The role of antibodies in kidney transplant(KT)has evolved significantly over the past few *** role of antibodies in KT is multifaceted,encompassing both the challenges they pose in terms of antibody-mediated rejection(AMR)and the opportunities for improving transplant outcomes through better detection,prevention,and treatment *** our understanding of the immunological mechanisms continues to evolve,so too will the approaches to managing and harnessing the power of antibodies in KT,ultimately leading to improved patient and graft *** narrative review explores the multifaceted roles of antibodies in KT,including their involvement in rejection mechanisms,advancements in desensitization protocols,AMR treatments,and their potential role in monitoring and improving graft survival.

Currently,display-based methods are well established and widely used in antibody engineering for affinity maturation and structural stability *** obtained a novel anti-human programmed death 1(PD-1)antibody using computer-aided design and a mammalian cell display technology *** used computer-aided modeling and distance geometry methods to predict and assign the key residues that contributed to the binding of human PD-L1 to ***,we analyzed the sequence of nivolumab(an anti-human PD-1 antibody,referred to as MIL75 in the article)to determine the template for antibody design and library *** identified a series of potential substitutions on the obtained template and constructed a virtual epitope-targeted antibody library based on the physicochemical properties and each possible location of the assigned key *** virtual antibody libraries were displayed on the surface of mammalian cells as the antigen-binding fragments of full-length immunoglobulin ***,we used flow cytometry and sequencing approaches to sort and screen the ***,we obtained a novel anti-human PD-1 antibody named ***78 competitively recognized the PD-1 epitopes that interacted with MIL75 and possessed an affinity comparable to *** results implied that FV78 possessed equivalent bioactivity in vitro and in vivo compared with MIL75,which highlighted the probability and prospect of FV78 becoming a new potential antibody therapy.

Inflammatory bowel disease(IBD)is caused by an uncontrolled immune response in the intestinal lumen,leading to inflammation in genetically predisposed *** may be a promising approach to the treatment of ***,we show that transforming growth factor-β1(TGF-β1)gene-modified immature dendritic cells(imDCs)could enhance the inhibitory function of imDCs and delay the progress of IBD induced by dextran sodium sulfate in *** results of fluorescence-activated cell sorter(FACS)demonstrated that this protective effect is mediated partially by inducing CD4^(+)Foxp3^(+)regulatory T cells(Tregs)in mesentery lymph nodes to control *** vitro experiments also supported this *** conclusion,we provide evidence that TGF-b1-modified bone marrow-derived imDCs may have a therapeutic effect to IBD.

Innate recognition of DNA within intracellular compartments or thecytoplasm leads to activation of the immune system, which can result in defense against DNA virus infections or the initiation of pathological responses to self-DNA. In the past several years, the field of DNA sensing has undergone an immense expansion. Many DNA sen- sors have been proposed, including cGAMP synthase （cGAS）, DNA-depen- dent activator of interferon （IFN）-regula- tory factors （also named Z-DNA binding protein 1）, absent in melanoma 2 （MM2）, leucine-rich repeat （in Flightless I） inter- acting protein-1, DExD/H box helicases （DHX9 and DHX36） and IFN-inducible protein IFI16^1-3 （Figure 1）.

Background DNA analysis has shown a lack of significant compatibility between couples affected by unexplained recurrent spontaneous abortion (URSA) compared with normal fertile couples, 8 although one study that made use of a PCR-sequence-specific oligonucleotide (SSO) method did observe evidence of significant compatibility in the HLA-DQA1 and DQB1 alleles between patients and aborted fetuses. 9 This study was designed to investigate whether URSA were associated with particular DQ alleles or promoter *** Thirty-two patients with URSA and 54 women who had had at least one successful pregnancy were included in this study. HLA-DQ genotyping was performed by the polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) method. The HLA-DQB1 promoter was detected by the SSO and sequence-specific primer (SSP) methods. The DQA1, DQB1, and DQB1 promoter (QBP) gene frequencies in the patients were compared with the gene frequencies in normal controls. The data were analyzed statistically with the χ 2 and Fisher’s exact *** The results showed that the frequency of DQB1 *0604/0605 was significantly higher and the frequency of DQB1 *0501/0502 was significantly lower in the patient group as compared with the normal controls. In addition, the frequencies of the DQA1 *01-DQB1 *0604/0605 and QBP6.2-DQB1 *0604/0605 haplotypes were overrepresented in the patients relative to the controls. Our results did not show any differences between URSA patients and the controls with regard to DQA1 and QBP allele frequencies. Conclusions Our data suggest that URSA is associated with the HLA-DQB1 coding region, and is not associated with its upstream regulatory region. The DQB1 *0604/0605, DQA1 *01-DQB1 *0604/0605, and QBP6.2-DQB1 *0604/0605 haplotypes may confer susceptibility to URSA, while the DQB1 *0501/0502 allele may protect women from URSA.

