for bacterial infection ***,data are discussed in relation to new control strategies with greater biosafety against pest insects with use of microbial biocontrol agents in combination with RNA interference against the insect's defensive AMP genes.

Genetic load and inbreeding are recognized as important factors to be considered in conservation *** levels of both can increase the risk of population extinction by negatively impacting fitness-related characters in many species of plants and animals,including humans(inbreeding depression).Genomic tech-niques are increasingly used in measuring and understanding genetic load and inbreeding and their importance in evolution and *** used whole genome resequencing data from two sibling grouse species in subarctic Eurasia to quantify *** found a large range of inbreeding measured as FROH(fraction of runs of homozygosity)in individuals from different populations of Chinese Grouse(Tetrastes sewerzowi)and Hazel Grouse(***).FROH estimated from genome-wide runs of homozygosity(ROH)ranged from 0.02 to 0.24 among Chinese Grouse populations and from 0.01 to 0.44 in Hazel *** from a population of Chinese Grouse residing in the Qilian mountains and from the European populations of Hazel Grouse(including samples from Sweden,Germany and Northeast Poland)were the most inbred(FROH ranged from 0.10 to 0.23 and 0.11 to 0.44,respectively).These levels are comparable to other highly inbred populations of *** Grouse from northern China and Chinese Grouse residing in the Qinghai-Tibetan Plateau showed relatively lower inbreeding *** of the ratio between deleterious missense mutations and synonymous mutations revealed higher levels in Chinese Grouse as compared to Hazel *** results are possibly explained by higher fixation rates,mutational melt down,in the range-restricted Chinese Grouse compared to the wide-ranging Hazel ***,when we compared the relatively more severe class of loss-of-function muta-tions,Hazel Grouse had slightly higher levels than Chinese Grouse,a result which may indicate that purifying selection(purging)has been more efficient in Chinese Grouse on this class of mutations.

The Aral Sea was the fourth largest lake in the world but it has shrunk dramatically as a result of irrational human activities, triggering the "Aral Sea ecological crisis". The ecological problems of the Aral Sea have attracted widespread attention, and the alleviation of the Aral Sea ecological crisis has reached a consensus among the five Central Asian countries(Kazakhstan, Uzbekistan, Tajikistan, Kyrgyzstan, and Turkmenistan). In the past decades, many ecological management measures have been implemented for the ecological restoration of the Aral Sea. However, due to the lack of regional planning and zoning, the results are not ideal. In this study, we mapped the ecological zoning of the Aral Sea from the perspective of ecological restoration based on soil type, soil salinity, surface water, groundwater table, Normalized Difference Vegetation Index(NDVI), land cover, and aerosol optical depth(AOD) data. Soil salinization and salt dust are the most prominent ecological problems in the Aral Sea. We divided the Aral Sea into 7 first-level ecological restoration subregions(North Aral Sea catchment area in the downstream of the Syr Darya River(Subregion Ⅰ);artificial flood overflow area in the downstream of the Aral Sea(Subregion Ⅱ);physical/chemical remediation area of the salt dust source area in the eastern part of the South Aral Sea(Subregion Ⅲ);physical/chemical remediation area of severe salinization in the central part of the South Aral Sea(Subregion Ⅳ);existing water surface and potential restoration area of the South Aral Sea(Subregion Ⅴ);Aral Sea vegetation natural recovery area(Subregion Ⅵ);and vegetation planting area with slight salinization in the South Aral Sea(Subregion Ⅶ)) and 14 second-level ecological restoration subregions according to the ecological zoning principles. Understanding the competition and coexistence of flagship carnivores is key to creating strategies for their conservation in the face of global carnivore *** studies exploring the dynamics and competition between tigers(Panthera tigris)and leopards(***)span decades,there is a lack of understanding regarding the factors that influence their coexistence mechanisms on a broad scale,as well as the drivers determining their exploitative and interference *** gathered a comprehensive list of research papers among which 36 papers explored the interspecific interactions between tigers and leopards and tested the influence of biotic and abiotic factors on the coexistence mechanisms along three dimensions using multiple response variables regression models;we also tested the influence of ecological drivers determining the exploitative or interference competition between tigers and *** and ungulate density were the most important predictors in regulating the coexistence *** and leopards exhibited more positive relations/higher overlaps as elevation increased in the spatial *** addition,they showed a higher dietary overlap in the prey-rich *** determined that interference competition between tigers and leopards was less frequently observed in habitats with dense tree cover and homogeneous vegetation ***,studies with multiple metrics would promote the detection of interference *** study provides new insight into the competitive interactions and coexistence mechanisms of tigers and leopards on a broad ***-makers and managers should pay more attention to the factors of elevation,prey abundance,and habitat structures for the conservation of tigers and leopards.

