Roles of Salicylic Acid-responsive Cis-acting Elements and W-boxes in Salicylic Acid Induction of VCH3 Promoter in Transgenic Tobaccos

A salicylic acid (SA)-inducible VCH3 promoter was recently identified from grapevine (Vitisarnurensis) that contains two inverse SA-responsive cis-acting elements and four W-boxes.To furtherdemonstrate the roles of these elements,four fragments with lengths from-1187,-892,-589,-276 to+7 bp were fused with the β-glucuronidase (GUS) reporter géne and transferred to Nicotiana tobacum,together with another four VCH3 promoter fragments with mutation in the two inverse SA-responsiveelements.The functions,of each promoter fragment were-examined by analysis of GUS activity in thetransgenic tobacco root treated with SA.Enhanced GUS activity was shown in the roots of transgenictobaccos with the VCH3 (-1187)-GUS construct containing two SA-responsive cis-acting elements andfour W-boxes.However,GUS activity directed by the VCH3 (-892)-GUS construct,containing one SA cis-acting element and four W-boxes,was reduced by up to 35% compared with that in tobaccos transformedwith the VCH3 (-1187)-GUS construct,indicating that the SA cis-acting element plays an important role inSA induction of the VCH3 promoter.Neither the m2VCH3 (-1187)-GUS nor the mVCH3 (-892)-GUSconstruct,with mutation on the SA-responsive elements,abolished the expression of GUS activity,demon-strating that the W-boxes in the VCH3 promoter are also involved in SA induction.Histochemical arialysis ofGUS activity directed by each of the eight VCH3 promoter fragments showed that GUS was expressedspecifically in vascular tissue.It was concluded that both the SA-responsive cis-acting elements and the W-boxes are important for the SA induction of the VCH3 promoter.This promoter might have a potential usein plant genetic engineering.

Additive manufactured osseointegrated screws with hierarchical design

Bone screws are devices used to fix implants or bones to bones.However,conventional screws are mechanically fixed with thread and often face long-term failure due to poor osseointegration.To improve osseointegration,screws are evolving from solid and smooth to porous and rough.Additive manufacturing(AM)offers a high degree of manufacturing freedom,enabling the preparation of predesigned screws that are porous and rough.This paper provides an overview of the problems currently faced by bone screws:long-term loosening and screw breakage.Next,advances in osseointegrated screws are summarized hierarchically(sub-micro,micro,and macro).At the sub-microscale level,we describe surface-modification techniques for enhancing osseointegration.At the micro level,we summarize the micro-design parameters that affect the mechanical and biological properties of porous osseointegrated screws,including porosity,pore size,and pore shape.In addition,we highlight three promising pore shapes:triply periodic minimal surface,auxetic structure with negative Poisson ratio,and the Voronoi structure.At the macro level,we outline the strategies of graded design,gradient design,and topology optimization design to improve the mechanical strength of porous osseointegrated screws.Simultaneously,this paper outlines advances in AM technology for enhancing the mechanical properties of porous osseointegrated screws.AM osseointegrated screws with hierarchical design are expected to provide excellent long-term fixation and the required mechanical strength.

Heavy use of conventional fertilizers can lead to negative environmental concerns.Controlled-release fertilizers(CRFs)can effectively reduce the amounts of fertilizers used,improve the availability of fer-tilizers,and which is conducive to the protection of the ecological environment and sustainable devel-opment of agriculture.Therefore,it is imperative to develop and use CRFs as an alternative to traditional fertilizers.This review aims to present the classification,raw material composition,benefits,release process,release mode,and manufacturing methods of fertilizers coated with organic-inorganic com-posite membranes(OICMs)in order to provide an overall update and summarize CRFs encapsulated by OICMs and provide an insight for future trends in the field of fertilizers.It is expected that utilizing CRFs encapsulated by OICMs and their characteristics for agricultural applications can provide innovative ideas and suggestions for developing novel CRFs suitable for modern and sustainable agriculture.

AZD1775 and anti-PD-1 antibody synergistically sensitize hepatoma to radiotherapy

