Mechanism of Shengmai Injection on Anti-Sepsis and Protective Activities of Intestinal Mucosal Barrier in Mice

Objective:To study the mechanism of Shengmai Injection(SMI)on anti-sepsis and protective activities of intestinal mucosal barrier.Methods:The contents of 11 active components of SMI including ginsenoside Rb1,Rb2,Rb3,Rd,Re,Rf,Rg1,Rg2,ophioposide D,schisandrol A and schisantherin A were determined using ultra-performance liquid chromatography.Fifty mice were randomly divided into the blank,the model,the low-,medium-and high-dose SMI groups(0.375,0.75,1.5 mL/kg,respectively)by random number table,10 mice in each group.On SMI group,SMI was administrated to mice daily via tail vein injection for 3 consecutive days,while the mice in the blank and model groups were given 0.1 mL of normal saline.One hour after the last SMI administration,except the blank group,the mice in other groups were intraperitoneally injected with lipopolysaccharide(LPS)saline solution(2 mL/kg)at a dosage of 5 mL/kg for development of endotoxemia mice model.The mice in the blank group were given the same volume of normal saline.Inflammatory factors including interferon-γ(INF-γ),tumor necrosis factor-α(TNF-α),interleukin(IL)-2 and IL-10 were measured by flow cytometry.Myosin light-chain kinase(MLCK),nuclear factorκB(NF-κB)levels,and change of Occludin proteins in jejunum samples were analyzed by Western blot.Results:The decreasing trends of INF-γ,TNF-α and IL-2 were found in serum of SMI treatment groups.In SMI-treated mice,the content of Occludin increased and MLCK protein decreased compared with the model group(P0.05).Conclusion:SMI may exert its anti-sepsis activity mainly through NF-κB-pro-inflammatory factor-MLCK-TJ cascade.

Are herbal sRNAs really novel precision medicines?

Dear Editor,In 2012,Zhang et al.(2012)published an interesting and widely cited article in Cell Research.They reported that exogenous plant miR168a could enter mammals through oral food intake to regulate the expression of mammalian LDLRAP1 gene.From then on,cross-kingdom regulation of diet-derived plant miRNAs has generated substantial interest and attracted media to report.Although there is evidence to support this point of view,the authenticity of cross-kingdom regulation of plant miRNAs has been widely questioned.In 2019,Science China Life Sciences published a special topic entitled“Herbal small RNA,novel precision medicine from Mother Nature”,in which the authors tried to demonstrate an idea that herbal sRNAs are novel precision medicines.This is a thought further than cross-kingdom regulation of sRNAs.However,are herbal sRNAs really novel precision medicines?

Comparing the effects of three decoctions for coronavirus disease 2019 on severe acute respiratory syndrome coronavirus 2-related toll-like receptors-mediated inflammations

OBJECTIVE:To compare the anti-inflammatory effects of three decoctions for coronavirus disease 2019(COVID-19)[Qingfei Paidu Tang(清肺排毒汤),QF;Huashi Baidu Fang(化湿败毒方),HS;Xuanfei Baidu Fang(宣肺败毒方),XF]in parallelly experimental models by using severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)-related toll-like receptors(TLRs)ligands and its spike(S)protein as stimulators.METHODS:RAW264.7 macrophages were used to investigate the effects of three decoctions on the inflammations induced by R848,poly(I:C),lipopolysaccharide(LPS),as well as SARS-CoV-2 S protein,in vitro.Mouse endotoxemia model was used for evaluating their anti-inflammatory actions in vivo.The levels of monocyte chemoattractant protein-1(MCP-1),interleukin-6(IL-6),tumor necrosis factorα(TNF-α),and interferon-β(IFN-β)were determined by enzyme-linked immunosorbent assay.RESULTS:Three decoctions could decrease supernatant IL-6,MCP-1,nitric oxide(NO)and TNF-αto varying degrees in activated macrophages.Meanwhile,they did not increase the level of antiviral cytokine IFN-βinduced by TLR3 and TLR4 ligands,but rather suppressed it,suggesting that externally administrated type I interferons(IFN-Is)may be needed for the severe COVID-19 cases characterized by deficient IFN-Is.In mouse endotoxemia model,all three decoctions could suppress serum pro-inflammatory cytokines,but only QF could relieve hypothermia and antagonize diarrhea.CONCLUSIONS:Collectively,our study compared,for the first time,the effects of three decoctions on SARSCoV-2-related TLRs-mediated inflammations.In vitro,three decoctions exert similar suppressive effects on inflammatory cytokines induced by SARS-CoV-2-related TLRs ligands,as well as S protein.In vivo,QF possesses the strongest effects compared with HS and XF.These findings may provide not only experimental basis for the clinical use of three decoctions,but also a rationale for the combination therapy with IFN-Is.

