27-Hydroxycholesterol-induced EndMT acts via STAT3 signaling to promote breast cancer cell migration by altering the tumor microenvironment

Objective:The endothelial to mesenchymal transition(EndMT)plays a major role in cancer metastasis by regulating the complexity of the tumor microenvironment(TME).Here,we investigated whether 27-hydroxycholesterol(27 HC)induces EndMT in endothelial cells(ECs).Methods:EndMT markers in the human microvascular endothelial cell-1(HMEC-1)cell line and human umbilical vein endothelial cells(HUVECs)stimulated with 27 HC were evaluated with Western *** to mesenchymal transition(EMT)markers in breast cancer(BC)cells cultured in conditioned medium were investigated with quantitative real time polymerase chain reaction(qRT-PCR).The MMP-2 and MMP-9 mRNA expression and activity were detected with qRT-PCR and gelatin zymography assays,*** effect of activated STAT3 on 27 HC-induced EndMT was validated by Western blot,immunofluorescence staining,and cell transfection *** migration ability of BC cells was evaluated with Transwell ***:We found that 27 HC induced EndMT in HMEC-1 and HUVECs,and 27 HC-induced EndMT facilitated EMT and BC cell *** 27 HC-induced EMT of BC cells also promoted EndMT and HUVEC *** of the underlying molecular mechanisms revealed that STAT3 knockdown repressed EndMT in HUVECs as well as migration in BC cells induced with 27 *** addition,C646 and resveratrol,inhibitors of STAT3 acetylation,repressed the expression of Ac-STAT3,p-STAT3,and EndMT markers in HUVECs exposed to 27 HC;these HUVECs in turn attenuated the migration ability of BC cells in 27 HC-induced ***:Cross-talk between 27 HC-induced EndMT and EMT was observed in the ***,activation of STAT3 signaling was found to be involved in 27 HC-induced EndMT.

AIM： To investigate the prevalence and clinical significance of ＂anti-HBc alone＂ in an unselected population of patients and employees of a university hospital in southern Germany. METHODS： All individuals with the pattern ＂anti-HBc alone＂ were registered over a time span of 82 mo. HBVDNA was measured in serum and liver samples, and clinical charts were reviewed. RESULTS： Five hundred and fifty two individuals were ＂anti-HBc alone＂（of 3004 anti-HBc positive individuals; 18.4%）, and this pattern affected males （20.5%） more often than females （15.3%; P〈0.001）. HBV-DNA was detected in serum of 44 of 545 ＂anti-HBc alone＂ individuals （8.1%）, and in paraffin embedded liver tissue in 16 of 39 patients tested （41.0%）. There was no association between the detection of HBV genomes and the presence of biochemical, ultrasonic or histological signs of liver damage. Thirty-eight ＂anti-HBc alone＂ patients with cirrhosis or primary liver carcinoma had at least one additional risk factor. HCV-coinfection was present in 20.4% of all individuals with ＂anti-HBc alone＂ and was the only factor associated with a worse clinical outcome. CONCLUSION： In an HBV low prevalence area, no evidence is found that HBV alone causes severe liver damage in individuals with ＂anti-HBc alone＂. Recommendations for the management of these individuals are given.

The purpose of this study was to investigate the potential of high-solids anaerobic mono-digestion of riverbank grass under thermophilic conditions, focusing on the effects of the strength and the amount of inoculum. Ensiled grass was inoculated with three different inocula; inoculum from liquid anaerobic digester（LI）, inoculum from dry anaerobic digester（DI）, and mixture of LI and DI（MI）, at feedstock-to-inoculum ratio（FIR） of 1, 2 and 4. The ensiling process of riverbank grass reduced moisture content（p 〉 0.05）, while the hemicellulose content was significantly increased from 30.88% to 35.15%（p 〈 0.05）, on dry matter basis. The highest methane production was at an FIR of 2 with MI（167 L/kg VSadded）,which was significantly higher（p 〈 0.05） than with DI, but not significant compared to LI（p 〉 0.05）. At an FIR of 4, digesters inoculated with LI and DI failed to produce methane,whereas 135 LCH4/kg VSaddedwas obtained with MI. The kinetic studies showed that at an FIR of 1 with LI and MI, the inoculum had less of effects on the hydrolysis rate constant（0.269 day-1and 0.245 day-1） and methane production（135 versus 149 L/kg VSadded）; rather,it affected the lag phase. In a thermophilic HS-AD of riverbank grass, the mixture of inoculum with low and high total solids content（TS） helps increase the TS of inoculum and digestion process. An FIR of 2 was deducted to be the limit for a better startup time and higher volumetric productivity of methane.

