Establishment and Application of Prevention and Control Techniques of New Sugarcane White Leaf Disease

Aiming at the basic and key technical problems in prevention and control of sugarcane white leaf disease(SCWL),this study systematically overcame key technical bottleneck of prevention and control of new SCWL after 10 years of collaborative research,and achieved several innovative *** phytoplasmas newly recorded in China and the new subgroup of SCWL phytoplasmas(16SrXI-D)were discovered for the first time in Yunnan,and the whole genome analysis of the epidemic subgroup was *** main transmission source of SCWL pathogens has been identified as infected seed canes,and Tettigoniella viridis and Clovia conifer were newly discovered as vectors for virus *** disease resistance of 25 main varieties was identified,and 10 control varieties were *** prevention and control strategy of"emphasizing early warning,strictly carrying out quarantine,blocking the vectors and controlling residual plants"was put forward,and a comprehensive prevention technique was established through integration of various techniques,and standardized technical regulations were formulated for demonstration *** promotion and application of these achievements have realized scientific prevention and control of SCWL,effectively curbed the spread of SCWL,and ensured the safety of sugarcane producing areas in China,achieving great economic,social and ecological benefits and providing technical support for high-quality development,loss reduction and efficiency improvement of China's sugar industry.

The mucosal barrier is crucial for intestinal homeostasis,and goblet cells are essential for maintaining the mucosal barrier *** proviral integration site for Moloney murine leukemia virus-1(PIM1)kinase regulates multiple cellular functions,but its role in intestinal homeostasis during colitis is ***,we demonstrate that PIM1 is prominently elevated in the colonic epithelia of both ulcerative colitis patients and murine models,in the presence of intestinal *** PIM1 leads to decreased goblet cells,thus impairing resistance to colitis and colitis-associated colorectal cancer(CAC)in ***,PIM1 modulates goblet cell differentiation through the Wnt and Notch signaling ***,PIM1 interacts with histone deacetylase 2(HDAC2)and downregulates its level via phosphorylation,thereby altering the epigenetic profiles of Wnt signaling pathway ***,these findings investigate the unknown function of the PIM1-HDAC2 axis in goblet cell differentiation and ulcerative colitis/CAC pathogenesis,which points to the potential for PIM1-targeted therapies of ulcerative colitis and CAC.

The Saccharum genus comprises species with large and variable chromosome numbers, leading to challenges in genomic studies and breeding improvement. Cytogenetics, including classical and molecular approaches, has played a central role in deciphering the genome structure, classification, and evolution of the genus Saccharum. The application of fluorescence in situ hybridization using oligonucleotide probes significantly improved our understanding of the complex genomes of Saccharum species. This paper reviews the application and progress of cytogenetic techniques in Saccharum. Future applications of cytogenetics are discussed, as they could benefit both genomic studies and breeding of sugarcane as well as other plants with complex genomes.

In recent years,with the adjustment of agricultural structure,the change of planting system,the continuous cropping phenomenon and the abnormal climate and environment have provided favorable conditions for the spread,reproduction and survival of sugarcane *** pest species that are harmful to sugarcane have increased,which pose potential threats to sugar cane ***,the occurrence dynamics and control of 6 sugarcane pests with potential threats were studied in order to prospectively grasp the dynamics of sugarcane pests,scientifically and effectively prevent and control sugarcane pests,and ensure the safety of sugarcane production.

Detection,Identification and Phylogenetic Analysis of Sugarcane Pokkah Boeng in Yunnan Sugarcane Areas

[Objectives]This study was conducted to determine the pathogen species and dominant species of sugarcane pokkah boeng in the sugarcane areas of Yunnan in the low-latitude plateau,so as to provide a basis for the pathogenic mechanism,disease resistance breeding and scientific prevention and control of pokkah boeng.[Methods]With 14 pokkah boeng samples collected from different sugarcane areas in Yunnan as materials,pokkah boeng-specific detection primers FvF4/Fv-R4 and Fp-F3/Fp-R3 were designed based on ribosomal DNA non-transcribed spacer(rDNA-ITS)sequences,respectively,and used to perform PCR on Fusarium verticillioides and ***.[Results]In 12 of the 14 pokkah boeng samples,two species of pokkah boeng pathogens were detected,and there was a phenomenon of composite *** composite infection samples were selected for sequence alignment and phylogenetic *** sequences of ***(Gen Bank accession number:MZ126549-MZ126555)and ***(Gen Bank accession number:MZ102259-MZ102265)from seven composite infection samples shared 98.6%-100%and 100%with *** strain 20(Gen Bank accession number:KU508286)and *** Dehong strain(GenBank accession number:KJ629482)published in Gen Bank,*** phylogenetic tree showed that the pathogens of sugarcane pokkah boeng in Yunnan were mainly divided into the *** group and *** *** the *** group,except for ROC 25(Lancang,Yunnan)and Funong10-1405(Maitreya,Yunnan)which were on an independent branch,the remaining five composite infection samples were grouped with *** strains from different geographical origins,and were closely related to *** Guangxi *** strains of *** from different geographic origins were clustered into another group.[Conclusions]This study provides a scientific basis for disease resistance breeding and disease prevention and control of sugarcane pokkah boeng.