Trace heavy metals and harmful elements in roots and rhizomes of herbs:Screening level analysis and health risk assessment

Objective:Heavy metal and harmful element contamination are frequently reported in Chinese herbal medicines(CHMs),and roots and rhizomes parts showed a higher content than other parts.To investigate the residue level and assess the potential human health risk of heavy metals and harmful elements in roots and rhizomes,720 batches of the sample representing 20 species of herbs from different sources were collected.Methods:The content of Pb,Cd,As,Hg,and Cu in the digests was determined using ICP-MS.The chronic hazard index estimate based on non-cancer hazard quotient(HQ)was applied for potential health risk assessment of Pb,Cd,As,Hg,and Cu via consumption of CHMs.Results:Compared with the Chinese limit standard(Chinese Pharmacopoeia Commission,2020 edition)of Pb,Cd,As,Hg,and Cu in Ginseng Radix et Rhizoma,the exceedance percentage of Pb in total samples was 14.1%,which were generally far higher than Cd,As,Hg,and Cu.Health risk assessment results based on hazard quotient calculating showed that total HQ of Cu,Pb,As,Cd,and Hg in Pulsatillae Radix and Clematidis Radix et Rhizoma exceeded 1,with the value of 1.543 and 1.235.Besides,Arsenic had the highest HQ value(0.957)in Pulsatillae Radix.Conclusion:Consuming raw materials of Pulsatillae Radix and Clematidis Radix et Rhizoma may pose a potential risk and Arsenic residues in Pulsatillae Radix deserved special attention.

Objective:Dalbergia odorifera has long been used as a Chinese herbal medicine for the treatment of cardiovascular and cerebrovascular diseases.This study aimed to determine the potential myocardial protective effect and possible mechanism of action of D.odor ife ra essential oil(DOEO).Materials and Methods:The essential oil of D.odorifera was extracted by hydrodistillation.The cardioprotective effects of DOEO were examined by histopathological observation,myocardial enzyme detection,peroxidation,anti-oxidant level detection,and related protein expression.The compounds in the blood were identified by gas chromatography-mass spectrometry.Results:These results showed that DOEO had significant myocardial cell protection,with IC50 values ranging from 17.64 to 24.78μg/mL in vitro.Compared to the myocardial ischemia group,the DOEO pretreatment groups had lower levels of myocardial injury,creatinine kinase,lactate dehydrogenase,alanine transaminase,aspartate transaminase,hydrogen peroxide,and nitric oxide,and higher levels of glutathione and superoxide dismutase.In addition,DOEO pretreatment significantly increased Na+-K+-ATPase and Ca2+-ATPase levels.Moreove r,immunohistochemical e xperiments showed that DOEO remarkably increased the protein levels of NF-E2-related nuclear factor 2(Nrf2)and heme oxygenase-1(HO-1)and reduced the expression of apoptotic caspases,including caspase 3 and caspase 9.The main components of the blood were transnerolidol and nerolidol oxide.Overall,the study showed that DOEO displayed myocardial protection by upregulating the NF-E2-related nuclear factor-antioxidant response element(Nrf2-ARE)and caspase pathways.DOEO has a therapeutic effect on MI by inhibiting the oxidant and apoptotic effects.Conclusions:D.odorifera may be a potential candidate drug for treating myocardial ischemic injury.

Sustained release of isoniazid from polylactide microspheres prepared using solid/oil drug loading method for tuberculosis treatment

Polylactide(PLA) microspheres were prepared using the solid-in-oil(S/O) spray-drying method to achieve the sustained release of a hydrophilic drug for the treatment of tuberculosis, via intratracheal instillation. Isoniazid(IN), a low-molecular-weight hydrophilic drug, was used as a model drug. The effects of various sizes of micronized IN powder, different drug/polymer ratios, spray-drying process parameters, and drug-release characteristics were studied to optimize the manufacturing parameters. A high entrapment efficiency(87.3%) was obtained using this method; furthermore, the microspheres were spherical and smooth. They were individually and homogenously distributed, with a mean diameter of 5.6 μm; furthermore, they showed a satisfactory extended sustained-release phase. After administration of the microspheres to rats, pulmonary drug concentrations were maintained at a relatively stable level for up to 4 weeks.

‘Food and medicine continuum’ in the East and West: Old tradition and current regulation

Food-medicine products are important materials for daily health management and are increasingly popular in the global healthy food market.However,because of the biocultural difference,food-medicine knowledge may differ among regions,which hinders the global sharing of such health strategies.Aim at bridging the food-medicine knowledge in the East and West,this study traced the historical roots of food and medicine continuum of the East and West,which was followed by a cross-cultural assessment on the importance of food-medicine products of China,thereafter,the current legislative terms for foodmedicine products were studied using an international survey.The results show that the food and medicine continuum in the East and West have their historical roots in the traditional medicines since antiquity,and the food-medicine knowledge in the East and West differs substantially;although the foodmedicine products have common properties,their legislative terms are diverse globally;with proofs of traditional uses and scientific evidence,food-medicine products are possible for cross-cultural communication.Finally,we recommend facilitating the cross-cultural communication of the food-medicine knowledge in the East and West,thus to make the best use of the traditional health wisdom in the globe.

