Gut microbiota and circulating metabolite dysbiosis predate important pathological changes in glucose metabolic disorders;however,comprehensive studies on impaired glucose tolerance(IGT),a diabetes mellitus(DM)precursor,are ***,we perform metagenomic sequencing and metabolomics on 47 pairs of individuals with IGT and newly diagnosed DM and 46 controls with normal glucose tolerance(NGT);patients with IGT are followed up after 4 years for progression to *** of baseline data reveals significant differences in gut microbiota and serum metabolites among the IGT,DM,and NGT *** addition,13 types of gut microbiota and 17 types of circulating metabolites showed significant differences at baseline before IGT progressed to DM,including higher levels of Eggerthella unclassified,Coprobacillus unclassified,Clostridium ramosum,L-valine,L-norleucine,and L-isoleucine,and lower levels of Eubacterium eligens,Bacteroides faecis,Lachnospiraceae bacterium 3_1_46FAA,Alistipes senegalensis,Megaspaera elsdenii,Clostridium perfringens,α-linolenic acid,10E,12Z-octadecadienoic acid,and dodecanoic acid.A random forest model based on differential intestinal microbiota and circulating metabolites can predict the progression from IGT to DM(AUC=0.87).These results suggest that microbiome and metabolome dysbiosis occur in individuals with IGT and have important predictive values and potential for intervention in preventing IGT from progressing to DM.

Irritable bowel syndrome (IBS) is a common chronic gastrointestinal disease with a significant impact on patients’ quality of life and a high socioeconomic burden. And the understanding of IBS has changed since the release of the Rome Ⅳ diagnosis in 2016. With the upcoming Rome Ⅴ revision, it is necessary to review the results of IBS research in recent years. In this review of IBS, we can highlight future concerns by reviewing the results of IBS research on epidemiology, overlap disorders, pathophysiology, and treatment over the past decade and summarizing the latest research.

Distribution of Traditional Chinese Medicine syndromes in diabetic kidney disease chronic kidney disease 1-5: a correlation study

OBJECTIVE: To analyze the distribution of Traditional Chinese medicine(TCM) syndromes in patients with diabetic kidney disease(DKD) and its related ***: We enrolled 435 patients with DKD, who were not undergoing dialysis, admitted to the Department of Nephrology, First Medical Center, Chinese PLA General Hospital from April 2020 to August *** of their TCM syndromes and related factors was carried ***: The 435 patients included 109, 117, 86, and 123 chronic kidney disease(CKD) 1-2, CKD3, CKD4, and CKD5 cases, respectively. With the progression of CKD1-5, the proportion of Yin deficiency and dry heat syndrome,and that of Qi and Yin deficiency syndrome showed a downward trend, whereas the proportion of spleen-kidney Yang deficiency, blood deficiency, blood stasis, water stagnation, and phlegm turbidity syndromes showed an upward trend; the differences were statistically significant(P spleen-kidney Yang deficiency syndrome was negatively correlated with hemoglobin(OR = 0.977, P < 0.001),albumin(OR = 0.891, P < 0.001), and eGFR(OR = 0.978,P < 0.001) but positively correlated with high density lipoprotein(OR = 3.376, P = 0.001). Formation of malignant tumor originating from normal healthy cell is a multistep process including genetic and epigenetic lesions. Previous studies of cell line model systems displayed that early important epigenetic events happened in stepwise fashion prior to cell immortalization. Once these epigenetic alterations are integrated into chromatin, they will perform vertical propagation through cell subculture. Hence, status of epigenetics is dramatically important in maintaining of cell identity. Histone modification is another factor of epigenetic alterations during human oncogenesis. Histones, one of main components of chromatin, can be modified post-translationally. Histone tail modifications are regulated by corresponding modification enzymes. This review focuses on the description of relationship between the main sites of histone modification and oncogenesis.