On the Shallowing of Antarctic Low-Level Temperature Inversions Projected by CESM-LE under RCP8.5

Panicle exsertion is one of the crucial agronomic traits in rice(Oryza sativa).Shortening of panicle exsertion often leads to panicle enclosure and severely reduces seed ***(GA)plays important roles in regulating panicle ***,the underlying mechanism and the relative regulatory network remain ***,we characterized the oswrky78 mutant showing severe panicle enclosure,and found that the defect of oswrky78 is caused by decreased bioactive GA *** analysis demonstrates that OsWRKY78 can directly activate GA biosynthesis and indirectly suppress GA ***,we found OsWRKY78 can interact with and be phosphorylated by mitogen-activated protein kinase(MAPK)kinase OsMAPK6,and this phosphorylation can enhance OsWRKY78 stability and is necessary for its biological *** together,these results not only reveal the critical function of OsWRKY78,but also reveal its mechanism via mediating crosstalk between MAPK and the GA signaling pathway in regulating panicle exsertion.

Actinobacterial community is a conspicuous part of aquatic ecosystems and displays an important role in the case of biogeochemical cycle,but little is known about the seasonal variation of actinobacterial community in reservoir ecological *** this study,the high-throughput techniques were used to investigate the structure of the aquatic actinobacterial community and its inducing water quality parameters in different *** results showed that the highest diversity and abundance of actinobacterial community occurred in winter,with Sporichthya(45.42%)being the most abundant genus and Rhodococcus sp.(29.32%)being the most abundant *** analysis and correlation analysis suggested that in autumn the dynamics of actinobacterial community were infuenced by more factors and Nocardioides ***2R5S2 was the potential keystone species which was negatively correlated with temperature(R=-0.72,Pmanagement and protection in water sources.

The wave-induced liquefaction of seabed is responsible for causing damage to marine *** composition and consolidation degree are the key factors affecting the pore water pressure response and liquefaction behavior of the seabed under wave *** present study conducted wave flume experiments on silt and silty fine sand beds with varying particle ***,a comprehensive analysis of the differences and underlying reasons for liquefaction behavior in two different types of soil was conducted from both macroscopic and microscopic *** experimental results indicate that the silt bed necessitates a lower wave load intensity to attain the liquefaction state in comparison to the silty fine sand ***,the duration and development depth of liquefaction are greater in the silt *** dissimilarity in liquefaction behavior between the two types of soil can be attributed to the variation in their permeability and plastic deformation *** permeability coefficient and compression modulus of silt are lower than those of silty fine ***,silt is more prone to the accumulation of pore pressure and subsequent liquefaction under external *** research has demonstrated that silt beds with varying consolidation degrees exhibit distinct initial failure ***,a dense bed undergoes shear failure,whereas a loose bed experiences initial liquefaction *** study utilized discrete element simulation to examine the microscopic mechanisms that underlie this phenomenon.

The microscopic characterization of isolated bubbles in gassy soil plays an important role in the macroscopic physical properties of sediments and is a key factor in the study of geological hazards in gas-bearing *** on the box-counting method and the pore fractal features in porous media,a fractal model of bubble microstructure parameters in gassy soil under different gas con-tents and vertical load conditions is established by using an industrial X-ray CT scanning *** results show that the fractal di-mension of bubbles in the sample is correlated with the volume fraction of bubbles,and it is also restricted by the vertical *** three-dimensional fractal dimension of the sample is about 1 larger than the average two-dimensional fractal dimension of all the slices from the same *** uniform porous media fractal model is used to test the equivalent diameter,and the results show that the variation of the measured pore diameter ratio is jointly restricted by the volume fraction and the vertical *** addition,the measured self-similarity interval of the bubble area distribution is tested by the porous media fractal capillary bundle model,and the fitting curve of measured pore area ratio in a small loading range is obtained in this paper.
