Different responses of arbuscular mycorrhizal fungal community to day-time and night-time warming in a semiarid steppe

Arbuscular mycorrhizal(AM) fungi form mutualistic symbioses with most plant species and play important roles in *** of the response of AM fungi to temperature change will improve our understanding of the function of AM fungal community under global climate change scenarios in *** effects of constant warming on AM fungal communities have been investigated previously,but responses to asymmetrical warming over 24-h periods have never been documented in natural *** this study,we examined AM fungal communities in a full factorial design including day-time and night-time warming in a semiarid steppe in northern ***-time and 24-h warming,but not night-time warming,significantly increased AM fungal spore *** contrast,none of the three warming regimes had a significant effect on AM fungal extra radical hyphal *** of 161 operational taxonomic units(OTUs) of AM fungi were recovered by 454 pyrosequencing of 18 S r ***-time,night-time,and 24-h warming all significantly increased AM fungal OTU *** AM fungal OTUs showed a significant bias toward day-time,night-time or24-h *** AM fungal community composition was significantly affected by night-time warming,but not by day-time and 24-h *** finding highlighted different responses of AM fungal spore density and community composition to asymmetrical *** study might improve our understanding of ecosystem functioning of AM fungal community under global climate change scenarios in a semiarid steppe ecosystem.

Infectious in vitro transcripts from cloned cDNA of beet necrotic yellow vein virus RNA3 and RNA4and their functional study

The full-length double-stranded cDNAs of beet necrotie yellow vein virus RNA3 and RNA 4were synthesized by using oligo(dT) 15 as well as RNA3 and RNA4 specific primers, and cloned downstream of the bacteriophage Sp6 RNA polymerase promoter of the transcription vector pGEM3Zf(+). The in vitro "run-off" transcription products obtained in the presence of Sp6 RNA polymerase and template DNA have high biological activities. In the 2 transcription systems, the transcription and capping in 2 separate reactions are more efficient. The simultaneous transcription and capping are not so efficient, but the transcripts are more infectious. Although there are a number of nonviral nucleotide sequences at the 5- and 3’-ends of RNA3 and RNA4 transcripts, the biological activities of both transcripts were not affected. The mechanical coinoculation of sugarbeet roots with the infectious transcripts of the prepared RNA3 and RNA4 and BNYW Rgl isolate has confirmed that RNA3 is the main cause of sugarbeet rhizomania.

Delayed resistance to Cucumber mosaic virus mediated by 3′ UTR-derived hairpin RNA

RNA silencing has been shown to function in the plant antivirus defense response,leading to viral RNA degradation induced by vsiRNA-containing RISC cleavage *** mosaic virus(CMV) 3′UTR sequences share a high conservation of nucleotide sequence and secondary structures that are important for CMV ***,in an attempt to simultaneously target the multiple genomic and subgenomic RNAs of CMV for degradation,CMV 3′UTR were used to design hairpin RNA(hpRNA) to transform tobacco(***) so as to constitutively produce viral *** of the transgenic plants expressing CMV Q strain(Q-CMV,subgroup Ⅱ strain) RNA3 3′UTR-derived hpRNA showed de-layed resistance to Q-CMV infection and exhibited recovery *** with Q-CMV-in-oculated leaves,the upper leaves showed weak or no disease symptoms and a reduced accumulation level of viral *** with transient assays,our results indicate that the 3′UTR-derived siRNAs were biologically active in targeting viral RNA for *** resistance in transgenic plants was also observed against subgroup IB strain SD-CMV infection,indicating a broad-spectrum anti-CMV effect of the 3′UTR-based antiviral *** blot assays indicated that there was no strong correlation between the degree of resistance and the accumulation level of 3′UTR-derived siRNAs,suggesting that to target a highly structured RNA,such as the CMV 3′UTR,the quantity of siRNAs may not be the only determinant of silencing *** RNA secondary structures may also affect target accessibility,siRNA-containing RISC-target recognition and the consequent antiviral effect.

Mapping paddy rice distribution using multi-temporal Landsat imagery in the Sanjiang Plain, northeast China

Information of paddy rice distribution is essential for food production and methane emission calculation. Phenology-based algorithms have been uti- lized in the mapping of paddy rice fields by identifying the unique flooding and seedling transplanting phases using multi-temporal moderate resolution （500m to 1 km） images. In this study, we developed simple algorithms to identify paddy rice at a fine resolution at the regional scale using multi-temporal Landsat imagery. Sixteen Landsat images from 2010-2012 were used to generate the 30 m paddy rice map in the Sanjiang Plain, northeast China-one of the major paddy rice cultivation regions in China. Three vegetation indices, Normalized Difference Vegetation Index （NDVI）, Enhanced Vegetation Index （EVI）, and Land Surface Water Index （LSWI）, were used to identify rice fields during the flooding/transplanting and ripening phases. The user and producer accuracies of paddy rice on the resultant Landsat-based paddy rice map were 90% and 94%, respectively. The Landsat-based paddy rice map was an improvement over the paddy rice layer on the National Land Cover Dataset, which was generated through visual interpretation and digitalization on the fine-resolution images. The agricultural census data substantially underreported paddy rice area, raising serious concern about its use for studies on food security.

