Interferon-induced transmembrane protein 3 (IFITM3) is a restriction factor that can be induced by viral infectionand interferons (IFNs). It inhibits the entry and replication of many viruses, which are independent of receptorusage but dependent on processes that occur in endosomes. In this study, we demonstrate that IFITM3 playsimportant roles in regulating the RNA-virus-triggered production of IFN-β in a negative-feedback *** of IFITM3 inhibited Sendai virus-triggered induction of IFN-β, whereas knockdown of IFITM3 hadthe opposite effect. We also showed that IFITM3 was constitutively associated with IRF3 and regulated thehomeostasis of IRF3 by mediating the autophagic degradation of IRF3. These findings suggest a novel inhibitoryfunction of IFITM3 on the RNA-virus-triggered production of type I IFNs and cellular antiviral responses.

Evaluation of Apatite-coated Chitosan Microspheres for Bone Regeneration

The apatite-coated chitosan microspheres were fabricated by incubating chitosan in five times simulated body fluid. The apatite deposited on the microspheres was similar to natural bone mineral, as demonstrated by scanning electron microscopy, X-ray diffraction spectra and Fourier transformed-infrared spectroscopy analyses. Rat bone marrow-derived mesenchymal stem cells(BMSCs) were seeded on apatitecoated microspheres to investigate the effect of this scaffold on cell proliferation and differentiation. BMSCs seeded on uncoated microspheres were served as a control. In vivo experiment was evaluated by transplanting the microspheres loaded with or without BMSCs in 5-mm cranial defects of Wistar rats. Bone regeneration was investigated via micro-CT and histological analysis. It was found that apatite-coated chitosan microspheres could significantly promote the proliferation and alkaline phosphatase activity of BMSCs compared with uncoated microspheres. Histological slices and Micro-CT images at 8 weeks revealed much better regeneration of bone in the apatite-coated microspheres loaded with BMSCs than the control. In addition, the defect filled with pure microspheres induced little new bone formation. Our findings suggest that the apatite-coated chitosan microspheres scaffold is a promising carrier of stem cells for cranial bone tissue engineering.

There are few studies on the membrane protein Ankfyl. We have found Ankfyl is specifically expressed in neural stem/precursor cells during early development in mice （murine）. To further explore Ankfyl function in neural development, we developed a gene knockout mouse with a mixed Balb/C and C57/BL6 genetic background. Using immunofluorescence and in situ hybridization, neural defects were absent in mixed genetic Ankfyl null mice during development and in adults up to 2 months old. However, Ankfyl gene knockout mice with a pure genetic background were found to be lethal in the C57/BL6 inbred mice embryos, even after seven generations of backcrossing. Polymerase chain reaction confirmed homozygotes were unattainable as early as embryonic day 11.5. We conclude that Ankfyl protein is dispensable in neural stem/precursor ceils, but could be critical for early embryonic murine development, depending on the genetic background.

Objective:To evaluate the personality and psychosocial status of adult malocclusion patients through the Eysenck Personality Questionnaire(EPQ) and the Symptom Checklist 90(SCL-90). Methods:The EPQ and the SCL-90 were administered to 348 adult respondents with Angle’s ClassⅠ,Ⅱ,Ⅲmalocclusion and normal *** and psychosocial traits were analyzed and compared among the ***:Patients in Angle’s ClassⅠ,D andⅢgroups scored lower on the EPQ-E and higher on the EPQ-N than those in the normal occlusion group, whereas patients in the Class II group scored higher on the *** mean scores of ClassⅠ,ⅡandⅢgroups were significantly higher than those of the normal group on somatization, obsessive-compulsiveness,interpersonal sensitivity,depression,anxiety,and paranoid ideation. All SCL-90 scores were significantly positively correlated with *** and neuroticism scores of female respondents were higher than those of male *** impact of education was greatest on the EPQ-P and the EPQ-E of adult personality,whereas the impact of deformity was greatest on the EPQ-N of adult ***:Personality and psychosocial status show differences in adult patients with ClassⅠ,Ⅱ,Ⅲmalocclusion and normal occlusion and can be influenced by gender,deformity,age and education.

