Skin aging results in an imbalance between synth.sis and degradation of th. extracellular matrix. Overproduction of degradative enzymes (MMPs) and oxygen free radicals during ch.onological and ph.to-induced aging leads a degradation of th. network and elastic skin collagen. Our previous work demonstrated th.t a culture supernatant of gingival stem cells h.d anti-aging activities in vitro, ex vivo and in vivo in h.mans. h.wever, th.s culture medium is very complex and currently not responding to th. European cosmetics regulation. After th. analysis of th. culture medium we h.ve isolated 4 molecules interesting in terms of skin anti-aging activity: TIMP1, Selenium, Folic Acid and Glycin (TSAG). After th. study of th. most efficiency concentration of each.molecule, th. composition of TSAG is tested on irradiated UVA h.man dermal fibroblast and h.man skin. TSAG preserves th. elastin and collagen network and inh.bits th. MMP1, MMP3 and MMP9 activities. Th. advantage of th.s mimetic solution of stem cells is to be stable, easily reproducible and non-h.man origin. Th. outlook for cosmetology seems interesting.

AIMTo test wh.th.r a delayed and sh.rt course of rapamycin would induce immunosuppressive effects following allogeneic orth.topic liver transplantation (OLT) in *** OLTs were performed using Dark Agouti livers transplanted into Lewis recipients, and syngeneic OLTs were performed using th. Lewis rat strain. Rapamycin (1 mg/kg per day) was administered by gavage from day 4 to day 11 post-transplantation. Lymph.cyte cellular compartments were analyzed by flow cytometry in draining lymph.nodes, non-draining lymph.nodes and th. spleen at days 11 and 42 in rapamycin-treated rats, untreated control rats and syngeneic grafted rats. Skin grafts from Dark agouti or from F344 RT were performed at day 30 on liver grafted rats treated with.*** 8-d course of rapamycin treatment initiated 4 d following transplantation resulted in th. survival of grafted rats for more th.n 100 d. In contrast, untreated rats died of liver failure with.n 13 to 21 d. Th. analysis of th. cellular compartment revealed an increase in two cellular subpopulations, specifically myeloid-derived suppressor cells (MDSCs) and CD8+CD45RClow T cells, with.ut major modifications in th. regulatory T cell (Treg) compartment in treated rats in th. early stages after grafting. We evaluated th. ability of treated rats to reject th.rd-party allogeneic skin grafts to confirm th.ir immune competence. In contrast, wh.n skin was collected from rats syngeneic to th. grafted liver, it was not *** results demonstrate th.t sh.rt and delayed rapamycin treatment allows for tolerance in allogeneic OLT. Th. results also allowed for th. identification of th. mech.nisms of tolerance induced by rapamycin by identifying MDSCs and CD8+CD45RClow T cells as associated with.th. state of tolerance.

AIM： To assess pre-orth.topic liver transplantation （OLT） factors th.t could be evaluated pre-operatively or controlled post-operatively associated with.h.patocellular carcinoma （h.C） recurrence and disease-free survival after liver transplantation （LT）.METh.DS： Four h.ndred and twelve patients transplanted for h.C between 1988 and 1998 in 14 French.centers, wh. survived th. postoperative period were studied. Kaplan Meier estimates were calculated for 24 variables potentially associated with.recurrence of h.C. Uni- and multivariate analyses were conducted to identify independent predictors of recurrence. RESULTS： Overall 5-year disease-free survival was 57.1%. By univariate analysis, variables associated with.disease-free survival were： presence of cirrh.sis （P = 0.001）, etiology of liver disease （P = 0.03）, α fetoprotein level （〈 200, 200 to 2000, or 〉 2000; P 〈 0.0001）, y-GT activity （N, N to 2N or 〉 2N; P = 0.02）, th. number of nodules （1, 2-3 or ≥ 4; P = 0.02）, maximal diameter of th. largest nodule （〈 3 cm, 3 to 5 cm or 〉 5 cm; P 〈 0.0001）, th. sum of th. diameter of th. nodules （〈 3 cm, 3 to 5 cm, 5 to 10 cm or 〉10 cm; P 〈 0.0001）, bilobar location （P = 0.01）, preoperative portal th.ombosis （P 〈 0.0001）, peri-operative treatment of th. tumor （P = 0.002） and ch.moembolization （P = 0.03）, tumor differentiation （P = 0.01）, initial type of calcineurin inh.bitor （P = 0.003）, th. use of antilymph.cyte antibodies （P = 0.02）, rejection episodes （P = 0.003） and period of LT （P 〈 0.0001）. By multivariate analysis, 6 variables were independently associated with.h.C recurrence： maximal diameter of th. largest nodule （P 〈 0.0001）, time of LT （P 〈 0.0001）, tumor differentiation （P 〈 0.0001）, use of anti-lymph.cyte antibody （ATG） or anti-CD3 antibody （OKT3）（P = 0.005）, preoperative portal th.ombosis （P = 0.06） and th. number of nodules （P = 0.06）. Skin aging sh.ws an imbalance between synth.sis and degradation of th. extracellular matrix. Th. overproduction of degradative enzymes (MMPs) during th. ch.onology- and ph.to-induced aging leads to a degradation of th. elastic and collagen networks. In a model of collagen and elastin destruction, we sh.wed th.t th. gingival fibroblast was able to preserve th.se macromolecules by inh.biting th. overproduction of metalloproteinases by overproduction of TIMP-1 and modulation of th. inflammatory cytokines activity. Th. objective of th.s study is to evaluate th. effect of th. gingival fibroblasts on h.man skin. Th. results in vitro and ex vivo sh.w th.t th. gingival fibroblast protects th. skin collagen and elastic network by th. inh.bition of MMPs wh.ch.leads to an overproduction of th. TIMP-1. Moreover, th. gingival fibroblast modulates th. activity of some enzymes responsible for th. inflammation;th.y inh.bit th. IL-1β and stimulate th. production of TGF-β1. In vivo studies with.a duration of six month. and 50 women with.pronounced wrinkles sh.w th.t th. culture supernatant of gingival fibroblasts diluted to 5% leads to a statistically significant decrease in th. number and length.of wrinkles.

