Real time quantitative PCR (RT-qPCR) requires a method to normalize the expression of target genes against an en- dogenous reference gene. It is known that commonly used housekeeping genes (HKGs) vary tremendously in inflam- matory conditions;however information about the stability and expression of HKGs in chronic inflammatory joint dis- ease such as rheumatoid arthritis (RA) is scarce. The expressional stability of 10 commonly used HKGs was analyzed in the neuronal (spinal cord, dorsal root ganglia) and in the musculoskeletal tissues (tendon, muscle, epiphysis, capsule, periosteum and ankle joint) using RT-qPCR in the rat model of RA. In individual tissues, suitable HKGs were selected by | △Ct| (│Ct control-Ct arthritis│) and further analyzed by using software programs;geNorm and normfinder. We found hypoxanthine-guanine phosphoribosyl tranferase (HPRT) as the most stable gene except ankle joint while glyce-raldehyde-3-phosphate dehydrogenase (GAPDH) was found as the least stable gene in musculoskeletal tissues. In in-flamed ankle joint where no reference gene was found to be stably expressed, an inflammatory cell marker CD3 was used to normalize peptidylprolyl isomerase B (PPIB), the most homogenous HKG identified among the 10 HKGs. The normalized PPIB was then used to analyze the gene expression of neurokinin 1 (NK1), receptor of substance P, a potent pro-inflammatory mediator. We observed a 3.5 fold increase (p = 0.009) in NK1 expression in inflamed ankle joint compared to control. Our results indicate that reference genes stability should be evaluated before using them as refer- ence during inflammatory conditions. In tissues with intense inflammatory cell infiltration, an inflammatory cell marker should be used to normalize the selected reference gene to avoid erroneous results.

Drugging the p53 pathway has been a goal for both academics and pharmaceutical companies since the designation of p53 as the'guardian of the genome'.Through growing understanding of p53 biology,we can see multiple routes for activation of both wild-type p53 function and restoration of mutant p53?In this review,we focus on small molecules that activate wild-type p53 and that do so in a nongenotoxit *** particular,we will describe potential approaches to targeting proteins that alter p53 stability and function through posttranslational modification,affect p53's subcellular localization,or target RNA synthesis or the synthesis of *** plethora of pathways for exploitation of p53,as well as the wide-ranging response to p53 activation,makes it an attractive target for anti-cancer therapy.

Propagated sensation along meridians （PSM） is a phenomenon that a sensation moves along meridians during stimulation of an acupoint. PSM has an appearance rate of 1.3% among people and have characteristics of low speed, going toward afflicted sites and being blocked by physical pressure which is difficult to be explained by known neural and blood transmission. Volume transmission （VT） is a widespread mode of intercellular communication in the central nervous system that occurs in the extracellular fluid and in the cerebrospinal fluid. VT signals moves from source to target cells via energy gradients leading to diffusion and convection （flow） which is slow, long distance and much less space filling. VT channel diffuse forming a plexus in the extracellular space with two parameters of volume fraction and tortuosity. Some experiments showed an information transmission between adjacent and distant acupoints along meridians cross spinal segments. This process is a cross-excitation between peripheral nerve terminals which is related to nonsynaptic transmission. Some neurotransmitters or neuropeptides such as glutamate, adenosine triphosphate （ATP） and neuropeptide such as substance P, neurokinin A and calcitonin gene-related peptide relate with the cross-excitation which can be regards as VT signals. Comparing the characteristics of PSM and VT, many similar aspects can be found leading to an assumption that PSM is a process of VT in peripheral tissue along meridians. The reason why VT signals transmit along meridians is that the meridian is rich in interstitial fluid under the condition of low hydraulic resistance which has been proven experimentally. According to Darcy＇s law which descript the flow of interstitial fluid and conservation equation, interstitial fluid will move toward meridians and flow along meridians that restrict the VT signals within the channel and accelerate the flow according to Fick＇s diffusion law. AIM To investigate the reliability and concurrent validity of the Baseline~? Body Level/Scoliosis meter for adolescent idiopathic scoliosis postural assessment in three anatomical *** This is an observational reliability and concurrent validity study of adolescent referrals to the Orthopaedic department for scoliosis screening at karolinskaUniversity Hospital, Stockholm, Sweden between MarchMay 2012. A total of 31 adolescents with idiopathic scoliosis(13.6 ± 0.6 years old) of mild-moderate curvatures(25°± 12°) were consecutively recruited. Measurement of cervical, thoracic and lumbar curvatures, pelvic and shoulder tilt, and axial thoracic rotation(ATR) were performed by two trained physiotherapists in one day. The intraclass correlation coefficient(ICC) was used to determine the inter-examiner reliability(ICC2,1) and the intra-rater reliability(ICC3,3) of the Baseline~? Body Level/Scoliosis meter. Spearman's correlation analyses were used to estimate concurrent validity between the Baseline~? Body Level/Scoliosis meter and Gold Standard Cobb angles from radiographs and the Orthopaedic Systems Inc. *** There was excellent reliability between examiners for thoracic kyphosis(ICC2,1 = 0.94), ATR(ICC2,1 = 0.92) and lumbar lordosis(ICC2,1 = 0.79). There was adequate reliability between examiners for cervical lordosis(ICC2,1 = 0.51), however poor reliability for pelvic and shoulder tilt. Both devices were reproducible in the measurement of ATR when repeated by one examiner(ICC3,3 0.98-1.00). The device had a good correlation with the Scoliometer(rho = 0.78). When compared with Cobb angle from radiographs, there was a moderate correlation for ATR(rho = 0.627).CONCLUSION The Baseline~? Body Level/Scoliosis meter provides reliable transverse and sagittal cervical, thoracic and lumbar measurements and valid transverse plan measurements of mild-moderate scoliosis deformity.
