co.stipation is co.mon in the diseases of the digestive system in *** the change in diet structure and the increase in life pressure,the prevalence rate increases year by *** traditional Chinese medicine(TCM),the location of the disease of co.stipation is in the large intestine,which is related to the dysfunction of lung,spleen,liver,kidney and other *** pathogenesis is co.ductive dysfunction of large *** on the theory,Shouhui Tongbian Capsule(SHTB)is co.posed of eight traditional Chinese medicines,including Polygoni multiflori Radix(Heshouwu in Chinese),Aloe(Luhui in Chinese),Cassiae Semen(Juemingzi in Chinese),Ginseng Radix et Rhizoma(Renshen in Chinese),Lycii Fructus(Gouqizi in Chinese),Asini co.ii co.la(Ejiao in Chinese),Aurantii Fructus Immaturus(Zhishi in Chinese),and Atractylodis Macrocephalae Rhizoma(Baizhu in Chinese),which co.ld help to release excessive turbid,and nourishing yin and supplementing qi in the *** study has been carried out to review the latest advances of SHTB in the treatment of *** results showed that significant effect of SHTB was found in the treatment of co.stipation,such as functional co.stipation,and co.stipation associated with tumor chemotherapy,co.itis,type 2 diabetes and chronic cardiac ***,obvious adverse reactions were not *** co.ld effectively treat five types of co.stipation,provide direction for the future exploration of SHTB in the treatment of other types of co.stipation。

Patients with refractory immune thrombocytopenia(ITP)frequently enco.nter substantial bleeding risks and demonstrate limited responsiveness to existing *** co.d-derived mesenchymal stem cells(UC-MSCs)present a promising alternative,capitalizing on their low immunogenicity and potent immunomodulatory effects for treating diverse autoimmune *** prospective phase I trial enrolled eighteen eligible patients to explore the safety and efficacy of UC-MSCs in treating refractory *** research design included administering UC-MSCs at escalating doses of 0.5×10^(6)cells/kg,1.0×10^(6)cells/kg,and 2.0×10^(6)cells/kg weekly for four co.secutive weeks across three co.orts during the dose-escalation phase,followed by a dose of 2.0×10^(6)cells/kg weekly for the dose-expansion *** events,platelet co.nts,and changes in peripheral blood immunity were monitored and reco.ded throughout the administration and follow-up ***,12(with an addition of three patients in the 2.0×10^(6)cells/kg group due to dose-limiting toxicity)and six patients were enrolled in the dose-escalation and dose-expansion phase,*** patients(13/18,72.2%)experienced one or more treatment emergent adverse *** adverse events occurred in four patients(4/18,22.2%),including gastrointestinal hemorrhage(2/4),profuse menstruation(1/4),and acute myocardial infarction(1/4).The response rates were 41.7%in the dose-escalation phase(5/12,two received 1.0×10^(6)cells/kg per week,and three received 2.0×10^(6)cells/kg per week)and 50.0%(3/6)in the dose-expansion *** overall response rate was 44.4%(8/18)among all enrolled *** sum up,UC-MSCs are effective and well tolerated in treating refractory ITP(*** ID:NCT04014166).

Optimization of Extraction Process for Total Flavonoids from Penthorum chinense Pursh and co.parison of Their co.tents from Different Parts

[Objectives]This study was co.ducted to optimize the extraction process of total flavonoids from Penthorum chinense Pursh and co.pare their co.tents from different parts.[Methods]Single factor and orthogonal experiments were designed to optimize the extraction process of total flavonoids from *** Pursh with the volume fraction of ethanol,the ratio of material to liquid,heating reflux extraction time and extraction times as factors,and the co.tent of total flavonoids as the index.A verification test was carried *** optimized extraction process was adopted to co.pare the co.tents of total flavonoids from different parts of *** Pursh.[Results]The best extraction process was extracting the powder of *** Pursh for 2.0 h with 20 times of 55%ethanol by reflux *** this co.dition,the co.tents of total flavonoids were 3.63%,8.90%,11.28%,and 4.36%from stems,leaves,flowers and whole grass of *** Pursh,respectively.[co.clusions]The process is reasonable,feasible and stable,and can effectively extract total flavonoids from *** *** co.tents of total flavonoids from different parts of *** Pursh were quite different,and the value was higher in the leaves and flowers,so the proportions of leaves and flowers should be paid attention to in the industrial processing of *** Pursh.

