Incorporating antioxidative peptides within nanofibrous delivery vehicles: Characterization and in vitro release kinetics

L-carnosine (Car), an antioxidative dipeptide, is a promising health-promoting bioactive agent, which can be isolated from animal waste. In the present study, Car loaded pullulan (Pul)-sodium alginate (NaAlg) based nanofibrous delivery vehicles were fabricated by uniaxial, coaxial and emulsion electrospinning. The CaCl2 crosslinking was applied after electrospinning process to evaluate the effect on release behavior of Car. Results showed that Car was successfully loaded in water-in-oil-in-water (W1/O/W2) double emulsion to produce nanofibers by emulsion electrospinning. Encapsulation efficiencies were found to be 74.11% and 81.69% for the uniaxial (UENFs) and coaxial nanofibers (CENFs), respectively. Encapsulation efficiency was determined for different formulas of emulsions, whereas among the samples that could form nanofibers (NFs) and encapsulate Car, the highest value was obtained for nanofibers from emulsion-VIII (EE-VIII NFs) at 68.63%. DPPH and CUPRAC assays revealed that all electrospinning methods demonstrated protective effect on the antioxidant activity of Car and thus helped in enhancing its antioxidant potential significantly. According to in vitro digestion results, the release of Car from all electrospun NFs was predominantly controlled by Fickian diffusion mechanism. The CaCl2 crosslinking treatment improved water resistance of NFs and enhanced sustained release of Car in the gastrointestinal tract. The initial burst release of Car from EE-VIII NFs was significantly lower than for UENFs and CENFs in the gastric phase, and the release from EE-VIII NFs in the intestinal phase was followed by sustained release, with/without crosslinking treatment. It can therefore be said that the simultaneously encapsulation of Car in double emulsion and in Pul-NaAlg based electrospun NFs can provide sustained release.

Bioactivity of non-extractable phenolics from lemon peel obtained by enzyme and ultrasound assisted extractions

Lemon peel waste constitutes a natural resource for bioactive phenolics with antioxidant, antihypertensive and antidiabetic activities. Bioactivities of non-extractable phenolics from lemon peel obtained by conventional heat-, enzyme-, ultrasound-, and ultrasound-enzyme-assisted extractions were evaluated in comparison with those of extractable phenolics. The antioxidant, angiotensin-I-converting enzyme (ACE), and alpha-amylase inhibitory ac-tivities and phenolic profile of the phenolic fractions were analysed. While the extractable fraction had higher total phenolic content, ascorbic acid content, and antioxidant activity, phenolic profile analysis indicated that the non-extractable fraction contained higher concentrations of phenolics especially hesperidin and hesperetin. The concentrations of hesperidin and hesperetin in the non-extractable fraction were 270.9 mg/100 g dry weight and 415.9 mg/100 g dry weight, respectively, which were about two-fold higher than those present in the extractable fraction. Moreover, ACE and alpha-amylase inhibitory activities of non-extractable fraction were stronger than those of the extractable fraction. Total phenolic content, total flavonoid content, and antioxidant activity were increased by enzyme and ultrasound treatments compared to those by conventional heat treatment. However, ACE inhibitory activities of all non-extractable fractions were similar while alpha-amylase inhibitory activity was higher in ultrasound-and ultrasound-enzyme-treated fractions. While ultrasound-assisted extraction slightly improved the yield of non-extractable phenolics, enzyme-assisted extraction yielded two-to four-fold increases in the amounts of phenolic compounds compared to heat-assisted extraction. Non-extractable phenolic fraction from lemon peel was found to have a significant potential as an antihypertensive and antidiabetic agent.

Evaluating bioactivity and bioaccessibility properties of the propolis extract prepared with L-lactic acid: An alternative solvent to ethanol for propolis extraction

Ethanol readily solubilizes the active components of propolis. Due to certain disadvantages of ethanolic extracts, developing new non-ethanolic extraction methods is highly desirable. This study aimed to determine the content of phenolic compounds and antioxidant activity of propolis by using lactic acid extraction compared to ethanol. Various concentrations of ethanol and lactic acid were utilized to produce extracts, and an in vitro digestion procedure was applied to the extracts to compare the bioaccessibility of phenolic compounds by using LC-MS/MS in extracts and HPLC-DAD in digested samples. The compounds detected in the lactic acid-based propolis so-lution had similar values to those in the ethanol solution at a propolis concentration of 10%. The propolis-lactic acid solution had a significantly higher antioxidant activity against DPPH radical than the one prepared with ethanol at the same concentration (p < 0.05). Chrysin and naringenin were found as the most abundant phenolic compounds in the initial samples during in vitro digestion experiments. Pinocembrin was the most bioaccessible component among the dialyzed IN fractions of ethanol and lactic acid-based solutions, whereas some compounds were not detected. The results suggest that lactic acid may be used as an alternative to ethanol for propolis extraction.

A new plant-based milk alternative of Pistacia vera geographically indicated in Turkiye: Antioxidant activity, in vitro bio-accessibility, and sensory characteristics

The plant-based food market is expanding and demand for novel plant-based milk alternatives is increasing. Compared to conventional milk, lower protein content, and lower sensory acceptance are some of the challenges of the plant-based milk alternative industry. This research aimed to develop novel plant-based milk from two different local pistachio varieties grown in Antep and Siirt, Turkiye that would be higher in protein, unsaturated fatty acids, and phenolic compounds, and have superior taste and sensory characteristics. Protein content, fatty acid composition by gas chromatography, phenolic profile by high-pressure liquid chromatography, total phenolic content, and antioxidant activity by spectrophotometric method, and in vitro gastrointestinal digestion along with sensory characteristics by quantitative descriptive test were evaluated. The protein contents of the prepared pistachio milk were 3.78% and 3.09% for Antep and Siirt varieties of pistachio milk, respectively. The most abundant fatty acid was oleic acid and the most abundant phenolic compounds were catechin and gallic acid. Moreover, the bio-accessibilities of total phenolic content were 201% and 213% and the bio-accessibility of total antioxidant activity was 200% and 205% for plant-based milk of Antep and Siirt pistachio, respectively. The overall sensory acceptance scores of the prepared pistachio milk were 5.6-7/10. These findings indicated that plant-based milk substitutes prepared from Antep and Siirt pistachio were higher in protein, had highly bioaccessible phenolic content, and overall accepted in terms of sensory except color challenges.

In this White Paper we present the potential of the enhanced X-ray Timing and Polarimetry(eXTP) mission for studies related to Observatory Science targets. These include flaring stars, supernova remnants, accreting white dwarfs, low and high mass X-ray binaries, radio quiet and radio loud active galactic nuclei, tidal disruption events, and gamma-ray bursts. eXTP will be excellently suited to study one common aspect of these objects: their often transient nature. Developed by an international Consortium led by the Institute of High Energy Physics of the Chinese Academy of Science, the eXTP mission is expected to be launched in the mid 2020s.
