Background:There exist few maximal oxygen uptake(VO_(2max))non-exercise-based prediction equations,fewer using machine learning(ML),and none specifically for older *** direct measurement of VO_(2max)is infeasible in large epidemiologic cohort studies,we sought to develop,validate,compare,and assess the transportability of several ML VO_(2max)prediction ***:The Baltimore Longitudinal Study of Aging(BLSA)participants with valid VO2_(max)tests were included(n=1080).Least absolute shrinkage and selection operator,linear-and tree-boosted extreme gradient boosting,random forest,and support vector machine(SVM)algorithms were trained to predict VO_(2max)*** developed these algorithms for:(a)the overall BLSA,(b)by sex,(c)using all BLSA variables,and(d)variables common in aging ***,we quantified the associations between measured and predicted VO_(2max)and ***:The age was 69.0±10.4 years(mean±SD)and the measured VO_(2max)was 21.6±5.9 mL/kg/*** absolute shrinkage and selection operator,linear-and tree-boosted extreme gradient boosting,random forest,and support vector machine yielded root mean squared errors of 3.4 mL/kg/min,3.6 mL/kg/min,3.4 mL/kg/min,3.6 mL/kg/min,and 3.5 mL/kg/min,*** quartiles of measured VO_(2max)showed an inverse gradient in mortality *** VO_(2max)variables yielded similar effect estimates but were not robust to ***:Measured VO_(2max)is a strong predictor of *** ML can improve the accuracy of prediction as compared to simpler approaches but estimates of association with mortality remain sensitive to *** studies should seek to reproduce these results so that VO_(2max),an important vital sign,can be more broadly studied as a modifiable target for promoting functional resiliency and healthy aging.

Pregnancy stressors and postpartum symptoms of depression and anxiety:the moderating role of a cognitivebehavioural therapy(CBT)intervention

Background Little is known about the association between stressors(especially positive stressors)during pregnancy and postpartum depression and *** We investigated the association between positive and negative stress events during different stages of pregnancy and postpartum mental health outcomes among low-income pregnant women with symptoms of anxiety in Pakistan and evaluated whether an intervention based on cognitive behavioural therapy(CBT)had a regulatory *** Participants were 621 pregnant Pakistani women with mild *** the Pregnancy Experience Scale-Brief Version,six scores were created to assess positive and negative *** performed a multivariate linear regression to examine whether these six scores,measured both at baseline and in the third trimester,were associated with postpartum anxiety and depressive *** effect of the intervention on this relationship was examined by adding an interaction term to the regression *** Hassles frequency measured in the third trimester was positively associated with depression(B=0.22,95%confidence interval(Cl):0.09 to 0.36)and anxiety(B=0.19,95%Cl:0.08to 0.30).At the same timepoint,uplifts intensity was negatively associated with symptoms of depression(B=-0.82,95%Cl:1.46 to-0.18)and anxiety(B=-0.70,95%Cl:-1.25 to-0.15),whereas hassles intensity was positively related to symptoms of depression(B=1.02,95%Cl:0.36 to 1.67)and anxiety(B=0.90,95%Cl:0.34 to 1.47).The intensity ratio of hassles to uplifts reported in the third trimester was positively related to both depression(B=1.40,95%Cl:0.59 to 2.20)and anxiety(B=1.26,95%Cl:0.57 to 1.96).The intervention strengthened the overall positive effects of uplfts and the negative effects of *** experiences at baseline during early pregnancy to mid-pregnancy were not associated with mental health *** Stressors in the third trimester but not earlier in pregnancy were associated with postpartum symptoms

Cognitive impairment associated with individual and joint exposure to PM_(2.5)constituents in a Chinese national cohort

