The gut microbiota plays a pivotal role in the maintenance of health for amphibians,and it has been fully recognized,but the effectiveness of various influencing factors has not yet been fully *** this association should be considered while the amphibian order Caudata is facing a severe situation of population decline and extinction,there is little understanding of the association between diets and the diversity of gut microbiota in the amphibian order ***,we conducted an extensive analysis of the gut microbiota of Cynops orientalis fed different diets using functional prediction and 16S rRNA amplicon sequencing ***,we found that wild individuals had greater gut microbial diversity and richness in comparison to captive ***,we identified the bacterial taxa associated with diets and observed differences in the relative abundance of gut microbiota among people on various ***,we have a predictive comprehension of the selection and adaptative significance of shared core ASVs in the gut microbiota in maintaining the healthy survival of *** in a large-scale spatiotemporal *** study emphasizes how diets alter the gut microbiota of Caudata and offers fresh perspectives on the conservation and captive management of species in Caudata.

Genome editing in plants using the TnpB transposase system

The widely used clustered regularly interspaced short palindromic repeats(CRISPR)/CRISPR-associated nuclease(Cas)system is thought to have evolved from IS200/IS605 *** proteins,encoded by one type of IS200/IS605 transposon,are considered to be the evolutionary ancestors of Cas12 nucleases,which have been engineered to function as RNA-guided DNA endonucleases for genome editing in bacteria and human *** nucleases,which are smaller than Cas nucleases,have been engineered for use in genome editing in animal systems,but the feasibility of this approach in plants remained ***,we obtained stably transformed genome-edited mutants in rice(Oryza sativa)by adapting three recently identified TnpB genome editing vectors,encoding distinct TnpB nucleases(ISAam1,ISDra2,and ISYmu1),for use in plants,demonstrating that the hypercompact TnpB proteins can effectively edit plant ***2 and ISYmu1 precisely edited their target sequences,with no off-target mutations detected,showing that TnpB transposon nucleases are suitable for development into a new genome editing tool for *** modifications improving the genome-editing efficiency of the TnpB system will facilitate plant functional studies and breeding programs.

Bird specimen number linked with species trait and climate niche breadth

Biological specimens are fundamental for taxonomy and flora/fauna *** importantly,they also play crucial roles in recording environmental impacts on morphology and behavior,which is vital for biodiversity research and ***,there are few systematic studies on the patterns and drivers of bird specimen number at regional *** study is the first attempt to examine the relationships between bird specimen number and species traits as well as climate niche breadth in China,aiming to answer two questions:1)how do species’temperature niche breadth and precipitation niche breadth influence specimen number?2)which trait is most associated with bird specimen number?The associations between bird specimen number and explanatory variables were examined using ordinary least squares,generalized linear models,phylogenetic generalized linear mixed models,and multiple *** results showed that Muscicapidae was the family with the highest specimen number,and Passeriformes was the order with the highest specimen *** specimen number significantly increased with larger temperature niche breadth and precipitation niche *** number was also positively associated with geographic range size,habitat specificity,hunting vulnerability and clutch size,but negatively associated with body *** findings suggest that future bird specimen collection should pay more attention to birds with limited ecological niches,large body sizes,and small clutch *** research enhances the use of bird specimen data to study and preserve biodiversity.

Genetic mechanism of body size variation in groupers:Insights from phylotranscriptomics

Animal body size variation is of particular interest in evolutionary biology,but the genetic basis remains largely *** studies have shown the presence of two parallel evolutionary genetic clusters within the fish genus Epinephelus with evident divergence in body size,providing an excellent opportunity to investigate the genetic basis of body size variation in ***,we performed phylotranscriptomic analysis and reconstructed the phylogeny of 13 epinephelids originating from the South China *** genetic clades with an estimated divergence time of approximately 15.4 million years ago were correlated with large and small body size,respectively.A total of 180 rapidly evolving genes and two positively selected genes were identified between the two *** enrichment analyses of these candidate genes revealed distinct enrichment categories between the two *** pathways and genes may play important roles in body size variation in groupers through complex regulatory *** on our results,we speculate that the ancestors of the two divergent groups of groupers may have adapted to different environments through habitat selection,leading to genetic variations in metabolic patterns,organ development,and lifespan,resulting in body size divergence between the two locally adapted *** findings provide important insights into the genetic mechanisms underlying body size variation in groupers and species differentiation.

Smarter in the city?Lizards from urban and semi-natural habitats do not differ in a cognitive task in two syntopic species

Urbanization occurs at a global scale,imposing dramatic and abrupt environmental changes that lead to biodiversity ***,some animal species can handle these changes,and thrive in such artifcial *** possible explanation is that urban individuals are equipped with better cognitive abilities,but most studies have focused on birds and mammals and yielded varied *** have received much less attention,despite some lizard species being common city *** Italian wall lizard,Podarcis siculus,and the common wall lizard,Podarcis muralis,are two successful lizards in anthropogenic habitats that thrive in urban *** test for differences in a cognitive skill between urban and semi-natural environments,we investigated inhibitory control through a detour task in syntopic populations of the two species,across 249 lizards that were tested in partially artifcial feld *** inhibitory control is considered essential for higher degrees of cognitive fexibility and other higher-level cognitive *** this task,we confronted lizards with a transparent barrier,separating them from a desired shelter area that they could only reach by controlling their impulse to go straight and instead detour the *** found no differences between lizards in urban and semi-natural environments,nor between species,but females overall performed better than ***,48%of the lizards in our study did not perform a correct trial in any of the 5 trials,hinting at the diffculty of the task for these spe*** study is among the frst to address lizard cognition,through their inhibitory control,as a potential explanation for success in cities and highlights one should be careful with assuming that urban animals generally have enhanced cognitive performance,as it might be taxa,task,or condition dependent.

