Quantitative assessment of the breast cancer marker HER2 using a gold nanoparticle-based lateral flow immunoassay

Human epidermal growth factor receptor 2(HER2)is an important biomarker for detection and treatment of breast *** this study,we developed monoclonal antibodies against the extracellular domain(ECD)of HER2 and established a rapid and accurate lateral flow immunoassay(LFIA)for use in community medical *** gene sequence of human HER2-ECD was obtained from the National Center for Biotechnology Information(NCBI)to construct the expression ***2-ECD protein expressed in HEK293F cells was used to immunize BALB/c *** monoclonal antibodies were produced in mouse ascites and isolated by hybridoma cell *** were analyzed for purity by SDS-PAGE(sodium dodecyl sulphate-polyacrylamide gel-electrophoresis)and affinity was assessed by enzyme-linked immunosorbent assay(ELISA)while subtypes were detected using the commercial *** HER2-ECD test strip was prepared based on the sandwich method and evaluated using a portable detection *** affinity of the paired antibodies,4D8 and 8D9,both reached 1×108 L/*** antibodies specifically recognized the HER2-ECD protein in *** limit of detection(LOD)of the gold nanoparticle(AuNP)-based LFIA was 1.7 ng/mL with a detection range of 1.7-400 ng/mL,and the performance of the HER2-ECD strip correlated well with that of a Siemens chemiluminescent immunoassay(CLIA)*** conclusion,the paired antibodies were successfully prepared with high affinity and *** AuNP-based LFIA of HER2-ECD provides a fast and accurate method to detect the concentration of HER2-ECD in serum samples for clinical use in community medical institutions,and could contribute to determining the progress of the disease or the effectiveness of treatment.

In this study,we investigated the in vivo behaviors of chiral cobalt oxide nanoparticles(Co_(3)O_(4) NPs)stabilized with D-or L-cysteine(denoted D-or L-NPs,respectively),as representative chiral metal oxide *** Co_(3)O_(4) NPs exerted no observable cytotoxicity within the tested dose *** D-NPs and L-NPs were internalized by dendritic cells through caveola-dependent endocytosis,and L-NPs entered the cells faster than ***,a metabolic analysis indicated that chiral L-NPs had the same effect on the level of bile acids in the liver as D-NPs,which is attributed to the almost equal amount of farnesoid X receptor(FXR)induced by the chiral *** study suggests that low chiroptical activity nanomaterials would not affect the metabolism and kinetics under physiological conditions even if there is some difference in their transport.

Gold-nanoparticle-based multiplex immuno-strip biosensor for simultaneous determination of 83 antibiotics

Antibiotic residues,generated by the irrational use of drugs and environmental pollution,have always been a great challenge to aquaculture ***,a quick,convenient,and performance-excellent way to detect antibiotic residues in aquaculture fish is urgently *** this study,a multiplex immunochromatographic strip biosensor based on gold nanoparticles was developed for the simultaneous detection of five classes of antibiotic residues(24β-lactam antibiotics,26 sulfonamides,five tetracyclines,24 quinolones,and four amphenicols)in aquaculture fish within 10 *** detection ranges of five representative antibiotics,penicillin G,sulfamethazine,tetracycline,enrofloxacin,and chloramphenicol,were 2.33–38.4,0.688–17.1,1.4–48.1,1.45–32.9,and 0.537–9.06μg/kg,*** accuracy and stability of these measurements were demonstrated by analyzing spiked fish samples,with recovery rates of 87.5%–115.2%and a coefficient of variation<9.5%.The cross-reaction rates,based on the five representative antibiotics,were 3.77%–202%forβ-lactams,3.95%–137%for sulfonamides,9.19%–100%for tetracyclines,4.9%–145%for quinolones,and 3.2%–100%for *** excellent testing performance of the biosensor strip to most of antibiotic residues in aquaculture fish ensures they meet the maximum residue limits required by countries or ***,this multiplex immunochromatographic strip biosensor is potentially applicable to the rapidly on-site determination of antibiotic residues in aquaculture fish.

A gold-based multiplex immunochromatographic sensor for detecting paclitaxel

Paclitaxel(PTX),methotrexate(MTX),and 5-fluorouracil(5-FU)are commonly-used small molecule anti-tumor drugs for breast ***,drug resistance occurs with long-term treatment or excessive use,and the high concentrations of PTX,MTX,and 5-FU in patients may lead to side effects with dose-limiting toxicities,such as myelosuppression,hepatotoxicity,and peripheral ***,concentration monitoring of PTX,MTX,and 5-FU in clinical treatment is very *** prepared highly sensitive and specific monoclonal antibodies using several haptens,and established a multiple paper-based sensor utilizing optical signals of gold nanoparticles,which simultaneously detected PTX,MTX,and 5-FU in human plasma or urine with 10 min.A portable scanner for quantitative detection in plasma and urine samples was used,and the calculated limit of detection(cLOD)values were 0.002 and 0.142μg/mL for PTX,0.187 and 0.976 ng/mL for MTX,and 0.057 and 0.128μg/mL for 5-FU,*** the recovery test,the recovery rates and the coefficient of variation were 85.84%–108.81%and 1.23%–8.80%,respectively,indicating the reliability and accuracy of the multiple gold immunochromatographic strip(GIS).In addition,for the determination of collected clinical plasma samples,the correlation between the test data of the multiple GIS and liquid chromatography–tandem mass spectrometry(LC–MS/MS)was ***,the developed multiple GIS could be applied to the rapid detection of PTX,MTX,and 5-FU in different clinical samples.