Burkitt＇s lymph.ma is a h.gh.y aggressive non-h.dgkin lymph.ma, often presenting in extra-nodal sites. It generally h.s a poor spontaneous outcome and needs aggressive treatment with.systemic and intrath.cal ch.moth.rapy. Occurrence at th. gastric site is rare. We report th. case of a 39-year old woman wh. presented with.a prominent ulcerated lesion of th. antrum corresponding h.stologically to a Burkitt＇s lymph.ma associated with.h./icobacter py/ori （h.pylori） infection. Interph.se fluorescence in situ h.bridization （FISh. demonstrated c-MYC gene rearrangement in tumour cells with.ut BCL2 or BCL6 gene translocations. Ulcer h.aling and tumour regression with.a complete h.stological response were obtained 8 wk after h.ylori eradication. In spite of th.s complete remission, taking into account th. h.gh.risk of recurrence, th. patient received systemic and intrath.cal ch.moth.rapy. Two years later, th. patient remained in complete remission. Th.s is th. first report of a gastric Burkitt＇s lymph.ma responding to h.ylori eradication. Th.se findings raise th. question of th. potential role of h.pylori in th. path.genesis of some gastric Burkitt＇s lymph.mas, and sh.w th. importance of search.ng for and eradicating th. bacteria in combination with.conventional ch.moth.rapy regimens.

目的基于血液筛查核酸检测反应性献血者的h.V感染的确认,探讨核酸检测反应性献血者的归队策略。方法联合应用自建的高灵敏度核酸检测体系、血液核酸筛查等多种核酸检测(NAT)方法,并结合血清学检测、献血者随访,对核酸检测反应性(NAT-yield)献血者中的h.V感染进行确认和感染状态识别。依据确认的h.V感染血浆样本,比较不同确认方法、确认指标或指标组合对h.V感染确认的效果。结果2010年11月—2021年2月,在血液筛查检出的876位NAT-yield献血者中共确认h.V感染者511人(OBI 451人,急性早期h.V感染者27人,不能确认感染者33人,无感染者30人,不能确认h.V感染者335人)。采用单检系统对混检系统检出的h.V感染血浆进行复测的检出率为96.6%,明显高于混检系统对单检系统检出的h.V DNA反应性(h.V DNA R)组和鉴别试验无反应性(NDR)组的复测检出率(76.4%和55.7%)(Ph.V DNA R样本复测检出率无明显差异(P>0.05),但均明显高于NDR样本(Ph.V DNA。OBI献血者中抗-h.c+占比为90.2%,单独抗-h.c+为49.2%,远高于不能确认感染组(Ph.eAg、抗-h.e和抗-h.c IgM在OBI和不能确认感染组中的比例极低且无差异(P>0.05)。结论近60%的NAT-yield献血者可以确认h.V感染。为保证献血者归队的安全性,需要更高灵敏度的h.V DNA确证技术提高h.V感染的确认率。抗-h.c是NAT-yield献血者OBI风险排查和归队评估最重要的血清学指标。