Bifidobacteriumlongumsubsp. infantis (***. infantis) and Bifidobacteriumadolescentis(B. adolescentis) play important roles in the guts of infants and adolescents， respectively. In this study， using a neonatal rat model， we compared the protective effects of these two bifidobacterial species against Salmonella infection. The results demonstrated that ***. infantis was more effective than B. adolescentis in alleviating the severity of infection in newborn rats exposed to Salmonella entericaserovarTyphimurium strain SL1344. B. *** attenuated intestinal inflammation and mucosal damage induced by Salmonella infection， as well as protecting intestinal nerves and intestinal barrier function through TLR4/MyD88 signalling. B. longumsubsp. infantis also displayed the potential to modulate gut metabolites by promoting the biosynthesis of unsaturated fatty acids (arachidonic acid， eicosapentaenoic acid and α-linolenic acid) and purine metabolism (guanine， adenine， inosine and adenosine)， thereby regulating metabolic disturbances. Additionally， the benefits of B. *** were also observed in the liver， spleen and brain， improving nerve reflexes and suppressing hepatosplenomegaly. Overall， these findings provide novel insights into the prevention and treatment of gut-related diseases in newborns， highlighting the potentially significant role of B. *** in clinical applications.

Insights into the Impact of Bifidobacterium bifidum， B. breve， and B. longum on the Immune Response Against Salmonella Infection in Neonatal Rats

Infants exhibit diminished susceptibility to external infections， a phenomenon closely linked to the pervasive presence of the bifidobacterial community within their intestinal tract. Nonetheless， a significant knowledge gap remains concerning the distinct species of bifidobacteria initiating anti-infection immune responses during the early developmental stages. In this study， the impact of early intervention with Bifidobacterium bifidum (B. bifidum)， B. breve， and B. longum on neonatal rats infected with Salmonella enterica serovar Typhimurium (S. typhimurium) SL1344 were compared. The study encompasses various immunity levels， including gut immunity， central immunity (thymus)， peripheral immunity (spleen)， and brain immunity. Following Salmonella infection， significant alterations in neonatal rats were observed in growth and developmental levels， immune markers， cytokine levels， balance of T lymphocyte subpopulations， intestinal barrier function， and blood-brain barrier integrity. Compared with B. breve and B. longum， B. bifidum demonstrated more pronounced efficacy in regulating these physiological processes. By conducting multi-level analyses of gut microbiota， bifidobacterial community， colonic content metabolomics， and serum metabolomics， the significance of B. bifidum's role is underscored， and the immune-enhancing function of messenger metabolites is unveiled. Among these metabolites， γ-L-Glutamyl-L-glutamic acid and Orotic acid were found to be shared by all three species， while Hippuric acid and 1a，1b-dihomo Prostaglandin F2α were unique to B. bifidum， and DL-Arginine was specific to B. longum. Overall， this study has provided novel insights into the intervention and immunomodulation by Bifidobacterium in early-life infections， emphasizing the significant role of B. bifidum.

Transcriptome analysis reveals immune-related genes in tissues of Vibrio anguillarum-infected turbot Scophthalmus maximus

Turbot Scophthalmus maximus is an important mariculture fish species with high economic ***,the bacterial diseases caused by Vibrio anguillarum infection bring huge economic losses to the turbot aquaculture *** understand the immune response of the turbot against *** infection and to explore novel immune-related genes,the transcriptome analysis of turbot spleen and gills were conducted after *** *** expressed genes(DEGs)were identified in spleen and gill of the turbot amounted to 17261 and 16436,respectively.A large number of immunerelated DEGs were enriched in cytokine-cytokine receptor interaction signaling pathway,and the others by the kyoto encyclopedia of genes and genomes(KEGG)*** gene ontology(GO)classification analysis revealed that *** infection had the greatest effect on biological processes and cellular *** immune-related DEGs were identified in the spleen(cstl.1,egfl6,lamb21,v2rx4,calcr,and gpr78a)and gills(ghra,sh3gl2a,cst12,inhbaa,cxcl8,and il-1b)by heat *** proteinprotein interaction(PPI)networks were constructed to analyze the immune *** results demonstrate that the maturation and antigen processing of major histocompatibility complex(MHC)class II molecule,and calcitonin-or adrenomedullin-regulated physiological activity were important events in the immunity of turbot against *** *** the gills,the protein interactions in TGF-βsignaling pathway,production of inflammatory factors,and endocytosis regulation were most *** research laid a foundation for discovering novel immune-related genes and enriching the knowledge of immune mechanisms of turbot against *** infection.
