Profile of Prosthetic Care Consumers at the institute of Odontology and stomatology Clinic of Dakar

The high frequency of dental extractions creates a high demand for prosthetic rehabilitation. Therefore, this study aimed to describe the profile of consumers of prosthetic care at the institute of Odontology and stomatology in Dakar. It was a descriptive and retrospective study of patients who had been consulted at the prosthetic clinic of the institute of Odontology and stomatology of Dakar. Data were collected from medical records from 09 November 2019 to 09 June 2020. The tools used to collect and process the information were the Sphinx software and the Excel spreadsheet. Out of the 101 patient records, 51% were male and 48% belonged to the age group of 18 to 39 years. Nearly 38.61% of the patients lived less than 5 km from the health care facility and 70.3% of the respondents had a monthly income of fewer than 100,000 FCFA. Clinically, 96% of patients came for functional reasons, 78.2% for aesthetic reasons, and partial edentulism was the most frequent diagnosis (96%). Chi-square tests were performed to find a dependency or non-dependency relationship between diagnosis and age, but also between income and treatment cost. In sum, age and diagnosis were independent of each other, just as patient income was not related to treatment cost. Proximity to health facilities, income, and cost of treatment are factors that influence accessibility to oral health care. It is important to integrate the profile of the consumer of care into global management.

The oral cavity is a complex physiological community encompassing a wide range of *** of oral microbiota can lead to various oral infectious diseases,such as periodontitis and tooth decay,and even affect systemic health,including brain aging and neurodegenerative *** studies have highlighted how oral microbes might be involved in brain aging and neurodegeneration,indicating potential avenues for intervention *** this review,we summarize clinical evidence demonstrating a link between oral microbes/oral infectious diseases and brain aging/neurodegenerative diseases,and dissect potential mechanisms by which oral microbes contribute to brain aging and *** also highlight advances in therapeutic development grounded in the realm of oral microbes,with the goal of advancing brain health and promoting healthy aging.

Hydrogel scaffolds have numerous potential applications in the tissue engineering ***,tough hydrogel scaffolds implanted in vivo are seldom reported because it is difficult to balance biocompatibility and high mechanical *** by Chinese ramen,we propose a universal fabricating method(printing-P,training-T,cross-linking-C,PTC&PCT)for tough hydrogel scaffolds to fill this ***,3D printing fabricates a hydrogel scaffold with desired structures(P).Then,the scaffold could have extraordinarily high mechanical properties and functional surface structure by cycle mechanical training with salting-out assistance(T).Finally,the training results are fixed by photo-cross-linking processing(C).The tough gelatin hydrogel scaffolds exhibit excellent tensile strength of 6.66 MPa(622-fold untreated)and have excellent ***,this scaffold possesses functional surface structures from nanometer to micron to millimeter,which can efficiently induce directional cell ***,this strategy can produce bionic human tissue with mechanical properties of 10 kPa-10 MPa by changing the type of salt,and many hydrogels,such as gelatin and silk,could be improved with PTC or PCT *** experiments show that this scaffold can effectively promote the new generation of muscle fibers,blood vessels,and nerves within 4 weeks,prompting the rapid regeneration of large-volume muscle loss injuries.

Cytotoxic Effects of Biodegradation of Pure Mg and MAO-Mg on Tumor Cells of MG63 and KB

Magnesium （Mg） based metals possess unique features of biodegradation that can be used for different purposes. In this study, two tumor cells, MG63 and KB, were cultured directly on a pure Mg with and without MAO coating to evaluate their cytotoxic effects on the tumor cells. It was found that both MG63 cells and KB cells adhered better on the surface of the coated Mg samples than those on the pure Mg samples. However, the survivals of two kinds of tumor cells were obviously inhibited on both Mg samples with and without coating, compared with that on the pure titanium. In addition, the survival of MG63 cells was more susceptive to the Mg degradation compared with the KB cells. Thus, the application of such effect should take account of that the degradation of Mg based metal may result in different cytotoxic effect on different cells.