Gold-based lateral-flow strip for the detection of penconazole in watermelon and cucumber samples

As a typical triazole fungicide,penconazole(PEN)is widely used in agriculture but has been proven to be *** this study,we designed a new hapten to prepare a highly sensitive and specific anti-PEN monoclonal antibody(mAb)and established a gold nanoparticle-based lateral-flow immunoassay(LFIA)for the detection of PEN residues in watermelon and *** 50%inhibitory concentration(IC_(50))of the mAb was 0.42 ng/mL and the LFIA strip had a visual limit of detection(vLOD)of 2.5 ng/g and a cut-off value of 10 ng/g in watermelon and *** calculated limit of detection(LOD)of the LFIA strip was 0.36 ng/g for watermelon and 0.29 ng/g for *** LFIA strip also gave a re-covery rate of 92.5%-109.0%for watermelon samples and 92.5%-106.7%for cucumber *** results using the LFIA strip are highly consistent with those seen using liquid chromatography-tandem mass *** our developed LFIA strip represents a potentially reliable tool for the rapid on-site screening for PEN in watermelons and cucumbers.

Electromagnetic field-enhanced chiral dimanganese trioxide nanoparticles mitigate Parkinson’s disease

The misfolding and aggregation ofα-synuclein(α-syn)is closely associated with Parkinson’s disease(PD).Here,chiral dimanganese trioxide(Mn_(2)O_(3))nanoparticles(NPs)were prepared for PD treatment enhanced by a noninvasive electromagnetic field(MF).The affinity constants of D-NPs towardα-syn monomer(mono)orα-syn fibril were 3.5 times or 5.2 times higher,respectively,than those of L-NPs,and the mechanical force generated by NPs under a MF further promoted the interaction between NPs andα-syn to amplify the difference between L-NPs and *** the synergy effect of the preferentially affinity ability and MF-induced mechanical forces,D-NPs exhibited a better inhibitory efficiency onα-syn fibrillization than ***,after differentially cellular uptake of L-/D-NPs via the caveolin-mediated pathway,as reactive oxygen species(ROS)-scavengers,D-NPs possess higher efficiency in decreasing intracellular ROS level than L-NPs to provide higher cytoprotective efficiency to neuron *** vivo data showed that after treatment with D-NPs under a MF for 60 days,α-syn concentration in the cerebrospinal fluid of PD mice decreased 81%,while dopamine level in the brain of PD mice increased *** findings indicated the potential of utilizing the synergic interplay of chiral NPs and MF for treating disease and opened a new path to explore the nanoscale chirality for regulating the biological effect.

Protein aggregation causes alpha-synuclein(α-syn)to change from its original physiological role to a pathological state,which is a potential pathogenic mechanism in Parkinson’s disease(PD).Chiral _(L/D)-Cu_(x)Co_(y)S supraparticles(_(L/D)-SPs)with a circular dichroism value of 35 mdeg at 805 nm were fabricated using a simple wet-chemical *** _(L/D)-SPs prevented the α-syn monomers from forming fibrils and triggered the α-syn fibrils to turn into monomers under 808 nm near-infrared(NIR)*** living MN9D cells,D-SPs reduced cellular damage,neuronal functional deficits,and neuron loss caused by α-syn fibrils after NIR spectroscopy treatment within 10 min to prevent α-syn ***,the reactive oxygen species produced by _(D)-SPs were 1.42 times higher than those produced by _(L)-*** vivo experiments showed that _(D)-SPs had a protective effect on neuron damage caused by α-syn aggregate deposition,reduced the symptoms in a mouse model of PD,and restored cognitive *** NIR light treatment,the amount of α-syn in a mouse model of PD decreased by more than 67.5%.At the same time,_(D)-SPs gradually decomposed into small nanoparticleswithin 60 days and were excreted through the blood-brain *** discovery paves theway for the treatment of neurodegenerative diseases using chiral SPs under NIR light irradiation.

Wheat gliadin is one of the most important causes of gluten *** study aimed to develop a sandwich enzyme-linked immunosorbent assay(ELISA)and gold nanoparticle-based lateral flow(LFIA)strips to detect wheat allergen in *** cell screening,ten monoclonal antibodies(mAbs)against gliadin and after optimization were obtained,a pair of mAbs suitable for its detection(capture antibody:mAb 7;detection antibody:HRP-labeled mAb 6)were *** on two of these antibodies,a sandwich ELISA with limit of detection(LOD)of 60 ng/mL in negative milk,and more importantly,negligible cross-reactivity to other allergens was *** average recoveries for gliadin in negative milk were 99.16%-100.07%using the sandwich *** also developed LFIA strips for the rapid detection of gliadin with visual limit of detection(vLOD)of 25 ng/mL and calculated LOD value of 6.56 ng/mL in negative *** test results of positive samples obtained from the LFIA strips were highly consistent with those of the sandwich ***,the developed LFIA strip is an effective and reliable tool for the rapid and on-site detection of wheat allergen in milk.