Human dental pulp stem cells(hDPSCs)are easily obtained multipotent cells,however,their potential value in regenerative medicine is hindered by the phenotypic and functional changes after conventional monolayer ***,we employed single-cell RNA sequencing(scRNA-seq)to comprehensively study the transcriptional difference between the freshly isolated and monolayer cultured *** cell cluster analysis based on our scRNA-seq data showed that monolayer culture resulted in a significant cellular composition switch compared to the freshly isolated ***,one subpopulation,characterized as MCAM(+)JAG(+)PDGFRA(−),maintained the most transcriptional characteristics compared to their freshly isolated ***,immunofluorescent staining revealed that the MCAM(+)JAG(+)PDGFRA(−)hDPSCs uniquely located in the perivascular region of human dental pulp ***-cytometry analysis confirmed that their proportion remained relatively stable(~2%)regardless of physiological senescence or dental *** with the annotation of scRNA-seq data,MCAM(+)JAG(+)PDGFRA(−)hDPSCs showed higher proliferation capacity and enhanced in vitro multilineage differentiation potentials(osteogenic,chondrogenic and adipogenic)compared with their counterparts PDGFRA(+)***,the MCAM(+)JAG(+)PDGFRA(−)hDPSCs showed enhanced bone tissue formation and adipose tissue formation after 4-week subcutaneous implantation in nude *** together,our study for the first time revealed the cellular composition switch of monolayer cultured hDPSCs compared to the freshly isolated *** in vitro expansion,the MCAM(+)JAG(+)PDGFRA(−)subpopulation resembled the most transcriptional characteristics of fresh hDPSCs which may be beneficial for further tissue regeneration applications.

The effect of glass ionomer cement and resin-modified glass ionomer cement incorporated with chlorhexidine and bioactive glass on antimicrobial activity and physicochemical properties were investigated. The experimental results showed that groups incorporated with 1% chlorhexidine exhibited a significant reduction of optical density values of the bacterial suspension and increased the degradation of Streptococcus mutans biofilm. However, groups incorporated with 10% bioactive glass did not affect the optical density values and the biofilm formation. The mechanical properties of the materials and the polymerization were not influenced by the addition of chlorhexidine. Nevertheless, the compressive strength was lower when the materials were incorporated with bioactive glass. It can be concluded that glass ionomer cements incorporated with chlorhexidine can maintain its mechanical properties as well as reduce early S mutans biofilm formation. Controlled release/sustained release technology may be required to optimize the antibacterial activity of glass ionomer cements incorporated with bioactive glass.

The cranial neural crest(CNC)cells play a vital role in craniofacial development and *** are multi-potent progenitors,being able to differentiate into various types of *** pre-migratory and post-migratory CNC cells are plastic,taking on diverse fates by responding to different inductive ***,what sustains the multipotency ofCNCcells andderivatives remains largely *** this study,we present evidence that FGF8 signaling is able to sustain progenitor status and multipotency of CNC-derived mesenchymal cells both in vivo and in *** show that augmented FGF8 signaling in pre-migratory CNC cells prevents cell differentiation and organogenesis in the craniofacial region by maintaining their progenitor ***-derived mesenchymal cells with Fgf8 overexpression or control cells in the presence of exogenous FGF8 exhibit prolonged survival,proliferation,and multi-potent differentiation capability in cell ***,exogenous FGF8 also sustains the capability of CNC-derived mesenchymal cells to participate in organogenesis such as ***,FGF8-mediated signaling strongly promotes adipogenesis but inhibits osteogenesis of CNC-derived mesenchymal cells in *** results reveal a specific role for FGF8 in the maintenance of progenitor status and in fate determination of CNC cells,implicating a potential application in expansion and fate manipulation of CNC-derived cells in stem cell-based craniofacial regeneration.