Crosstalk between adipose-derived stem cells and chondrocytes:when growth factors matter

Adipose-derived stem cells（ASCs）and mesenchymal stem cells are promising for tissue repair because of their multilineage differentiation *** previous data confirmed that the implantation of mixed ASCs and chondrocytes into cartilage defects induced desirable in vivo healing ***,the paracrine action of ASCs on chondrocytes needs to be further *** this study,we established a co-culture system to achieve cell-to-cell and cell-to-tissue crosstalk and explored the soluble growth factors in both ASCs and chondrocytes supplemented with 1%fetal bovine serum to mimic the physiological *** ASCs,we screened for growth factors by semi-quantitative PCR and quantitative real-time PCR and found that the expression of bone morphogenetic protein 2（BMP-2）,vascular endothelial growth factor B（VEGFB）,hypoxia inducible factor-1α（HIF-1α）,fibroblast growth factor-2（FGF-2）,and transforming growth factor-β1 significantly increased after co-culture in comparison with *** chondrocytes,VEGFA was significantly enhanced after ***,the expression of collagen II and aggrecan was significantly down-regulated in the co-culture group compared with the mono-culture ***,among all the growth factors screened,we found that the BMP family members BMP-2,BMP-4,and BMP-5 were down-regulated and that VEGFB,HIF-1α,FGF-2,and PDGF were significantly decreased after *** results suggest that crosstalk between ASCs and chondrocytes is a pathway through the regulated growth factors that might have potential in cartilage repair and regeneration and could be useful for tissue engineering.

Comparisons of Condylar Movements with the Functional Occlusal Clutch and Tray Clutch Recording Methods in CADIAX~ system

Aim The purpose of this study is to compare the effects of the two clutches on recording the condylar movement. Methodology Ten subjects （6 women,4 men; mean age 25.4 years） participated in the study. The mandibular move-ment,sagittal condylar inclination angle,and transversal condylar inclination angle of each subject were recorded with the CADIAX？ using the two clutches,respectively. The characteristics of the tracings of the protrusion,opening,and mediotrusion were analyzed with the t-test statistics at α = 0.05 level. The Kappa values were calculated for an assessment of the congruence of the tracings. Results The results showed that the contour,direction,and dimension of the tracings in the two clutches were approxi-mately same,but the tracings determined by the functional occlusal clutch were more regular and congruent. In the group segment recorded with the tray clutch,opening/ closing paths of one subject showed crossed and time curves of three subjects appeared peak-like changes of velocity,but none were statistically different （P0.05）. Conclusion The research suggests that the functional occlusal clutch should be preferred in the evaluation of the mandibular function,as the tracings with the tray clutch are more likely to produce false positive results.

Cytocompatibility and Hemolysis of AZ31B Magnesium Alloy with Si-containing Coating

In the present study, a Si-containing coating was fabricated on AZ31B Mg alloy. Cytocompatibility of the coated alloy was evaluated by both indirect and direct contact methods, respectively. Effects of a number of incubation variables on the sensitivity and reproducibility of the hemolysis test were also examined by using positively and negatively responding biomaterials. Cytocompatibility testing results indicated that cell condition, cell adherence, cell proliferation and extracellular matrix secretion of the coated alloy were improved compared with those of the uncoated alloy for different extraction and co-culture time. The hemolysis test suggested that hemolysis testing conditions were critical to determine the hemolysis of the alloy. It was also found that 1 day in vitro degradation of the uncoated AZ31B alloy had no destructive effect on erythrocyte. As for the coated AZ31B alloy at any time point, the hemolysis rate was much lower than 5%, the safe value for biomaterials. These in vitro experimental results indicate that the Si-containing coating is effective to improve the cytocompatibility and hemolysis behaviors of AZ31B alloy during its degradation.