Evaluation of the Effect of High-efficiency Compound Formulations on the Prevention and Control of Sugarcane Brown Stripe

[Objectives]This study was conducted to screen high-efficiency compound formulations and precise application technologies for the prevention and control of sugarcane brown stripe disease.[Methods]Carbendazim,benomyl,chlorothalonil,azoxystrobin,pyraclostrobine and difenoconazole·azoxystrobin were selected for field efficacy tests.[Results]Three formulations,(50%carbendazim WP 1500 g+75%chlorothalonil WP 1500 g+potassium dihydrogen phosphate 2400 g+agricultural synergist 300 ml)/hm^(2),(50%benomyl WP 1500 g+75%chlorothalonil WP 1500 g+potassium dihydrogen phosphate 2400 g+agricultural synergist 300 ml)/hm^(2) and(25%pyraclostrobin 600 ml+potassium dihydrogen phosphate 2400 g+agricultural synergist 300 ml)/hm^(2) had good control effects on sugarcane brown stripe disease,and showed disease index below 14.02 and control efficacy above 84.41%.The three formulations are ideal high-efficiency compound formulations for the prevention and control of sugarcane brown stripe *** can be sprayed manually and by unmanned aerial vehicles on the foliar surface from July to August,once every 7 to 10 d,continuously for 2 times.[Conclusions]This study provides new technical support for accurate and efficient prevention and control of sugarcane brown stripe disease.

Analysis of Combining Ability for Resistance of Sugarcane Parents and Cross Combinations to Brown Stripe Disease

[Objectives] The paper was to screen resistant sugarcane varieties against brown stripe disease,and to breed disease-resistant germplasm resource.[Methods]The combining ability for resistance to sugarcane brown stripe disease was analyzed based on 23 female parents,21 male parents and 29 cross combinations. [Results]The average heritability of resistance to sugarcane brown stripe disease successively were female parents（ 95. 3%）,cross combinations（ 93. 0%）and male parents（ 79. 1%）. The general combining ability of 12 female parents showed negative effect,including Pma 98-40,Yacheng 93-26,Yunrui 05-283,Yuetang 91-976,Chuanzhe 19,ROC10,Yunzhe 06-80,ROC26,Zhanzhe 74-141,K86-110,Yunzhe 03-194 and ROC25. The general combining ability of 10 male parents showed negative effect,including Q 199,Yunrui 06-649,Yunrui 05-733,CP 84-1198,CP 88-1762,Yacheng 84-125,Yunrui 05-784,Yuetang 00-236,CP72-3591 and CP 94-110. The special combining ability of 16 cross combinations showed negative effect,including Pma 98-40 × Yunrui 05-649,Yacheng 93-26 ×Yunrui 05-733,Yunrui 05-283 × Q199,Yuetang 91-976 × CP 84-1198,Chuanzhe 19 × CP 88-1762 and ROC10 × Yuenong 73-204. [Conclusions] There were significant differences in combining ability among female parents,male parents and cross combinations,which were mainly controlled by additive and non-additive gene.

Estimation of Probability of Unprotected Heterosexual Vaginal Transmission of HIV-1 from Clients to Female Sex Workers in kaiyuan,Yunnan Province, China

Objective To estimate the probability of unprotected heterosexual vaginal transmission of HIV-1 from clients to Female Sex Workers （FSWs） in kaiyuan County, Yunnan province, China, and analyze factors which impact the transmission probability. Methods An open cohort research of FSWs in kaiyuan was created from surveys conducted in April 2008, October 2008, and April 2009. Two cross-sectional surveys of local clients were also carried out in May 2008 and November 2008. A model was developed to estimate the probability of unprotected heterosexual vaginal transmission of HIV-1 from clients to FSWs. Results The transmission probability from clients to FSWs was estimated as 0.0023 [95% CI 0.0014-0.0032] per unprotected heterosexual act. Conclusion The transmission probability among this group engaging in commercial sex in Yunnan province differs from that found in studies from other countries, and future studies should estimate the influence factors of HIV-1 transmission probability, such as stage of HIV infection, co-morbid sexually transmitted infections （STIs）.