Background:Radiation(IR)-induced DNA damage triggers cell cycle arrest and has a suppressive effect on the tumor microenvironment(TME).Wee1,a cell cycle regulator,can eliminate G2/M arrest by phosphorylating cyclin-dependent kinase 1(CDK1).Meanwhile,programed death-1/programed death ligand-1(PD-1/PDL-1)blockade is closely related to TME.This study aims to investigate the effects and mechanisms of Wee1 inhibitor AZD1775 and anti-PD-1 antibody(anti-PD-1 Ab)on radiosensitization of hepatoma.Methods:The anti-tumor activity of AZD1775 and IR was determined by 3-(4,5-dimethylthiazol-2-y1)-2,5-diphenyltetrazolium bromide(MTT)assay on human and mouse hepatoma cells HepG2,Hepa1-6,and H22.The anti-hepatoma mechanism of AZD1775 and IR revealed by flow cytometry and Western blot in vitro.A hepatoma subcutaneous xenograft mice model was constructed on Balb/c mice,which were divided into control group,IR group,AZD1775 group,IR+AZD1775 group,IR+anti-PD-1 Ab group,and the IR+AZD1775+anti-PD-1 Ab group.Cytotoxic CD8^(+)T cells in TME were analyzed by flow cytometry.Results:Combining IR with AZD1775 synergistically reduced the viability of hepatoma cells in vitro.AZD1775 exhibited antitumor effects by decreasing CDK1 phosphorylation to reverse the IR-induced G2/M arrest and increasing IR-induced DNA damage.AZD1775 treatment also reduced the proportion of PD-1^(+)/CD8^(+)T cells in the spleen of hepatoma subcutaneous xenograft mice.Further studies revealed that AZD1775 and anti-PD-1 Ab could enhance the radiosensitivity of hepatoma by enhancing the levels of interferonγ(IFNγ)^(+)or Ki67^(+)CD8 T cells and decreasing the levels of CD8^(+)Tregs cells in the tumor and spleen of the hepatoma mice model,indicating that the improvement of TME was manifested by increasing the cytotoxic factor IFNγexpression,enhancing CD8^(+)T cells proliferation,and weakening CD8^(+)T cells depletion.Conclusions:This work suggests that AZD1775 and anti-PD-1 Ab synergistically sensitize hepatoma to radiotherapy by enhancing IR-induced DNA damage and improv

In this study,sodium alginate(SA),a non-toxic natural polysaccharide with good biocompatibility and biodegradability,was developed for targeted delivery of curcumin(CUR)in tumor therapy.The strategy is to sulfhydrylate the folic acid(FA)modified SA,and the CUR dissolved in ethyl acetate(EAC)phase is coated in microcapsules by a quick,efficient and environment-friendly sonochemical method.The EAC in the microcapsule core is volatile,which can be recycled and reused to reduce cost.The prepared mi-crocapsules(FA-RSMCs@CUR)exhibited similar toxicity to free curcumin in anti-tumour evaluation in vitro.FA-RSMCs@CUR also exhibited effective antibacterial properties in the antibacterial evaluation in vitro.It is expected to become a low-cost tumor targeting vector in the future,and has the potential to be promoted in clinical application.

High quality repair of osteochondral defects in rats using the extracellular matrix of antler stem cells

BACKGROUND Cartilage defects are some of the most common causes of arthritis.Cartilage lesions caused by inflammation,trauma or degenerative disease normally result in osteochondral defects.Previous studies have shown that decellularized extracellular matrix(ECM)derived from autologous,allogenic,or xenogeneic mesenchymal stromal cells(MSCs)can effectively restore osteochondral integrity.AIM To determine whether the decellularized ECM of antler reserve mesenchymal cells(RMCs),a xenogeneic material from antler stem cells,is superior to the currently available treatments for osteochondral defects.METHODS We isolated the RMCs from a 60-d-old sika deer antler and cultured them in vitro to 70%confluence;50 mg/mL L-ascorbic acid was then added to the medium to stimulate ECM deposition.Decellularized sheets of adipocyte-derived MSCs(aMSCs)and antlerogenic periosteal cells(another type of antler stem cells)were used as the controls.Three weeks after ascorbic acid stimulation,the ECM sheets were harvested and applied to the osteochondral defects in rat knee joints.RESULTS The defects were successfully repaired by applying the ECM-sheets.The highest quality of repair was achieved in the RMC-ECM group both in vitro(including cell attachment and proliferation),and in vivo(including the simultaneous regeneration of well-vascularized subchondral bone and avascular articular hyaline cartilage integrated with surrounding native tissues).Notably,the antler-stem-cell-derived ECM(xenogeneic)performed better than the aMSC-ECM(allogenic),while the ECM of the active antler stem cells was superior to that of the quiescent antler stem cells.CONCLUSION Decellularized xenogeneic ECM derived from the antler stem cell,particularly the active form(RMC-ECM),can achieve high quality repair/reconstruction of osteochondral defects,suggesting that selection of decellularized ECM for such repair should be focused more on bioactivity rather than kinship.

