PI3K/AKT signaling and neuroprotection in ischemic stroke:molecular mechanisms and therapeutic perspectives

It has been reported that the PI3K/AKT signaling pathway plays a key role in the pathogenesis of ischemic *** a result,the development of drugs targeting the PI3K/AKT signaling pathway has attracted increasing attention from *** article reviews the pathological mechanisms and advancements in research related to the signaling pathways in ischemic stroke,with a focus on the PI3K/AKT signaling *** key findings include the following:(1)The complex pathological mechanisms of ischemic stroke can be categorized into five major types:excitatory amino acid toxicity,Ca^(2+)overload,inflammatory response,oxidative stress,and apoptosis.(2)The PI3K/AKT-mediated signaling pathway is closely associated with the occurrence and progression of ischemic stroke,which primarily involves the NF-κB,NRF2,BCL-2,mTOR,and endothelial NOS signaling pathways.(3)Natural products,including flavonoids,quinones,alkaloids,phenylpropanoids,phenols,terpenoids,and iridoids,show great potential as candidate substances for the development of innovative anti-stroke medications.(4)Recently,novel therapeutic techniques,such as electroacupuncture and mesenchymal stem cell therapy,have demonstrated the potential to improve stroke outcomes by activating the PI3K/AKT signaling pathway,providing new possibilities for the treatment and rehabilitation of patients with ischemic *** investigations should focus on the direct regulatory mechanisms of drugs targeting the PI3K/AKT signaling pathway and their clinical translation to develop innovative treatment strategies for ischemic stroke.

Clinical Efficacy of GBR Technique Combined with Temporary Bridgework-Guided Gingival Contouring in Treating Upper Anterior Tooth Loss with Labial Bone Defects

Objective:To investigate the clinical effect of the guided bone regeneration(GBR)technique combined with temporary bridgework-guided gingival contouring in treating upper anterior tooth loss with labial bone ***:From July 2023 to April 2024,80 patients with upper anterior tooth loss and labial bone defects were admitted to the hospital and selected as evaluation *** were divided into an observation group(n=40)and a control group(n=40)using a numerical table lottery *** control group received treatment with the GBR technique,while the observation group received treatment with the GBR technique combined with temporary bridges to guide gingival *** two groups were compared in terms of clinical red aesthetic scores(PES),labial alveolar bone density,labial bone wall thickness,gingival papillae,gingival margin levels,and patient ***:The PES scores of patients in the observation group were higher than those in the control group after surgery(P<0.05).The bone density of the labial alveolar bone and the thickness of the labial bone wall in the observation group were higher than those in the control *** levels of gingival papillae and gingival margins were lower in the observation group after surgery(P<0.05).Additionally,patient satisfaction in the observation group was higher than in the control group(P<0.05).Conclusion:The GBR technique combined with temporary bridge-guided gingival contouring for treating upper anterior tooth loss with labial bone defects can improve the aesthetic effect of gingival soft tissue,increase alveolar bone density and the thickness of the labial bone wall,and enhance patient *** approach is suitable for widespread application in healthcare institutions.

Analysis of the Effect of Hormone Therapy Combined with Hydroxychloroquine in the Treatment of IgA Nephropathy

Objective:To explore and analyze the effect of hormone therapy combined with hydroxychloroquine in the treatment of IgA ***:30 patients with IgA nephropathy who were admitted to the Urology Department of jiuquan hospital from August 2021 to May 2023 were selected as the research subjects,and they divided into an observation group and control group by drawing lots,with 15 cases in each *** observation group underwent hydroxychloroquine treatment in addition of hormone therapy,and the control group underwent conventional treatment and hormone *** rate of effectiveness of treatment,serum index,and renal function of both groups were ***:The incidence of adverse reactions in the observation group was significantly lower than that in the control group(P0.05)between the groups before treatment;after treatment,the serum creatinine and serum albumin in the steroid group were significantly better than those in the reference group(P0.05);after treatment,the hormone group’s urine protein quantification and glomerular filtration rate were significantly better than those in the reference group(P<0.05).The rate of effectiveness of the hormone group was significantly higher than that of the reference group(P<0.05).Conclusion:Hormone therapy combined with hydroxychloroquine in the treatment of IgA nephropathy is more effective than hormone therapy alone,thus this treatment plan is worthy of promotion and application.

Characteristics of shell thickness in a slab continuous casting mold

The key to reduce shell breakout in the continuous casting process is to control shell thickness in the mold. A numerical simulation on the turbulent flow and heat transfer coupled with solidification in the slab mold using the volume of fluid （VOF） model and the enthalpy-porosity scheme was conducted and the emphasis was put upon the flow effect on the shell thickness profiles in longitudinal and transverse directions. The results show that the jet acts a stronger impingement on the shell of narrow face, which causes a zero-increase of shell thickness in a certain range near the impingement point. The thinnest shell on the slab cross-section locates primarily in the center of the narrow face, and secondly near the comer of the wide face. Nozzle optimization can obviously increase the shell thickness and make it more uniform.