Aldehyde oxidase(AOX)is a molybdoenzyme that is primarily expressed in the liver and is involved in the metabolism of drugs and other ***-mediated metabolism can result in unexpected outco.es,such as the production of toxic metabolites and high metabolic clearance,which can lead to the clinical failure of novel therapeutic *** models can assist medicinal chemists in rapidly evaluating the AOX metabolic risk of co.pounds during the early phases of drug disco.ery and provide valuable clues for manipulating AOX-mediated metabolism *** this study,we developed a novel graph neural network called AOMP for predicting AOX-mediated *** integrated the tasks of metabolic substrate/non-substrate classification and metabolic site prediction,while utilizing transfer learning from 13C nuclear magnetic resonance data to enhance its performance on both *** significantly outperformed the benchmark methods in both cross-validation and external *** AOMP,we systematically assessed the AOX-mediated metabolism of co.mon fragments in kinase inhibitors and successfully identified four new scaffolds with AOX metabolism liability,which were validated through in vitro ***,for the co.venience of the co.munity,we established the first online service for AOX metabolism prediction based on AOMP,which is freely available at https://***.

In this study, etherification of ginkgolide B and dimethylaminoethyl chloride hydrochloride was investigated as a model reaction in a micro-flow system （MFS）, providing the resulting ethers in high yield with fewer side effects. Meanwhile, this novel process in MFS worked well for other ginkgolides from Ginkgol biloba and halides, giving moderate yields.

In the face of increasingly serious environmental pollution,the health of human lung tissues is also facing serious *** IIE(M2E)is the main metabolite of sweetening agents mogrosides from the anti-tussive Chinese herbal Siraitia *** study elucidated the anti-inflammatory action and molecular mechanism of M2E against acute lung injury(ALI).A lipopolysaccharide(LPS)-induced ALI model was established in mice and MH-S cells were employed to explore the protective mechanism of M2E through the western blotting,co.immunoprecipitation,and quantitative real time-PCR *** results indicated that M2E alleviated LPS-induced lung injury through restraining the activation of secreted phospholipase A2 type IIA(Pla2g2a)-epidermal growth factor receptor(EGFR).The interaction of Pla2g2a and EGFR was identified by *** addition,M2E protected ALI induced with LPS against inflammatory and damage which were significantly dependent upon the downregulation of AKT and m TOR via the inhibition of ***2g2a may represent a potential target for M2E in the improvement of LPS-induced lung injury,which may represent a promising strategy to treat ALI.

Clinical Effect of Yinhuang Qingfei Capsules in Treatment of Asymptomatic and Mild/co.mon Severe Acute Respiratory Syndrome co.onavirus 2 Infection:An Analysis of 242 Cases

[Objectives]To investigate the clinical effect of Yinhuang Qingfei capsules in the treatment of asymptomatic and mild/co.mon severe acute respiratory syndrome co.onavirus 2(SARS-co.-2)infection.[Methods]A total of 362 patients with SARS-co.-2 infection were divided into the treatment group with 242 patients and co.trol group with 120 patients acco.ding to their treatment *** patients in the co.trol group were given standard treatment regimen and those in the treatment group were given Yinhuang Qingfei capsules in addition to the treatment in the co.trol *** two groups were observed in terms of average length of hospital stay,mean time for nucleic acid clearance,TCM syndrome sco.e,and progression to severe/critical illness,and clinical outco.e was co.pared between the two groups.[Results]There was a significant difference in the overall response rate between the treatment group and the co.trol group[97.52%(236/242)vs 95.00%(114/120),Pco.pared with the co.trol group,the treatment group had significantly shorter length of hospital stay and time for nucleic acid clearance(Pco.e,and there was a significant difference in TCM syndrome sco.e between the two groups(Pco.e of *** to severe/critical illness was not observed in either group.[co.clusions]co.pared with the standard treatment regimen alone,standard treatment regimen co.bined with Yinhuang Qingfei capsules can effectively shorten the length of hospital stay and time for nucleic acid clearance and improve TCM symptoms in patients with asymptomatic and mild/co.mon SARS-co.-2 infection.

Synthesis and Crystal Structure of(Z)-5-fluoro-3-(phenyl((5-(pyridin-3-ylmethyl)thiophen-2-yl)amino)methylene)Indolin-2-one

The crystal structure of the title co.pound （Z）-5-fluoro-3-（phenyl（（5-（pyridin-3- ylmethyl）thiophen-2-yl）amino）methylene） indolin-2-one （C25H18FN3OS, Mr = 427.50） has been prepared and determined by single-crystal X-ray diffraction. The crystal is of orthorhombic, space group P212121 with a = 5.931（1）, b = 12.413（1）, c = 28.102（2） A, V= 2090.4（3） A3, Z = 4, Dc = 1.358 g/cm3, F（000） = 888, μ= 0.186 mm-1, MoKa radiation （2 = 0.71073）, R = 0.041 and wR = 0.105 for 4567 observed reflections with I 〉 2σ（I）. X-ray diffraction analysis reveals that the indole and thiophene rings are not co.lanar with a dihedral angle of 46.26（5）°. Intramolecular and intermolecular N-H＇..O hydrogen bonds together with C-H...π interations can be observed in the lattice.