Nationwide longitudinal evidence linking cognitive decline with exposure to fine particulate matter(PM_(2.5))constituents remains scarce in *** constructing a dynamic cohort based on the China health and Retirement Longitudinal Study,we aimed to assess individual and joint associations of PM_(2.5)constituents with cognitive function among middle-aged and older adults in *** mixed-effects models incorporated with quantile-based g-computation were applied to investigate individual and joint associations of long-term exposures to PM_(2.5)constituents with cognitive *** 13,507 respondents,we evaluated 38,950 follow-up records of cognitive function *** in global cognitive score associated with an interquartile range(IQR)increase in exposure were−1.477(95%CI:−1.722,−1.232)for nitrate,followed by−1.331(−1.529,−1.133)for ammonium,−1.033(−1.184,−0.883)for sulfate,−0.988(−1.144,−0.832)for organic matter and−0.822(−0.946,−0.699)for black *** IQR-equivalent increase in joint exposure to these PM_(2.5)constituents was associated with a decline of−1.353(−1.659,−1.048)in global cognitive ***,younger,and well-educated individuals were at greater vulnerability to cognitive impairment related to individual and joint exposure to PM_(2.5)*** study suggested that later-life exposures to PM_(2.5)constituents were associated with cognitive decline in middle-aged and older adults in China.

AIM:To develop and evaluate the validity and reliability of a knowledge,attitude,and practice questionnaire related to vision screening(KAP-VST)among preschool teachers in ***:The questionnaire was developed through a literature review and discussions with *** and face validation were conducted by a panel of experts(n=10)and preschool teachers(n=10),respectively.A pilot study was conducted for construct validation(n=161)and test-retest reliability(n=60)of the newly developed ***:Based on the content and face validation,71 items were generated,and 68 items were selected after exploratory factor *** content validity index for items(I-CVI)score ranged from 0.8-1.0,and the content validity index for scale(S-CVI)/Ave was *** consistency was KR^(2)0=0.93 for knowledge,Cronbach’s alpha=0.758 for attitude,and Cronbach’s alpha=0.856 for ***:The KAP-VST is a valid and reliable instrument for assessing knowledge,attitude,and practice in relation to vision screening among preschool teachers in Malaysia.

Fibrous dysplasia(FD)is a rare,disabling skeletal disease for which there are no established *** evidence supports inhibiting the osteoclastogenic factor receptor activator of nuclear kappa-B ligand(RANKL)as a potential treatment *** this study,we investigated the mechanisms underlying RANKL inhibition in FD tissue and its likely indirect effects on osteoprogenitors by evaluating human FD tissue pre-and post-treatment in a phase 2 clinical trial of denosumab(NCT03571191)and in murine in vivo and ex vivo preclinical *** analysis of human and mouse tissue demonstrated increased osteogenic maturation,reduced cellularity,and reduced expression of the pathogenic Gαs variant in FD lesions after RANKL *** sequencing of human and mouse tissue supported these *** interaction between osteoclasts and mutant osteoprogenitors was further assessed in an ex vivo lesion model,which indicated that the proliferation of abnormal FD osteoprogenitors was dependent on *** results from this study demonstrated that,in addition to its expected antiosteoclastic effect,denosumab reduces FD lesion activity by decreasing FD cell proliferation and increasing osteogenic maturation,leading to increased bone formation within *** findings highlight the unappreciated role of cellular crosstalk between osteoclasts and preosteoblasts/osteoblasts as a driver of FD pathology and demonstrate a novel mechanism of action of denosumab in the treatment of bone disease.

Design of an automated robotic microinjection system for batch injection of zebrafish embryos and larvae

This paper presents the design of a vision-based automated robotic microinjection system for batch injection of both zebrafish embryos and larvae.A novel visual recognition algorithm based on an automatic threshold and excessive dilatation is introduced to accurately identify the center of zebrafish embryos and larval yolks.A corresponding software system is developed using the producer-consumer model as the framework structure,and a friendly user interface is designed to allow operators to choose from a range of desired functions according to their different *** addition,a novel microstructural agarose device is designed and fabricated to simultaneously immobilize mixed batches of embryos and ***,a prototype microinjection system is fabricated by integrating hardware devices with visual *** experimental study is conducted to verify the performance of the robotic microinjection *** results show that the reported system can accurately identify zebrafish embryos and larvae and efficiently complete batch microinjection tasks of the mixtures with an injection success rate of 92.05%in 13.88 s per *** with manual and existing microinjection systems,the proposed system demonstrates the merits of versatility,excellent efficiency,high success rate,high survival rate,and sufficient stability.

