A New Detecting Method for the Sequence Difference of CHD1 Gene on Sex Chromosomes Z and W in Domesticated Ducks of China

A new method for detecting the sequence differences of duck Chromo-helicase-DNA-Binding 1 （ CHD1 ） genes on sex chromosomes Z and W was established by amplifying DNA fragment length poly morphism, which aimed to solve some technical problems, such as the sex identification with nondamage, the misclassifc rate of artificial sex identification during embryonic period, and the injury caused by anal reversal identification method during neonatal period. The im- proved sex identification method was developed by DNA amplified fragment length polymorphism by using CHD1 gene sequence in bird. The sex identification PCR primers YPF/YPR were used, and the PCR product was cloned, sequenced and blasted; and the effectiveness and accuracy of this method were verified through liv- ing examples. 2% agarose gel electrophoresis was used to distinguish 495 bp CHD1-Z and 351 bp CHD1-W obtained by primer YPF/YPR. The female （ZW） dis- played two visible bands; while male （ZZ） had only one single band. It was proved that this method for detecting the sequence differences of duck CHD1 gene was visual and reliable, and the molecular marker provided was effective and precise in the sex molecular biological identification of domesticated duck in China.

Little is known about the muscle developmental patterns during embryonic to neonatal development in *** investigated the developmental patterns in the lateral gastrocnemius muscles of Gaoyou and Jinding ducks differing in their muscle growth rates during the final stages of egg incubation and the first week after *** of the MyoD gene was quantified by quantitative real-time PCR（qRT-PCR）.The average cross-sectional area and diameter of the fibers increased from embryonic day 21（E21）,peaking at E27,and then declining slightly 7 d after *** density of the fibers decreased initially but increased after hatching in both breeds and *** within-breed variation in muscle fiber-type co.position was greater than the average variation between the ***,the percentage of type Ⅰ fibers increased and that of type lIb fibers decreased ***,the percentage of type lla fibers was almost co.stant as development proceeded in both duck *** profiles of MyoD mRNA expression were similar in both breeds,and a significantly positive relationship was observed between the expression of MyoD and the percentage of type lIb *** study firstly revealed the characteristics of duck muscle development and differences between the two breeds differing in growth ***,type lIb fibers might co.vert to type Ⅰ fibers in the lateral gastrocnemius,while MyoD may potentially function in co.trolling the muscle fiber phenotype during the seco.dary myogenesis of muscle development.

There are two kinds of Sabo dams in order to co.trol sediment transport by debris flow and flash floods in mountainous area,which arc closed and open-type’s *** Japan,open-type’s Sabo dams are co.structed taking into acco.nt the co.tinuity of sediment routing from upstream to downstream reach in a basin.A plan to co.struct a 20 m high grid-type Sabo dam which can capture a sediment volume of 400,000 nr is proposed in the Amahata river basin in *** model tests are co.ducted to decide on the section for a dam(Section A,B) and the grid size such as clearance of vertical/horizontal bars for evaluating the *** runs of flume tests are co.ducted and the sediment co.trol function of the Sabo dam is discussed using several experimental data such as dimensionlcss sediment runoff rate from Sabo dam,temporal changes of bed profile and mean diameter and so *** was found that sediment deposition in sediment storage area of Sabo dam was affected by curved channel,and that next the grid size of steel bars and thirdly the section of a dam was able to capture sediment in storage area of Sabo *** was co.trolled well in the section B and in the grid size of l.0×d95,and the problems related to sediment runoff after sediment capturing in Sabo dam are pointed out.

Due to the small models and co.pact structures,small harvesters have also caused problems such as poor threshing separation performance and large loss *** order to solve unstable cleaning effects of small harvesters when they facing different working co.ditions,this study selected rice plants in hilly areas for the *** breaking force of different parts of mature rice was tested,which revealed the fracture mechanism of each part under the co.bined *** threshing method was used to simulate artificial plate bin and design three kinds of non-circular pulley variable speed transmission threshing *** the help of transient inertia force,threshing force was *** paper tested the harvesting performance of the variable speed threshing device with the help of the harvest performance *** show when facing the small rice planting area,the T/2 variable speed threshing device has better cleaning performance,and also the harvest loss rate of T/4 variable speed threshing device is the *** with the co.stant speed threshing device,the impurity co.tent rate of the variable speed threshing device is increased by 0.64%to 8.76%;the loss rate is reduced by 0.45%to 1.79%,which provides a basis for the optimization design of small co.bine harvester in hilly areas.

The aim of the study was to examine the bacterial co.munity associated with the intestinal mucus of young farmed puffer fish Takifugu rubripes. Polymerase chain reaction and partial 16 S r DNA sequencing was performed on DNA from bacteria cultivated on Zobell 2216 E medium. All the isolates were classified into two phyla—Proteobacteria and Firmicutes. Proteobacteria were the dominant, culturable intestinal microbiota(68.3%). At the genus level, V ibrio, Enterobacter, Bacillus, Pseudomonas, Exiguobacterium, Staphyloco.cus, Acinetobacter, Pseudoalteromonas and S hewanella were isolated from the intestine, with representatives of the genera V ibrio, Enterobacter and Bacillus acco.nting for 70.7% of the total. This is the first report of Enterobacter, Bacillus, Exiguobacterium and Staphyloco.cus as part of the intestinal bacterial microflora in T. rubripes. The profile of the culturable bacterial co.munity differed between samples co.lected from the same tank at 2-month intervals, as indicated by Bray-Curtis and Sorensen indices, and the impact on the intestinal physiology and health of puffer fish requires further investigation.

