Sub-chronic effects of a Phthirusa pyrifolia aqueous extract on reproductive function and comparative hormone levels in male rats

Objective: To explore the effects of aqueous extract from Phthirusa pyrifolia leaves(67 mg/kg body weight for 12 days) on the reproductive function of male Wistar rats through oral administration.Methods: Animals(n = 30), aged 13 weeks and weighing(378.5 ± 5.0) g, were housed in a vivarium under controlled environmental conditions [photoperiod of 12 h light/dark,temperature of(23 ± 1)C] and were fed standard rations ad libitum. The experiment ran for 12 days, wherein animals were divided into three groups: negative control(n = 6)received water, positive control(n = 12) with finasteride at a concentration of 1.0 mg/kg;and a test group(n = 12) submitted to aqueous extract. At the end of the experiment, the animals were sacrificed and submitted to analyses.Results: The morphological results of the testes showed that the aqueous extract induced significant changes in the diameter and cross-sectional area of the seminiferous tubules as well as the thickness of the seminiferous epithelium. Furthermore, the extract was able to abruptly decrease testosterone concentrations by about 81.88% in the treated group when compared with the negative control,(47.0 ± 4.8) ng/d L and(255.0 ± 2.0) ng/d L,respectively, and 76.8%,(211.0 ± 8.7) ng/d L, when compared with finasteride. However,the extract causes neither liver damage nor impairment of renal function.Conclusions: These results suggest that the high amounts of flavonoids shown to be in the extract may be responsible for its hepato-protective effects and suggest a possible decrease in the libido and reproduction of rats.

Long COVID has been studied as different sequelae that some individuals can develop after the acute phase of the disease. Persistent symptoms such as dry cough, fatigue, and dyspnea can remain after six months of COVID-19 cure. Others such as lung fibrosis, kidney injury, and thrombotic risk also are observed. Here, a deep review of each human organ and system infected by the virus was performed aiming to show how molecules expression and cell signaling can induce the organism cure or injuries and, subsequently sequelae. The review also suggests the importance of public health surveillance for these cases including a more comprehensive analysis of molecular biology tools that can clarify and assist in the prognosis, treatment, and preventive methods for potentially more serious disorders in post-COVID-19 patients.

Mayaro virus is an emergent alphavirus that infects humans,leading to Mayaro fever.Approximately fifty percent of infected patients develop arthritis symptoms in the recovery phase,a phase that can last up to a year.The literature about Mayaro virus infection and its immune response is scarce,which may hamper the development of treatment strategies.We summarize changes in cytokines and chemokines in the acute and recovery phase in Mayaro virus infected patients,and relate this molecular characterization with the immune response.VEGF and IL-12/p70 show pronounced changes in patients in the acute phase,suggesting the development of cellular immunity and Th1 response.IL-6,IL-7,CXCL8/IL-8,IL-13,IL-17,and IFN-γare elevated in patients with arthritis symptoms in the long-term recovery phase,which may be related to the continuous inflammatory process,a possible Th2 inhibiting and promoting Th17 process.Although few studies discuss the issue,with a small number of patients and different backgrounds,inflammatory and immune response and manifestations seem to be closely linked.This information may help to develop the appropriate treatment strategies in Mayaro virus infection.Therefore,we analyzed and summarized data available in literature.

Study on enzymes of industrial interest in digestive viscera: Greater amberjack (Seriola dumerili)

Objective:To investigate the presence of industrial enzymes (trypsin and chymotrypsin) as well as collagenolytic and fibrinolytic activities of the neglected processing waste of greater amberjackSeriola dumerili(S. dumerili). Methods: The investigations on trypsin, chymotrypsin, collagenolytic and fibrinolytic activities in residues in digestive viscera were performed. The enzymes trypsin and chymotrypsin were physicochemically characterized (including pH- and thermo-stabilities and sensitivity to metal ions and inhibitors). The kinetic parameters were also determined. Collagenolytic and fibrinolytic activities of the crude extract were also investigated. Results:The crude extract ofS. dumerili showed enzymatic activity of (1.4600 ± 0.0011 and (0.7100 ± 0.0079) IU/mg protein for trypsin and chymotrypsin, respectively. The optimal temperature and pH was 60°C and 9 for trypsin and 40°C and 8 for chymotrypsin. The main inhibitory ions were in the following order: Fe3+> Cu2+> Zn2+> Cd2+> Hg2+ (trypsin) and Hg2+> Fe3+> Zn2+> Cd2+> Mn2+(chymotrypsin). TLCK and benzamidine were the strongest inhibitors for both enzymes. The crude extract also presented collagenolytic activity [(42.44 ± 0.01) IU/mg] and fibrinolytic activity ((26.70 ± 0.05) IU/mL)Conclusions:The crude extract ofS. dumerili showed potential for reuse of its residues and to provide biomolecules of industrial interest like trypsin and to be a source of fibrinolytic enzymes.