Influence of Garlic Intercropping or Active Emitted Volatiles in Releasers on Aphid and Related Beneficial in Wheat Fields in China

In order to develop biological control of aphids by a ＂push-pull＂ approach, intercropping using repellent emitting plants was developed in different crop and associated plant models. Garlic is one of the potential plant that could be inserted in crops to decrease the pest occurrence in neighboring crop plots. In this study, field works were conducted in wheat fields in Langfang Experimental Station, Hebei Province in China from October 2009 to July 2010 during wheat developmental season. The effect of wheat intercropping with garlic but also the volatiles emission on the incidence of the English grain aphid, Sitobion avenae Fabricius （Homoptera： Aphididae） was assessed. Natural beneficial occurrence and global yields in two winter wheat varieties that were susceptible or resistant to cereal aphid were also determined comparing to control plots without the use of garlic plant intercrop nor semiochemical releaser in the fields. S. avenae was found to be lower in garlic oil blend treatment （GOB）, diallyl disulfide treatment （DD） and wheat-garlic intercropping treatment （WGI） when compared to the control plots for both two varieties （P〈0.01）. Both intercropping and application of volatile chemicals emitted by garlic could improve the population densities of natural enemies of cereal aphid, including ladybeetles and mummified aphids. Ladybeetle population density in WGI, GOB and mummified aphids densities in WGI, DD were significantly higher than those in control fields for both two varieties （P〈0.05）. There were significant interactions between cultivars and treatments to the population densities of S. avenae. The 1 000-grain weight and yield of wheat were also increased compared to the control. Facile synthesis of enzyme functional metal-organic framework for colorimetric detecting H_2O_2 and ascorbic acid

In this work,a metal-organic frameworks material MIL-88 was prepared easily using solvent-thermal method,and was first found to have catalytic activities similar to those of biological enzymes such as catalase and *** material was characterized by XRD,SEM,TEM,EDX,FT-IR techniques and an N2 adsorption *** exhibited peroxidase-like activity through catalytic oxidation of the peroxidase substrate 3,3＇,5,5＇-tetramethylbenzidine（TMB） in the presence of H2O2,producing a blue-colored *** optimal conditions,the absorbance at 652 nm is linearly correlated with the concentration of H2O2 from 2.0×10^-6 mol/L to 2.03×10^-5 mol/L（R-2=0.981） with a detection limit of 5.62×10^-7 mol/L（S/N=3）.More importantly,a sensitive and selective method for ascorbic acid detection was developed using this material as a *** analytical method for ascorbic acid detection was observed to have a linear range from 2.57×10^-6 mol/L to 1.01×10^-5 mol/L（R-2=0.989） with a detection limit of 1.03×10^-6 mol/L（S/N=3）.This work suggests MOFs have advantages of preparing biomimetic catalysts and extends applications of the functional MOFs in the field of biosensor.

The Russian dandelion Taraxacum kok-saghyz Rodin(TKS), a member of the Composite family and a potential alternative source of natural rubber(NR) and inulin, is an ideal model system for studying rubber biosynthesis. Here we present the draft genome of TKS, the first assembled NR-producing weed plant. The draft TKS genome assembly has a length of 1.29 Gb, containing 46 731 predicted protein-coding genes and68.56% repeats, in which the LTR-RT elements predominantly contribute to the genome enlargement. We analyzed the heterozygous regions/genes, suggesting its possible involvement in inbreeding *** comparative studies between rubber-producing and non-rubber-producing plants, we found that enzymes of the mevalonate(MVA) pathway and rubber elongation might be critical for rubber biosynthesis, and several key isoforms have been isolated and shown to be predominantly expressed in the latex, indicating their crucial functions in rubber biosynthesis. Moreover, for two important families in rubber elongation, the CPT/CPTL and REF/SRPP families, diverse evolutionary tracks have been revealed. These results provide valuable resources and new insights into the mechanism of NR biosynthesis,and facilitate the development of alternative NR-producing crops.

A five-gene cluster cvhABCDE was identified from Streptomyces hygroscopicus *** thefirst gene of this cluster,cvhA encoded a putative sensor histidine kinase with a predicted sensor domainconsisting of two trans-membrane segments at the N-terminus and a conserved HATPasecdomain at the *** C-terminus polypeptide of CvhA expressed in Escherichia coli was purified and shown to beautophosphorylated with [γ-32P]ATP in *** phosphoryl group was acid-labile and basic-stable,whichsupported histidine as the phosphorylation *** obvious difference of mycelia development was ob-served between the null mutant of cvhA generated by targeted gene replacement and the wild-type parentalstrain 10-22 grown on solid soya flour medium with 2%-8% glucose or sucrose,but the cvhA mutant couldform much more abundant aerial mycelia and spores than the wild-type strain on solid soya flour mediumsupplemented with 6%-8% mannitol,6%-8% sorbitol,4%-6% mannose,or 4%-6% *** pheno-type was complemented by the cloned wild-type cvhA gene,and no difference was observed for growthcurves of the cvhA mutant and the wild strain in liquid minimal medium with the tested sugars at a concen-tration of 4%,6% and 8%.We thus propose that CvhA is likely a sensor histidine kinase and negativelyregulates the morphological differentiation in a sugar-dependent manner in *** 10-22.

Structure, expression pattern and chromosomal localization of the rice Osgrp-2 gene

Glycine-rich proteins (GRPs) belong to a kind of important structural proteins of plant cell walls. The expression of GRP genes is regulated spatially and developmentally as well as by various environmental stresses, thus providing a good model for the study of plant gene expres-sion. We obtained the genomic sequence of a new GRP gene (Osgrp-2) from a rice genomic li-brary. The transcription start site of Osgrp-2 was determined by 5’-rapid amplification of cDNA ends (RACE) and a 2.4-kb promoter sequence was thus delimited. The spatial and developmental expression pattern as well as the wound-inducible character of Osgrp-2 in rice plants was ana-lyzed in detail. Furthermore, the gene was mapped onto rice chromosome 10 by analysis of re-striction fragment length polymorphism (RFLP).