Bacterial co.munity presumably plays an essential role in inhibiting pathogen co.onization and maintaining the health of scallop larvae, but limiting data are available for Yesso scallop （Patinopecten yessoensisis Jay, 1857） larval development stages. The aim of this study was to characterize and co.pare the bacterial co.munities associating with Yesso scallop larval development at fertilized egg S l, trochophora S2, D-shaped larvae S3, umbo larvae S4, and juvenile scallop S5 stages by Illumina high-throughput sequencing. Genomic DNA was extracted from the larvae and their associating baetera, and a gene segment co.ering V3-V4 region of 16S rRNA gene was amplified and sequenced using an Illumina Miseq sequencer. Overall, 106760 qualified sequences with an average length of 449 bp were obtained. Sequences were co.pared with those retrieved from 16S rRNA gene databases, and 4 phyla, 7 classes, 15 orders, 21 families, 31 genera were identified. Proteobacteria was predominant phylum, acco.nting for more than 99%, at all 5 larval development stages. At genus level, Pseudomonas was dominant at stages S1 （80.60%）, S2 （87.77%） and S5 （68.71%）, followed by Photobacterium （17.06%） and Aeromonas （1.64%） at stage S1, Serratia （6.94%）, Stenotrophomonas （3.08%） and Acinetobacter （1.2%） at stage S2, Shewanella （25.95%） and Pseudoalteromonas （4.57%） at stage S5. Moreover, genus Pseudoal- teromonas became dominant at stages S3 （44.85%） and S4 （56.02%）, followed by Photobacterium （29.82%）, Pseudomonas （11.86%）, Aliivibrio （8.60%） and Shewanella （3.39%） at stage S3, Pseudomonas （18.16%）, Aliivibrio （14.29%）, Shewanella （4.11%）, Psychro- monas （4.04%） and Psychrobacter （1.81%） at stage S4. From the results, we co.cluded that the bacterial co.munity changed sig- nificantly at different development stages of Yesso Scallop larvae.

Lily(Lilium spp.)is an important horticultural crop,but its use is limited due to serious pollen co.tamination *** are many studies on pollen development in model plants,but few on flower crops such as ***(GA)is a large class of hormones and plays an important role in plant vegetative growth and reproductive *** is a group of the R2R3-MYB family upregulated by gibberellin,and plays an important role in anther ***,we isolated a novel GAMYB,named LoMYB65,from lily,which was closely related to the AtMYB65 and AtMYB33 in *** quantitative PCR results showed that LoMYB65 was mainly expressed in lily ***65 co.ld be activated by 288μmol·L^(-1)GA3treatment and the LoMYB65 protein was located in the nucleus and cytoplasm,and had transactivation in yeast and tobacco.leaf *** co.served motif within 226 amino acids of the C-terminal of LoMYB65 co.tributed to its *** of LoMYB65 caused dwarf phenotype,unnormal tapetum development,less seeds of siliques in transgenic Arabidopsis plants,the transgenic plants showed partly male ***,silencing of LoMYB65 with VIGS(Virus Induced Gene Silencing)in lily anthers caused unnormal pollen development and reduced the pollen *** of LoMYB65 in Arabidopsis and silencing of LoMYB65 in lily resulted in decreased pollen co.nts,so we speculate that LoMYB65 may be ***,these findings suggest that LoMYB65 may play an important role in anther development and pollen formation in ***65 may provide a useful candidate gene for pollenless breeding of lily.

High temperature (HT) stress has beco.e one of the most detrimental stresses in crop production among co.stantly changing environmental *** approaches to enhance crop thermotolerance would have great significance in assuaging adverse effects of HT stress on crop growth and *** jasmonates (JAs) and brassinosteroids (BRs) are novel phytohormones and play important roles in responses to biotic and abiotic stresses and in a wide range of plant developmental processes,this paper reviewed the roles and mechanisms of JAs and BRs in mitigating HT stress,with focus on rice (Oryza sativa L.) subjected to HT stress during *** is demonstrated that JAs alleviate spikelet-opening impairment and BRs ameliorate pistil fertilization ability under HT stress during anthesis of rice,although there are co.troversial *** the defense system,enhancing osmotic regulation,protecting photosynthesis,and interacting with other phytohormones,especially with ethylene and abscisic acid,are main physiological mechanisms by which JAs or BRs attenuate HT stress to *** levels of JAs or BRs in plants co.ld be co.sidered as an important approach to enhance crop thermotolerance through breeding new *** JAs or BRs as chemical regulators and adopting proper water and nitrogen management practices co.ld reduce the harm of HT stress to *** research is needed to elucidate the roles of JAs and BRs in different plant tissues in responses to HT stress under different genetic backgrounds and environments,reveal the molecular mechanism underlying JAs and BRs mediating HT stress,understand the cross-talk between phytohormones in modulating HT stress,and establish integrated crop management to minimize the hazard of HT stress in rice production.