Identifying miRNA biomarkers of polycystic ovary syndrome through text mining

Objective:Polycystic ovary syndrome(PCOS)is an endocrine disorder with diverse clinical manifestations that often occurs in women of childbearing ***,its molecular pathogenesis remains unclear,and this study aimed to identify miRNA targets in PCOS through text mining and database ***:First,three different sets of text mining genes(TMGs)associated with"polycystic ovary syndrome","obesity/adiposis",and"anovulation"keywords were retrieved from the GenCLiP3 database,and overlapping genes were ***,Gene ontology annotation and biological pathway enrichment analyses of these overlapping TMGs were performed,followed by protein-protein interaction(PPI)network ***,genes in the gene module clustered in the PPI were selected to predict potential miRNAs for PCOS via miRNA-mRNA ***:A total of 4291 TMGs related to three different keywords were obtained through text mining;72 intersect TMGs were retained among the three gene sets,and 62 TMGs participated in the establishment of the PPI network,of which 18 were aggregated in the gene ***,11 miRNAs that simultaneously bound to two TMGs(IGF1,ESR1,MAPK1,NAMPT,PIK3CA,and SERPINE1)could be prioritized as targets to study ***(s):The discovery of 11 miRNAs(miR-301a-3p,miR-301b-3p,miR-3666,miR-454-3p,miR-130a-3p,miR-130b-3p,miR-4295,miR-190a-3p,miR-5011-5p,miR-548c-3p,and miR-4799-5p)and 6 TMGs,which are associated with the HIF-1 signaling pathway(P=4.799E-08),could be used as potential targets for PCOS.

Objective:Polycystic ovarian syndrome(PCOS)is a common endocrine disorder affecting women of reproductive *** study aimed to use text mining and microarray data analysis to identify drugs that target genes and potential pathways associated with ***:We extracted a common set of genes associated with PCOS using text mining and the microarray dataset ***,we performed Gene Ontology and Kyoto Encyclopedia of Genes and Genomes analyses of these genes,as well as protein–protein interaction(PPI)network ***,we used MCODE and cytoHubba to cluster significant common genes in the PPI network and performed gene–drug interaction analyses to identify potential drugs for further ***,we annotated pathways associated with the genes ***:Text mining and microarray analysis yielded 696 text mining genes(TMGs)and 2804 differentially expressed genes(DEGs).Among these,a set of 77 genes was found in both TMGs and ***,67 of these genes participated in constructing the PPI *** common hub genes were selected using the MCODE and CytoHubba ***,five out of seven genes were targeted by 15 existing ***:Four genes(FASLG,IL13,IL17A,andIL2RA),which are mainly related to the cytokine–cytokine receptor interaction pathway,could be prioritized as targets for PCOS.

Effects of Qigongwan on Wnt/β-catenin Signaling Pathway in Rats with Polycystic Ovary syndrome

[Objectives] To explore the therapeutic effect and mechanism of Qigongwan on PCOS model rats by detecting the changes in sex hormone levels in rats with polycystic ovary syndrome (PCOS), and observing the effects of ovarian pathological morphological changes, apoptosis and expression of Wnt/β-β catenin signaling pathway protein. [Methods] Ten of 40 female SD rats were randomly selected as the normal group, and the other 30 rats were treated with letrozole combined with high-fat diet to establish the PCOS rat model. After successful modeling, the model group was randomly divided into Qigongwan group, positive Daying-35 (Ethinylestradiol and Cyproterone Acetate Tablets) group and model group, with 10 rats in each group. Qigongwan group was given 14.7 g/(kg·d) by gavage, Daying-35 group was given 0.21 mg/(kg·d) by oral gavage, and normal group and model group were given the same amount of distilled water, and the intervention lasted for 21 d. ELISA method was used to detect the levels of hormones such as follicle-stimulating hormone (FSH), luteinizing hormone (LH), testosterone (T), estradiol (E 2) and progesterone (P) in serum. HE staining was used to observe the pathological morphological changes of ovarian tissues;TUNEL staining was used to observe apoptosis of ovarian tissue granule cells;the expression of Wnt, β-catenin protein in rat ovarian tissue was detected by immunohistochemistry. [Results] (i) Compared with the model group, Qigongwan group and Daying-35 group could significantly increase serum E 2 and P levels, significantly reduce serum T levels ( P <0.01), significantly reduce serum LH levels and LH/FSH ratio ( P <0.01), and increase serum FSH levels ( P <0.05) in different degrees. (ii)The results of HE staining showed that compared with the model group, Qigongwan and Daying-35 groups could improve follicular development and reduce atretic follicles in different degrees.