Reshaping the active pocket of an aldo-keto reductase to enhance its detoxification activity towards 3-keto-deoxynivalenol

Deoxynivalenol （DON）， the most common mycotoxin in the world， poses serious health hazards to both humans and animals and causes significant economic losses to the food and feed industry. An intriguing method for DON detoxification involves enzymatic cascade catalysis by dehydrogenase （DADH） and aldo-keto reductase （AKR13B3）. However， the poor catalytic activity of AKR13B3 has limited its usefulness. Here， structure-guided steric hindrance engineering， computer-assisted protein engineering， and combinatorial mutagenesis were carried out to increase the catalytic efficiency of AKR13B3. The best mutant in this project was M28S/S65V， which outperformed wild-type AKR13B3 in terms of catalytic efficiency (kcat/Km) and specific activity by factors of 45.08 and 43.11， respectively. Kinetic parameter determination revealed that the enhanced catalytic efficiency of M28S/S65V toward 3-keto-DON was attributed to the decreased Kmand increased kcatvalues. Moreover， the engineered enzyme was applied to degrade DON in contaminated corn steep liquor， for a 90.62% removal rate. Structure-based computational analysis provided insights into the enhanced catalytic efficiency of M28S/S65V， which was attributed to the enlarged substrate-binding pocket and reshaped pocket with a favorable hydrophilic attack distance. The results provide a reference for improving the catalytic activity of an enzyme toward non-native bulky substrates and pave the way for the development of enzymes as detoxification agents to mitigate DON in the food and feed industry.

Genetic load and inbreeding are recognized as important factors to be considered in conservation *** levels of both can increase the risk of population extinction by negatively impacting fitness-related characters in many species of plants and animals,including humans(inbreeding depression).Genomic tech-niques are increasingly used in measuring and understanding genetic load and inbreeding and their importance in evolution and *** used whole genome resequencing data from two sibling grouse species in subarctic Eurasia to quantify *** found a large range of inbreeding measured as FROH(fraction of runs of homozygosity)in individuals from different populations of Chinese Grouse(Tetrastes sewerzowi)and Hazel Grouse(***).FROH estimated from genome-wide runs of homozygosity(ROH)ranged from 0.02 to 0.24 among Chinese Grouse populations and from 0.01 to 0.44 in Hazel *** from a population of Chinese Grouse residing in the Qilian mountains and from the European populations of Hazel Grouse(including samples from Sweden,Germany and Northeast Poland)were the most inbred(FROH ranged from 0.10 to 0.23 and 0.11 to 0.44,respectively).These levels are comparable to other highly inbred populations of *** Grouse from northern China and Chinese Grouse residing in the Qinghai-Tibetan Plateau showed relatively lower inbreeding *** of the ratio between deleterious missense mutations and synonymous mutations revealed higher levels in Chinese Grouse as compared to Hazel *** results are possibly explained by higher fixation rates,mutational melt down,in the range-restricted Chinese Grouse compared to the wide-ranging Hazel ***,when we compared the relatively more severe class of loss-of-function muta-tions,Hazel Grouse had slightly higher levels than Chinese Grouse,a result which may indicate that purifying selection(purging)has been more efficient in Chinese Grouse on this class of mutations.

Odor cues rather than personality affect tadpole deposition in a neotropical poison frog

animals constantly need to evaluate available external and internal information to make appropriate ***,assessing,and acting on relevant cues in contexts such as mate choice,intra-sexual competition,and parental care is particularly important for optimizing individual reproductive *** factors can infuence decision-making,such as external environmental cues and the animal’s own internal state,yet,we have limited knowledge on how animals integrate available ***,we used an entire island population(57 males,53 females,and 1,109 tadpoles)of the neotropical brilliant-thighed poison frog Allobates femoralis to investigate how 2 factors(olfactory cues and personality traits)infuence the ability of males to fnd and use new resources for tadpole *** experimentally manipulated the location of tadpole deposition sites and their associated olfactory cues,and repeatedly measured exploration and boldness in adult *** further reconstructed tadpole deposition choices via inferred parent–offspring relationships of adult frogs and tadpoles deposited in our experimental pools using molecular parentage *** found that the discovery and use of new rearing sites were heavily infuenced by olfactory cues;however,we did not fnd an effect of the measured behavioral traits on resource discovery and *** conclude that in highly dynamic environments such as tropical rainforests,reliable external cues likely take priority over personality traits,helping individuals to discover and make use of reproductive resources.