Study on Relationship between Carbon Disulfide Exposure and Symptoms and Signs of Nervous System in Workers

The relationship between carbon disulfide (CS2) and the symptoms and signs of nervous system in rayon workers was investigated. In a cross sectional study of CS2-exposed workers from the viscose industry and controls, data on neurological symptoms and clinical neurological examination were evaluated. Theexposed subjects includes 326 workers and 105 workers from the same factory,without exposure to CS2 or to any other neurotoxic agents, served as *** median exposure to CS2(8 h time weighted) in the low-level exposure group was 4. 6 mg/m3, and 18. 3 mg/m3 in high-level exposure group. Non-conditional multiple logistic regression analysis was employed for evaluating statistical differ-ences by using SAS software. In the analysis, age, sex, work time, body massindex (BMI), smoking and drinking habits, educational background and marriage status were controlled. The odds ratio (OR) significantly increased of some symptoms in workers exposed to CS2 at levels below 10 mg/m3, as compared to con-trols. The dose-effect relationship between the exposure to CS2 and the symptoms of nervous system was demonstrated. In terms of neurological parameters, no other abnorrnal change but that in temperature sensitivity was observed in work-ers exposed to CS2. In conclusion, the influence of low level CS2 on nervous system of workers should not be ignored.

BACKGROUND： Folic acid is essential for normal functioning of the nervous system. Previous studies have focused on the effects of folic acid on astrocyte proliferation. OBJECTIVE： To explore the effects of folic acid on astrocyte differentiation of neural stem cells （NSCs） and the related mechanisms in vitro. DESIGN, TIME AND SETTING： A randomized, controlled, grouping experiment was performed in Tianjin Medical University between August 2007 and October 2008. MATERIALS： Folic acid and 5-bromo-2-deoxyuridine （BrdU） were obtained from Sigma, MO, USA. Primary antibodies [rabbit anti-rat nestin, β-tubulin-Ⅲ, glial fibrillary acidic protein, and neurogeninl （Ngnl）; mouse anti-rat BrdU and β-actin monoclonal antibodies] were purchased from Santa Cruz Biotechnology, USA. METHODS： At 6 days of NSC proliferation from 24-hour-old neonatal rats, BrdU incorporation assay was performed. Seven days after primary culture, NSCs were induced to differentiate with medium containing 5% fetal bovine serum. Cultured NSCs were assigned to three groups： control, low-dose （liquid media with 8 mg/L folic acid）, and high-dose folic acid （liquid media with 44 mg/L folic acid）. MAIN OUTCOME MEASURES： At day 7 after primary culture, the cells were identified as NSCs by immunocytochemical methods. Double-label immunofluorescence technique for glial fibrillary acidic protein/BrdU detected differentiated cells 7 days after induction. Western blot was used to analyze expression of Ngnl protein in NSCs. RESULTS： In serum-free suspension medium, neurospheres comprised a large number of Nestin-, glial fibrillary acidic protein-, β-tubulin-Ⅲ-, and BrdU-positive cells. Compared with the control group, high-dose folic acid supplementation led to an marked increase in the percentage of glial fibrillary acidic protein/BrdU-positive cells （P 〈 0.05）, and significantly decreased Ngnl protein expression （P 〈 0.05）. Objective To study the effect of chronic noise exposure on expression of N-methyI-D-aspartic acid receptor 2B （NR2B） and tau phosphorylation in hippocampus of rats. Methods Twenty-four male SD rats were divided in control group and chronic noise exposure group. NR2B expression and tau phosphorylation in hippocampus of rats were detected after chronic noise exposure （100 dB SPL white noise, 4 h/dx30d） and their mechanisms underlying neuronal apoptosis in hippocampus of rats with TUNEL staining. Results The NR2B expression decreased significantly after chronic noise exposure which resulted in tau hyperphosphorylation and neural apoptosis in hippocampus of rats. Immunohistochemistry showed that the tau hyperphosphorylation was most prominent in dentate gyrus （DG） and CA1 region of rat hippocampus. Conclusion The abnormality of neurotransmitter system, especially Glu and NR2B containing NMDA receptor, and tau hyperphosphorylation in hippocampus of rats, may play a role in chronic noise-induced neural apoptosis and cognition impairment.

Phenolic acids (PAs) are widely found in many daily consumed vegetables and *** inhibitory effects of PAs on Nnitrosomorpholine (NMOR) formation in vitro undersimulated gastric juice condition were studied. The results showed that the inhibjtoryIX)tency of thirteen varieties of PAs differed greatly, which may be related to theirchemical structures; the blocking rate (BR) of different killds of PAs were as follows:caf feic acid, 92.5%; tannic acid, 90.0%; gallic acid, 86.8%; sinaPinic acid, 86.2%; ferulicacid, 8l.l%; chlorogenic acid, 69.4%; gentisic acid, 69.2%; syringic acid, 62.1%; protocatechuic acid, 56.0%; p-coumaric acid, 52.5%; vannilic acid, 35.4%; moreover,p-hydroxybenzoic acid and m-coumaric acid had the least blocking effect or even slightcatalyzing effect. The results also demonstrated that amounts of NMOR formed werenegatively correlated with molar ratio of PAs to nitrite and that the optimum pH forinhibition was between 2 and 3.
