Biochemical and microbial soil functioning after application of the insecticide imidacloprid

Imidacloprid is one of the most commonly used insecticides in agricultural practice, and its application poses a potential risk for soil microorganisms. The objective of this study was to assess whether changes in the structure of the soil microbial community after imidacloprid application at the field rate（FR, 1 mg/kg soil） and 10 times the FR（10 × FR, 10 mg/kg soil）may also have an impact on biochemical and microbial soil functioning. The obtained data showed a negative effect by imidacloprid applied at the FR dosage for substrate-induced respiration（SIR）, the number of total bacteria, dehydrogenase（DHA）, both phosphatases（PHOS-H and PHOS-OH）, and urease（URE） at the beginning of the experiment. In 10 × FR treated soil, decreased activity of SIR, DHA, PHOS-OH and PHOS-H was observed over the experimental period. Nitrifying and N2-fixing bacteria were the most sensitive to imidacloprid. The concentration of NO3-decreased in both imidacloprid-treated soils,whereas the concentration of NH4＋in soil with 10 × FR was higher than in the *** of the bacterial growth strategy revealed that imidacloprid affected the r- or K-type bacterial classes as indicated also by the decreased eco-physiological（EP） *** affected the physiological state of culturable bacteria and caused a reduction in the rate of colony formation as well as a prolonged time for growth. Principal component analysis showed that imidacloprid application significantly shifted the measured parameters, and the application of imidacloprid may pose a potential risk to the biochemical and microbial activity of soils.

Membrane fusion is a key step in enveloped virus entry. Highly conserved heptad repeat regions (HR1 and HR2) of Newcastle disease virus (NDV) fusion protein (F) are critical functional domains for viral membrane fusion. They display different conformations in the membrane fusion states and are viewed as candidate targets for neutralizing antibody responses. We previously reported that an analog of heptad repeat peptides HR2-HR1-HR2(HR212) and HR2 could inhibit NDV induced cell-cell membrane fusion. Here, we show that HR212 can induce the production of highly potent antibody in immunized rabbits, which could recognize full length peptides of both HR1 and HR2, and inhibit NDV hemagglutination and NDV entry. These suggest that either HR212 or its antibody could be an inhibitor of virus-induced cell-cell membrane fusion.

Objective: To evaluate NS1 antigen detection ELISA for the early laboratory diagnosis of dengue virus infection. Methods: The present study was conducted to evaluate the overall positivity of NS1 antigen detection ELISA and its comparison with viral RNA detection via real time PCR and Ig M antibodies detection by ELISA. Results: A total of 1 270 serum samples were tested 86%(1 097/1 270) were detected positive by one or more than one diagnostic test. Out of 1 270, 64%(807/1 270) were positive by NS1 ELISA and 52%(662/1 270), 51%(646/1 270) were positive by real-time RT-PCR and Ig M ELISA ***: NS1 antigen detection ELISA is highly suitable diagnostic tools and it also has great value for use in outbreak and epidemic situation.

Born in Dinghai County of Zhejiang province in June, 1926, Guan-Fu Zhu began to study at School of Medicine, Tongji University, in October, 1946 （Fig. 1）. Then he moved to the Department of Bacteriology and Immunology of Peking Union Medical College in September, 1954, where he got his master degree under the supervision of Prof. Shao-Wen Xie, a renowned microbiologist and immunologist. In July, 1951, Mr. Zhu served as an assistant in teaching and research section of the Second Military Medical University of PLA. In January, 1958, he began to work at the institute of Microbi- ology and Epidemiology, Academy of Military Medical Sci- ences, successively serving as a research assistant, assistant professor, associate professor and professor. Due to cardiovascular and cerebrovascular diseases, Mr. Guan- Fu Zhu unfortunately passed away on November 7th, 2015, at the age of 89.

Objective:To High light some epidemiological,clinical and diagnostic features of dengue fever during an outbreak and the role of different diagnostic techniques to achieve the highest level of accuracy in ***:Blood samples(n=323) were collected along with epidemiological and clinical data from suspected dengue patients who visited different hospitals in Swat and Mansehra district of Pakistan between May-November 2013 during a dengue *** were tested for the detection of viral nucleic acid by real-lime *** structural protein-1(NS1antigen and IgM antibodies by ***:Out of 323 cases with clinical dengue infection,304 were positive by one or more diagnostic parameter:201 samples were positive by real-time PCR,209 were positive by NS1 ELISA and 190 were positive by IgM *** of real-time PCR and NS1 *** were comparable for early diagnosis of dengue virus *** antibody detection assay was found useful for the diagnosis in the samples collected later than day 5 of ***:The use of real-lime PCR or detection of non stnictural protein NS 1 by ELISA followed by IgM antibodies detection can be recommended for early diagnosis of dengue virus infection with a high level of accuracy.

Effects of mutated replicase and movement protein genes on attenuation of tobacco mosaic virus

Our previous reports showed that one opal mutation (UGA) and one ochre mutation (UAA) respectively located in the replicase and movement protein (MP) genes of the attenuated tomato mosaic virus K(ToMV-K) contribute to the viral attenuation. To explore a wider application of this attenuation pattern to other plant viruses, we have constructed three mutants which respectively contain one opal mutation of the replicase gene and/or one ochre mutation of the MP using PCR-mediated site-directed mutagenesis from a virulent tobacco mosaic virus isolated from China (TMV-Cv). Plant infection performed by in vitro transcripts revealed that the MP truncated mutant TMV-Cvmp and the replicase-MP truncated mutant TMV-Cvrase-mp were infectious on both local lesion (Nicotiana tabacum cv. Xanthi NC) and systemic (N. tabacum cv. K326) host plants, while the replicase truncated mutant TMV-Cvrase was non-infectious. The K326 plant infected by TMV-Cvrease-mp displayed on|y a little mild mosaic. By electronic microscopy (EM), plant re-inoculation, RNA Dot-blot, RT-PCR and sequencing we demonstrated that the progeny viruses of TMV-Cvmp and TMV-Cvrease-mp shared similar morphological character with TMV-Cv, owned the abilities to infect, replicate and propagate in the assayed plants, and maintained the mutated sites during infection. These data showed that both the opal and the ochre mutations are able to cooperatively induce the attenuated phenotypes of TMV-Cvrase-mp on plants, indicating that the mutation pattern of ToMV-K could be used to attenuate other virulent plant viruses.

China is experiencing the most severe dengue outbreak since the 1980s. Till 31 October 2014, a total of 44497 la- boratory-confirmed dengue cases with six deaths have been reported in China's Mainland, among which 43031 cases were from Guangdong. Although dengue fever is classified as a notifiable infectious disease according to Chinese Ministry of Health, the public and health care system has not fully appreciated the medical burden of this disease. The unprec- edented dengue cases in China have set off the alarm for public health to respond to emerging and re-emerging dis- eases. A comprehensive strategy is needed to win the battle against dengue and other infectious diseases, including sur- veillance, diagnostics, vector control, vaccine, and drug development.