An infection by Zika virus(ZIKV), a mosquito-borne flavivirus, broke out in South American regions in 2015, and recently showed a tendency of spreading to North America and even worldwide. ZIKV was first detected in 1947 and only 14 human infection cases were reported until 2007. This virus was previously observed to cause only mild flu-like ***, recent ZIKV infections might be responsible for the increasing cases of neurological disorders such as GuillainBarre′ syndrome and congenital defects, including newborn microcephaly. Therefore, researchers have established several animal models to study ZIKV transmission and pathogenesis, and test therapeutic candidates. This review mainly summarizes the reported animal models of ZIKV infection, including mice and non-human primates.

Relationship between Microsatellite Alterations of RASSF1A Gene and Development of Cervical Carcinoma

Objective： To explore the relationship between microsatellite alterations of RASSFIA gene and the development of cervical carcinoma, and its relationship with HPV16 infection. Methods： Two sites of microsatellite polymorphism of RASSFIA gene were selected. Polymerase chain reaction （PCR） technique was used to detect LOH and MSI in 50 cases of cervical carcinoma and 40 cases of cervical intraepithelial neoplasia （CIN）, and to detect the infection state of HPV16. Results： At D3S1478 and D3S4604, the LOH rates of cervical carcinomas were 32.6% （14/43） and 48.9% （23/47）, the MSI rates were 14% （6/43） and 19.1% （9/47）, respectively. The LOH rates of CINs were 31.4% （11/35） and 39.5% （15/38）, the MSI rates were 11.4% （4/35） and 15.8% （6/38）, respectively. There were no significant differences between cervical carcinomas and CINs in respect to their positive rates of LOH and MSI at D3S1478 and D3S4604 （P〉0.05）. There were significant differences in LOH rates at D3S1478 and D3S4604 between the stage Ⅰ-Ⅱ and Ⅲ-Ⅳ cervical carcinomas and between the well/moderately differentiated cervical carcinomas and the poorly differentiated cervical carcinomas （P〈0.05）. The positive rates of LOH and MSI for CIN Ⅲ and noninvasive cervical carcinomas were higher than those in CIN Ⅰ-Ⅱ. The rates of infection of HPV16 in cervical cancer was obviously higher than that in CIN and in normal cervical tissues （P〈0.05）, and the incidence of LOH of RASSFIA gene was higher in HPV16（＋） than that in HPV16（-）（P〈0.05）. Conclusion： The RASSFIA gene change is a relatively late event in cervical carcinomas. The detection of LOH and MSI of RASSFIA gene might be helpful to the early diagnosis and the screening of cervical carcinoma. It might also be useful for predicting the prognosis of cervical carcinoma.

Qinghaosu （青蒿素）, isolated and purified from the Chinese herb Artemisia annua Linn（黄花蒿）, and identified as a sesquiterpene with a peroxide bridge and lactone structure,is a highly potent and non-toxic new antimalaria drug. This paper reports the immunosup-pressive action of its water soluble derivative （hemisuccinate Na, QHS）. The remarkable sup-pression by QHS of the in vitroHTdR incorporation by mitogen-stimulated mouse spleencells and human peripheral lymphocytes, as well as the spontaneous incorporation by mousethymocytes and blood cells from some leukemia patients is presented and its characteristics aredescribed. The in vivo effect as shown by quantitative PFC is studied and the differencebetween the present in vitro and in vivo effects is investigated. The possible mechanism ofinhibition and discrepancy in effects are discussed.