Achieving superior performance in powder-metallurgy near-βtitanium alloy by combining hot rolling and rapid heat treatment followed by aging

Heat treatment plays an important role in tailoring the mechanical properties of powder-metallurgy(PM)titanium alloys.However,only limited work about the rapid heat treatment(RHT)of PM titanium alloys has been reported.In this work,RHT was applied to PM Ti-5Al-5Mo-5V-1Cr-1Fe alloy after hot rolling to study the evolution of its mechanical properties and the influence of residual pores on its properties.Through hot rolling and annealing,a fine and uniformα+βstructure with few residual pores is ob-tained.During RHT,the primaryαdissolves gradually and completes in theβregion,and theβgrains then grow,resulting in the continuous decrease in elongation after aging.Moreover,the tensile strength first increases and then decreases with increasing RHT temperature,owing to the increase in volume fraction of secondaryαinα+βregion and theβgrain growth inβregion.In contrast to the RHT of cast-and-wrought titanium,the negative influence of residual pores lowers the RHT temperature for obtaining the highest tensile strength to a temperature below theβ-transus temperature.Despite the negative influence of the residual pores,retained primaryαand fineβgrains with fine secondaryαinside contribute to achieving a good strength/ductility balance(1570 MPa and 6.1%,respectively).Addi-tionally,although at higher cycles to failure,the negative influence of residual pores increases as it affects the crack initiation zone at the subsurface,the good resistance of secondaryαto fatigue crack propaga-tion still enhances the fatigue strength considerably(about 300 MPa).This work suggests a cost-effective strategy to produce titanium alloys with high performance.

Anthocyanins play crucial roles in pollen protection and pollinator attraction in flowering plants.However,the mechanisms underlying flower color determination and whether floral anthocyanin regulators participate in other processes remain largely unresolved in soybeans(Glycine max).In this study,we investigated the genetic components and mechanisms governing anthocyanin biosynthesis in soybean flowers.Molecular and genetic studies have characterized two antagonistic regulators,the positive activator GmMYBA3 and the negative repressor GmMYBR1,that modulate the gene expression of anthocyanin biosynthesis in soybean flowers.Further findings revealed a regulatory interplay between GmMYBA3 and GmMYBR1 bridged by GmTT8a,highlighting the complexity of anthocyanin regulation in different soybean organs.Exploration of additional soybean cultivars demonstrated the universality of GmMYBA3 and GmMYBR1 in regulating floral anthocyanin biosynthesis-related genes,with GmF3’5’H identified as a crucial determinant of white flower color.This study provides a molecular mechanism underlying soybean flower color determination,paving the way for the molecular modification of soybean flowers to probably enhance their resistance to abiotic stresses and attractiveness to pollinators.

Coronavirus disease 2019(COVID-19)is an acute respiratory infection caused by severe acute respiratory syndrome coronavirus 2(SARS-CoV-2).SARS-CoV-2 infection typically presents with fever and respiratory symptoms,which can progress to severe respiratory distress syndrome and multiple organ failure.In severe cases,these complications may even lead to death.One of the causes of COVID-19 deaths is the cytokine storm caused by an overactive immune response.Therefore,suppressing the overactive immune response may be an effective strategy for treating COVID-19.Mesenchymal stem cells(MSCs)and their derived exosomes(MSCs-Exo)have potent homing abilities,immunomodulatory functions,regenerative repair,and antifibrotic effects,promising an effective tool in treating COVID-19.In this paper,we review the main mechanisms and potential roles of MSCs and MSCs-Exo in treating COVID-19.We also summarize relevant recent clinical trials,including the source of cells,the dosage and the efficacy,and the clinical value and problems in this field,providing more theoretical references for the clinical use of MSCs and MSCs-Exo in the treatment of COVID-19.

Emerging strategies for nerve repair and regeneration in ischemic stroke:neural stem cell therapy

Ischemic stroke is a major cause of mortality and disability worldwide,with limited treatment options available in clinical practice.The emergence of stem cell therapy has provided new hope to the field of stroke treatment via the restoration of brain neuron function.Exogenous neural stem cells are beneficial not only in cell replacement but also through the bystander effect.Neural stem cells regulate multiple physiological responses,including nerve repair,endogenous regeneration,immune function,and blood-brain barrier permeability,through the secretion of bioactive substances,including extracellular vesicles/exosomes.However,due to the complex microenvironment of ischemic cerebrovascular events and the low survival rate of neural stem cells following transplantation,limitations in the treatment effect remain unresolved.In this paper,we provide a detailed summary of the potential mechanisms of neural stem cell therapy for the treatment of ischemic stroke,review current neural stem cell therapeutic strategies and clinical trial results,and summarize the latest advancements in neural stem cell engineering to improve the survival rate of neural stem cells.We hope that this review could help provide insight into the therapeutic potential of neural stem cells and guide future scientific endeavors on neural stem cells.