Controlling inflammation meanwhile facilitating tissue regeneration has been considered as a promising strategy to treat inflammatory bone defect. Herein, we describe the synthesis of a bio-sensitive poly(lactic-co-glycolic acid)/mesoporous silica nanocarriers core-shell porous microsphere(PLGA/MSNsPMS) encapsulated poly(L-lactic acid)(PLLA) spongy nanofibrous micro-scaffold as a new generation of therapeutic platform for effective reconstruction of bone defects caused by periodontal *** PLGA/MSNs-PMS were designed as stimuli-responsive carriers for on-demand co-delivery of multiple biomolecules to provide proper physiological environment, while the multi-level(from macro-,micro-to nanometers) nanofibrous and porous structures in PLLA micro-scaffold were in charge of the reconstruction of ECM, which synergistically contribute to the enhancement of new tissue formation under inflammatory condition. After local injection into periodontal tissue, this construct could sequentially release bone growth factor(BMP-2) as well as anti-inflammatory drug(celecoxib) loaded MSNs in response to the over-expressed matrix metalloproteinases(MMP) in periodontal region. During alveolar bone regeneration induced by BMP-2 and ECM like structure, the MSNs would further deliver celecoxib in target cells to achieve inflammation inhibition, resulting in effective treatment of periodontal disease.

Screening the Optimal Patterned Surfaces Consisting of Cell Morphology Mimicking Micro-pillars and Nanotube Arrays for the Design of Titanium Implants#br#

Micron/nano scale topographic modification has been a significant focus of interest in current titanium(Ti)surface ***,the influence of micron/nano structured surface on cell or bacterium behavior on the Ti implant has rarely been systematically ***,except for popular microgrooves,little work has been carried out on the reaction of cells to the bionic *** this study,several micro-pillars mimicking cell morphology were prepared on Ti surfaces by lithography and contact printing(ICP)method,and they were further decorated with nanotube arrays by anodization *** surface modifications remarkablly increased the surface roughness of pristine Ti surface from 91.17 nm±5.57 nm to be more than 1000 nm,and reduced their water contact angles from 68.3°±0.7°to be 16.9°±2.4°.Then,the effects of these hierarchical micron/nano scale patterns on the behaviors of MG63 osteoblasts,L929 fibroblasts,SCC epithelial cells and *** were studied,aiming to evaluate their performance in osseointegration,gingival epithelial sealing and antibacterial *** an innovative scoring strategy,our findings showed that square micro-pillars with 6μm width and 2μm height combined with 85 nm diameter nanotubes was suitable for implant neck design,while square micro-pillars with 3μm width and 3.6μm height combined with 55 nm diameter nanotubes was the best for implant body *** study reveals the synergistic effect of the hierarchical micron/nano scale patterns on MG63 osteoblasts,L929 fibroblasts,SCC epithelial cells and *** *** provides insight into the design of biomedical implant surfaces.

Novel MSX1 variants identified in families with nonsyndromic oligodontia

The goal of this study was to identify MSX1 gene variants in multiple Chinese families with nonsyndromic oligodontia and analyse the functional influence of these ***-exome sequencing(WES)and Sanger sequencing were performed to identify the causal gene variants in five families with nonsyndromic oligodontia,and a series of bioinformatics databases were used for variant confirmation and functional *** characterization of the members of these families was described,and an in vitro analysis was performed for functional *** novel MSX1 heterozygous variants were identified:three missense variants[c.662A>C(p.Q221P),c.670C>T(p.R224C),and c.809C>T(p.S270L)],one nonsense variant[c.364G>T(p.G122*)],and one frameshift variant[c.277delG(p.A93Rfs*67)].Preliminary in vitro studies demonstrated that the subcellular localization of MSX1 was abnormal with the p.Q221P,p.R224C,p.G122*,and p.A93Rfs*67 variants compared to the wild *** variants(p.Q221P,p.G122*,and p.A93Rfs*67)were classified as pathogenic or likely pathogenic,while p.S270L and p.R224C were of uncertain significance in the current ***,we summarized and analysed the MSX1-related tooth agenesis positions and found that the type and variant locus were not related to the severity of tooth *** results expand the variant spectrum of nonsyndromic oligodontia and provide valuable information for genetic counselling.