Phase Transitions and Seepage Characteristics during the Depletion development of Deep Condensate Gas Reservoirs

Deep condensate gas reservoirs exhibit highly complex and variable phase behaviors,making it crucial to understand the relationship between fluid phase states and flow *** study conducts a comprehensive analysis of the actual production process of the deep condensate gas well A1 in a certain oilfield in *** phase behavior analysis and CMG software simulations,the study systematically investigates phase transitions,viscosity,and density changes in the gas and liquid phases under different pressure conditions,with a reservoir temperature of 165°*** research covers three crucial depletion stages of the reservoir:single-phase flow,two-phase transition,and two-phase *** findings indicate that retrograde condensation occurs when the pressure falls below the dew point pressure,reachingmaximum condensate liquid production at around *** pressure decreases,gas phase density and viscosity gradually decrease,while liquid phase density and viscosity show an increasing *** the initial single-phase flow stage,maintaining a consistent gas-oil ratio is observed when both bottom-hole and reservoir pressures are higher than the dew point ***,a sudden drop in bottom-hole pressure below the dew point triggers the production of condensate oil,significantly reducing subsequent gas and oil *** the transitional two-phase flow stage,as the bottom-hole pressure further decreases,the reservoir exhibits a complex flow regime with coexisting areas of gas and *** the subsequent two-phase flow stage,when both bottom-hole and reservoir pressures are below the dew point pressure,a significant increase in the gas-oil ratio is *** reservoir manifests a two-phase flow regime,devoid of single-phase gas flow *** lowpressure conditions in deep condensate gas reservoirs,considerations include gas injection,gas lift,and cyclic gas injection and production in surrounding ***,techniques such as hot nitrogen or CO

development of a Proof-of-Concept Multi-Method Computer Simulation to Support Rural healthcare Disaster Preparedness Planning

Due to a lack of resources, rural communities often face challenges when planning catastrophic events. This project involved applying systems thinking and model-based systems engineering to develop a proof-of-concept, multi-method computer simulation and then determining whether the simulation could be used to assess the efficacy of disaster planning approaches on health outcomes in rural communities, as a function of primary healthcare. The project focus was a rural or non-urban healthcare system experiencing a natural hazard. Both system dynamics and discrete event models were incorporated to represent subsystem operations, crucial disaster responses, as well as three key response systems: public health, emergency management, and healthcare. The subsystem models included several components: policies/procedures, communications,resources, exercises/drills/training, healthcare space and staff, and the flow of affected people into and through the *** combined simulation can serve as a first step to a more comprehensive approach to helping rural communities achieve more efficient and effective healthcare planning for disaster responses.

Individual and joint exposures to PMconstituents and mortality risk among the oldest-old in China

Cohort evidence linking long-term survival of older adults with exposure to fine particulate matter (PM2.5) constituents remains scarce in *** constructing a dynamic cohort based on the Chinese Longitudinal healthy Longevity Study,we aimed to assess the individual and joint associations of major PM2.5constituents with all-cause death in Chinese oldest-old （≥80 years） ***-dependent Cox proportional hazards models were adopted to estimate death risks of long-term exposure to *** 14,884 participants,totaling 56,342 person-years of follow-up,12,346 deaths were *** highest mortality risk associated with an interquartile range（IQR） increase in exposure was 1.081 （95%confidence interval[CI]:1.055–1.108） for sulfate (IQR=4.1μg m–3),followed by 1.078 （95%CI:1.056–1.101） for black carbon (IQR=1.6μg m–3),1.056 （95%CI:1.028–1.084） for ammonium (IQR=3.2μg m–3),1.050 （95%CI:1.021–1.080） for nitrate (IQR=5.8μg m–3),and 1.049 （95%CI:1.024–1.074） for organic matter (IQR=10.3μg m–3).In joint exposure,each IQR-equivalent rise of all five PM2.5constituents was associated with an 8.2%（95%CI:4.0%–12.6%） increase in mortality *** weight analysis indicated the predominant role of sulfate and black carbon in driving PM2.5-related *** （aged 80–89 years） and rural dwellers were at significantly greater risk of mortality from individual and joint exposures to *** study suggests that later-life exposure to PM2.5constituents,particularly sulfate and black carbon,may curtail long-term survival of the oldest-old in China.
