A strategy for uncovering germline variants altering anti-tumor CD8 T cell response

Among many factors known to alter the outcomes of T cell receptor(TCR)-induced proximal signaling,the role of human germline variants in dictating the individuality of the anti-tumor CD8 T cell response has remained challenging to ***,we describe a convenient strategy for molecular and functional characterization of phosphotyrosine-altering non-synonymous single nucleotide variations(pTyr-SNVs)that directly impact TCR-induced proximal phosphotyrosine motif-based signaling *** devise an experimental co-cultivation set-up comprising a C57BL/6 mouse-derived metastatic melanoma cell line engineered to constitutively present ovalbumin(OVA)antigens and retrovirally engineered syngeneic major histocompatibility complex(MHC)Class I restricted OVA TCR-transgenic CD8 T cells(OT-I).Using the synthetic version of pTyr-SNV rs1178800678-G/T-encoding integrin alpha 4(ITGA4)p.S1027I variant as a prototype,we show that under identical TCR stimulation conditions,genetically determined membrane-proximal immunoreceptor tyrosin activation motif(ITAM)results in increased tyrosine phosphorylation of 70 kDa zeta-chain-associated protein(ZAP70)and the levels of cytotoxic effector molecule granzyme B(GZMB),which in turn result in enhanced cytotoxic activity against metastatic melanoma cell *** strategy paves the way for rapid molecular and functional characterization of anti-tumor immune response-linked germline pTyr-SNVs so as to improve our understanding of the genetic basis of individual-to-individual differences in anti-tumor CD8 T cell response.

On-Station and On-Farm Assessment of the Effects of Soil Cover on Conservation Agriculture Performances in Western Burkina Faso

Conservation agriculture (CA) is one option for dealing with such a challenge, but its main difficulty in West Africa is in maintaining permanent soil cover, particularly with crop residues, due to their preferential use for livestock fodder. The aim of our study was to determine the effect of crop residue mulching on the efficiency of cropping systems based on the principles of conservation agriculture. The experimental design was based on on-station experiments, intended to assess the specific effect of different levels of crop residue mulching on the biological and chemical soil fertility parameters, while in on-farm experiments we mainly assessed the ability of farmers to actually collect crop residues for soil cover and the associated effects on weeds and yields. The on-station experimental design was in fully randomized factorial blocks comprising five treatments and three replicates. The treatments compared the conventional system, tillage and cropping without mulching, and CA systems with 1 ton, 2 tons, and 4 tons of straw per hectare in plots of 10 m2. The effects of CA on the macrofauna, respiratory activity, and soil chemical parameters were evaluated in the 2014 and 2015 growing seasons. For the on-farm experiments, the conventional and CA practices of 15 farmers were compared to conventional practices in 2013 and 2014 in plots of 626 m2. The on-station results showed that the presence of crop residue mulching induced an increase in the density of termites. A significant release of carbon dioxide (CO2) in the CA treatments compared to the conventional treatment was also observed. For the chemical parameters (pH, N, P and K) were significantly and positively affected by mulch in the top 5 centimeters of soil. The on-farm experiments confirmed the difficulty of farmers in collecting enough biomass, with negative effects on grass cover leading to generally lower yields than conventional treatments. Quality Assessment of Transcriptome Data Using Intrinsic Statistical Properties

In view of potential application to biomedical diagnosis, tight transcriptome data quality control is compulsory. Usually, quality control is achieved using labeling and hybridization controls added at different stages throughout the processing of the biologic RNA samples. These control measures, however, only reflect the performance of the individual technical manipulations during the entire process and have no bearing as to the continued integrity of the RNA sample itself. Here we demonstrate that intrinsic statistical properties of the resulting transcriptome data signal and signal-variance distributions and their invariance can be identified independently of the animal species studied and the labeling protocol used. From these invariant properties we have developed a data model, the pa- rameters of which can be estimatei：l from individual experiments and used to compute relative quality measures based on similarity with large reference datasets. These quality measures add supplementary, non-redundant in- formation to standard quality control estimates based on spike-in and hybridization controls, and are exploitable in data analysis. A software application for analyzing datasets as well as a reference dataset for AB 1700 arrays are provided. They should allow AB 1700 users to easily integrate this method into their analysis pipeline, and might instigate similar developments for other transcriptome platforms.

In vitro antiplasmodial activity of crude extracts from Togolese medicinal plants

Objective:To investigate the antimalarial effect of a few plants in Togo folk medicine. Methods:After ethnobotanical survey,Opilia celtidifolia,Pavetta corymbosa（***） and Tamarindus indica（***） were selected for *** vitro antimalarial tests were performed on crude extracts against fresh clinical isolates of Plasmodium falciparum using the semi ***:Different IC50,values of the extracts ranged from 2.042 to 100.000μg/mL. According to the results,the methanol extract of aerial part of *** followed by aqueous extract of fruit of *** were the most active(IC50 of 2.042 and 4.786μg/mL,respectively). Qualitative test revealed the presence of alkaloids in the leaves of *** that may be responsible for the activity of the ***:Our study provides scientific evidence for usage of plant in the folk medicine,and further studies are needed for identification and purification of the active principles.

植物需要多种基本元素调节并保持其特定比例以维持生长发育,叶片元素含量反映了植物的功能及其对环境的适应性.然而,在森林海拔梯度上,我们对于资源获取、光合作用和生长相关的叶片元素如何调节森林生物量仍知之甚少.本研究在喜马拉雅山干城章嘉峰地区沿热带森林(海拔80m)至高山树线(海拔4200m)设置海拔梯度样带,每间隔海拔100m设置一个样点,对116个树种的1859个个体测定了叶片中10种元素(C、N、P、K、Ca、Mg、Zn、Fe、Cu和Mn)的含量,通过计算叶片元素含量的群落加权平均值(反映优势物种的性状优势度,表征质量比效应)和功能离散度(反映物种在生态位空间内的聚集或者离散程度,表征互补效应),探究叶片元素含量及其离散程度沿海拔梯度分异特征,解析其对森林群落生物量的影响.研究结果表明,较强的叶片元素质量比效应和较弱的互补效应有利于森林群落生物量的积累.叶片元素性状多样性与海拔的共同作用对生物量的解释量(解释方差:52.2%)高于元素性状多样性的解释量(解释方差:0.05%~21%).海拔调节了树种性状多样性在生物量积累中的作用,互补效应在低海拔和高海拔地区对生物量的影响分别体现为积极和消极作用,这表明海拔和互补效应之间也存在交互作用.此外,海拔和质量比效应的交互作用对海拔梯度上生物量的影响也存在异质性.本研究表明,不同森林群落生物量的积累可能受到由海拔调控的性状多样性的影响.尽管较高的性状分异不利于生物量的积累,但能增强高海拔地区树种对胁迫环境的适应性.

Forecasting the development of boreal paludified forests in response to climate change: a case study using Ontario ecosite classification