Cloning and expression of SgCYP450-4from Siraitia grosvenorii

CYP450 plays an essential role in the development and growth of the fruits of Siraitia grosvenorii. However, little is known about the SgCYP450-4 gene in S. grosvenorii. Here, based on transcriptome data, a full-length c DNA sequence of SgCYP450-4 was cloned by reverse transcriptasepolymerase chain reaction(RT-PCR) and rapid-amplification of c DNA ends(RACE) strategies. SgCYP450-4 is 1677 bp in length(Gen Bank accession No. AEM42985.1) and contains a complete open reading frame(ORF) of 1422 bp. The deduced protein was composed of 473 amino acids, the molecular weight is 54.01 kDa, the theoretical isoelectric point(PI) is 8.8, and the protein was predicted to possess cytochrome P450 domains. SgCYP450-4 gene was highly expressed in root, diploid fruit and fruit treated with hormone and pollination. At 10 days after treatment with pollination and hormones, the expression of SgCYP450-4had the highest level and then decreased over time, which was consistent with the development of fruits of S.Grosvenorii. Hormonal treatment could significantly induce the expression of SgCYP450-4. These results provide a reference for regulation of fruit development and the use of parthenocarpy to generate seedless fruit, and provide a scientific basis for the production of growth regulator application agents.

Mining Therapeutic Efficacy from Treasure Chest of Biodiversity and Chemodiversity:Pharmacophylogeny of Ranunculales medicinal plants

Ranunculales,comprising of 7families that are rich in medicinal species frequently utilized by traditional medicine and ethnomedicine,represents a treasure chest of biodiversity and chemodiversity.The phylogenetically related species often have similar chemical profile,which makes them often possess similar therapeutic spectrum.Thishas been validatedby both ethnomedicinal experiences and pharmacological investigations.This paper summarizes molecular phylogeny,chemical constituents,and therapeutic applications of Ranunculales,i.e.,a pharmacophylogeny study of this representative medicinal order.The phytochemistryl metabolome,ethnomedicine and bioactivity/pharmacology data are incorporated within the phylogenetic framework of Ranunculales.The most studied compounds of this order include benzylisoquinoline alkaloid,flavonoid,terpenoid,saponin and lignan,etc.Bisbenzylisoquinolinealkaloids are especially abundant in Berberidaceae and Menispermaceae.The most frequent ethnomedicinal uses are arthritis,heat-clearing and detoxification,carbuncle-abscess and sore-toxin.The most studied bioactivities are anticancer/cytotoxic,antimicrobial,and anti-inflammatory activities,etc.The pharmacophylogeny analysis,integrated with both traditional and modern medicinal uses,agrees with the molecular phylogeny based on chloroplast and nuclear DNA sequences,in which Ranunculales is divided into Ranunculaceae,Berberidaceae,Menispermaceae,Lardizabalaceae,Circaeasteraceae,Papaveraceae,and Eupteleaceaefamilies.Chemical constituents and therapeutic efficacy of each taxonomic group are reviewed and the underlying connection between phylogeny,chemodiversityand clinical uses is revealed,which facilitate the conservation and sustainable utilization of Ranunculales pharmaceutical resources,as well as developing novel plant-based pharmacotherapy.

Bornyl acetate extracted from Sharen(Fructus Amomi)inhibits proliferation,invasion and induces apoptosis by suppressing phosphatidylinositol-3-kinase/protein kinase B signaling in colorectal cancer

OBJECTIVE:To investigate the antitumor effects of bornyl acetate(BA)isolated from Sharen(Fructus Amomi)in colorectal cancer(CRC)and the underlying mechanisms.METHODS:SW480 and HT29 cells were treated with increasing doses of BA in order to determine its antitumor effects in vitro.Cell viability,colony formation,cell cycle,and apoptosis as well as migration and invasion were assessed using various assays.In addition,the in vivo antitumor effects of BA were assessed using a xenograft mouse model.We then assessed the mechanism of action of BA by conducting pathway activator-mediated rescue experiments and assessed the protein levels by Western blot analysis.RESULTS:BA showed anti-CRC tumor activities in vitro by suppressing cell proliferation and colony formation,inducing apoptosis,blocking cell cycle,and inhibiting migration and invasion.These effects were mediated via suppression of the phosphatidylinositol-3-kinase/protein kinase B(PI3K/AKT)pathway.In the tumor xenograft experiment,BA was found to repress tumor growth in vivo with low toxicity.CONCLUSIONS:The results demonstrated that BA exerts antitumor effects by suppressing the PI3K/AKT pathway,with low toxicity.Thus,BA might be a potential novel therapeutic agent for CRC.