Objective:To investigate the effect of ions on brain acetylcholinesterase(AChE;EC 3.***.1.7)activities from economic important fish[pirarucu,Arapaima gigas;tambaqui,Colossoma macropomum;cobia,Rachycentron canadum(R.canadum)and Nile tilapia,Oreochromis niloticus(O.niloticus)]comparing with a commercial enzyme from electric eel[Electrophorus electricus(E.electricus)].Methods:Thein vitro exposure was performed at concentrations ranging from 0.001 to 10 mmol/L(except for ethylene diamine tetraacetic acid;up to 150 mmol/L).Inhibition kinetics onR.canadum andO.niloticus were also observed through four methods(Michaelis-Menten,Lineweaver-Burk,Dixon and Cornish-Bowden plots)in order to investigate the type of inhibition produced by some ions.Results:Hg^(2+),As^(3+),Cu^(2+),Zn^(2+),Cd^(2+)caused inhibition in all the species under study.Ca^(2+),Mg^(2+)and Mn^(2+)induced slight activation inR.canadum enzyme while Pb^(2+),Ba^(2+),Fe^(2+),Li+inhibited the AChE from some of the analyzed species.The lowest IC50 and Ki values were estimated forE.electricus AChE in presence of Hg^(2+),Pb^(2+),Zn^(2+).Under our experimental conditions,the results forR.canadum andO.niloticus,As^(3+),Cu^(2+),Cd^(2+),Pb^(2+)and Zn^(2+)showed a non-competitive/mixed-type inhibition,while Hg^(2+)inhibited the enzyme in a mixed/competitive-like manner.Conclusions:E.electricus AChE activity was affected by ten of fifteen ions under study showing that this enzyme could undergo interference by these ions when used as pesticide biosensor in environmental analysis.This hindrance would be less relevant for the crude extracts.

Expression Variability of Carbohydrates at the Cell Wall Surface of Aspergillus Species and in Vitro Susceptibility to Amphotericin B, Voriconazole and Itraconazole

This study evaluated the expression of N-acetyl-D-glucosamine, L-fucose, D-galactose and glucose/mannose on the cell wall surface of Aspergillus species using lectins and the in vitro antifungal activity of AMB (amphotericin B), VOR (voriconazole) and ITC (itraconazole). Con A (Concanavalin A), WGA (wheat germ agglutinin), UEA-I (Ulex europeus agglutinin I) and PNA (peanut agglutinin), all conjugated with horseradish peroxidase, were used. For the susceptibility tests, sterilized flat-bottomed 96-well microtitre plates were used and the procedures were followed of the Clinical and laboratory Standard Institute. The inoculum was added to the wells with the tested drugs and the plates were incubated at 25 °C for 48 h before reading the results to determine the MIC (minimal inhibitory concentration) of AMB, VOR and ITC. All Aspergillus strains showed low MIC for AMB (0.031-1 μg/mL) and ITC (0.031-0.25 μg/mL). However, the isolates were less susceptible to VOR, for which the MIC ranged from 4 to 16 μg/mL. The results of this study indicate that the expression of N-acetyl-D-glucosamine and glucose/mannose on the cell wall surface of the Aspergillus species evaluated showed greater stability of expression in the fungal growth both in vitro and in parasitism.

In Vitro Evaluation of Gamma Irradiation on a Gel Formulation of Cratylia Mollis: Rheological Proporties and Microbiological Control

Lectin Cramoll-1,4, obtained from Cratylia mollis seeds (beans camaratu) was structurally characterized, biologically and pharmacologically, but its use as a biopharmaceutical is not well documented. The objective of this study is to propose a biopharmaceutical formulation lectin Cramoll-1,4, test their hemagglutinating properties in vitro as well as the use of gamma radiation as a continuous process of decontamination formulation. It was made of the extraction and purification Cramoll-1,4, was developed a gel formulation using Carbopol? as a vehicle, at concentrations of 50, 100, 200, 300 and 600 μg was irradiated with 60Co gamma rays in a dose of 7.549 kGy·h–1. The proposed formulation at a concentration of 300 μg produced an increase in the hemagglutinating units Cramoll-1,4 due to the synergistic effect caused by gamma radiation. Considering the diverse use of lectins, specific molecular and structural factors, as well as changes resulting from its formulation, concentration, irradiation and route of administration is of utmost importance to continue the studies in vitro, for subsequent application in vivo to characterize the physiological and molecular processes involved in the response and cellular effects.
