Chrysanthemum morifolium cv.‘Huaihuang’has ornamental,edible,medicinal,and tea product ***,its field growth,yield,and quality are negatively affected by black spot disease caused by Alternaria sp.(Strain:HQJH10092301;GenBank accession number:KF688111).In this study,we transcriptionally and transgenically characterized a new cultivar,‘Huaiju 2#’(henan Traditional chinese Medicine Plant Cultivar identification number:2016002),which was bred from‘Huaihuang’and shows resistance to Alternaria ***‘Huaiju 2#’plants were inoculated with Alternaria *** three or five *** analysis showed increases in both salicylic acid(SA)and jasmonic acid(JA)in infected plants compared to the *** activity analysis also revealed a significant increase in defense enzyme activities in infected ***-Seq of plants infected for 3 or 5 days produced a total of 58.6 GB of clean *** these reads,16,550 and 13,559 differentially expressed genes(DEGs)were identified in Cm_3 dpi(sample from 3 days post-inoculation labeled as Cm_3 dpi)and Cm_5 dpi(sample from 5 days post-inoculation labeled as Cm_5 dpi),respectively,compared with their controls(Cm_0 d:a mixture samples from 0 d(before inoculation)and those treated with sterile distilled water at 3 dpi and 5 dpi).Gene annotation and cluster analysis of the DEGs revealed a variety of defense responses to Alternaria ***,which were characterized by increases in resistance(R)proteins and the reactive oxygen species(ROS),Ca2+,mitogen-activated protein kinase(MAPK),and JA signaling *** particular,SA signaling was highly responsive to Alternaria *** qPCR analysis of 12 DEG candidates supported their differential expression characterized by using the RNA-Seq *** candidate was CmNPR1(nonexpressor of pathogenesis-related gene 1),an important positive regulator of SA in systemic acquired resistance(SAR).Overexpression of CmNPR1 in‘Huaiju 2#’increased the resistance of transgen

[ Objective ] The paper was to explore effects of Tomato aspermy virus （TAV） -free on chlorophyll content and nitrogen metabolism of Chrysanthemum morifolium Ramat. ＇Huaibai＇, in order to provide a theoretical basis for field popularization and application of TAV-free ＇ Huai bai＇ plantlets. [ Method] With previously acquired TAV-free test-tube plantlets of ＇ Huaibai＇ as the experimental materials and its ordinary test-tube plantlets as the control （CK）, chlorophyll content and some indexes related to nitrogen metabolism in leaves were determined during subculture. [ Result] During subculture, the contents of chlorophyll a and chlorophyll b in leaves of CK and TAV-free ＇ Huaibai＇ test-tube plantlets increased with the subculture days. Both realized the largest increase at the 22＇h day of subculture, and their increase amplitudes were 5.24% and 41.69%, respectively. Compared with CK, the chlorophyll a and chlorophyll b contents of TAV-free test-tube plantlets increased at the same time point of subculture, with significant difference （P 〈0.05） or extremely significant difference （P 〈0.01 ）. Further- more, the nitrate reductase （NR） activity, free proline and soluble protein contents of TAV-free ＇ Huaibai＇ were all significantly higher than those of CK, and the highest increase amplitudes were 27.99% （ subculture for 22 d）, 28.10% （ subculture for 22 d） and 5.89% （ subculture for 38 d）, respectively. [ Conclusion] TAV-free could effectively improve chlorophyll content and nitrogen metabolism level of ＇ Huaibai＇ , and was conducive to its growth and development.

To explore the effect of tomato aspermyvirus(TAV)-free on physiology of Chrysanthemum morifolium Ramat.’ Huaibai’.TAV-free ’ Huaibai’ test-tube plantlets were used as experimental materials in this *** results showed that;(1) TAV-free didn’ t change the trend of photosynthetic indexes(the chlorophyll a,chlorophyll b and total chlorophyll contents) and N metabolism related indexes(the NR activity,Proline and soluble protein contents) of during subculture,but significantly increased the chlorophyll a contents,chlorophyll b contents and total chlorophyll contents at the same time,significantly increased the NR activity,Proline and soluble protein contents,so TAV-free improved photosynthetic capacity and N metabolism level.(2) TAV-free didn’ t change the trend of lipid peroxidation related indexes(O · produce rate,the MDA and HO contents) and protection system related indexes(protective enzymes SOD,POD,CAT,GR and APX activities;antioxidants ASA and GSH contents) during subculture,but significantly reduced the absolute value of lipid peroxidation related indexes and significantly increased activities of protective enzyme and the contents of antioxidants at the same time of *** TAV-free reduced the active oxygen metabolism and improved its oxidation resistance.

Breeding and Evaluation of New Iron Yam (Dioscorea opposita Thunb) Varieties

[Objective] This study was conducted to develop some new iron yam （Dioscorea opposita） varieties with excellent cooking, eating and nutritional qualities. [Method] The fresh weight per plant, drying rate, dry weight per plant, yield, allan- toin content, water soluble extract content and disease resistance of seven new iron yam cultivars （No.1, No.2, No.4, No.6, No.8, No.9 and No.10） obtained through spaceflight mutation breeding were measured and compared with control （No.11）. Then, the nutrition quality of two elite cultivars No. 6 and No.10 was determined by comparing with the control. [Result] The eight iron yam cultivars ranked in descend- ing order of their fresh weight per plant as follows： No.6（0.255 kg）〉No.10（0.254 kg）〉 No.4（0.242 kg）〉No.9（0.237 kg）〉No.1（0.233 kg）〉No.11（0.206 kg）〉No.2（0.191 kg）〉No.8 （0.157 kg）. There was no significant difference in fresh weight per plant between No.6 and No.10, but extremely significant difference between No.6 and the control, No.10 and the control. The eight iron yam cultivars ranked in descending order of their drying rate as follows： No.2 （32.641%）〉No.10 （32.230%）〉No.9 （28.223%）〉No.6 （25.174%）〉No.8（25.122%）〉No.11 （25.043%）〉No.1 （24.291%）〉No.4（20.234%）. The dry- ing rate of both No.6 and No.10 was higher than that of the control. The eight iron yam cultivars ranked in descending order of their dry weight per plant as follows： No.10（0.082 kg）〉No.9（0.067 kg）〉No.6（0.064 kg）〉No.2（0.062 kg）〉No.1（0.056 kg）〉No.11 （0.052 kg）〉No.4 （0.049 kg）〉No.8 （0.039 kg）. The dry weight per plant of both No.6 and No.10 is higher than that of the control. The eight iron yam cultivars ranked in descending order of their allantoin content as follows No.6 （0.484%）〉No.4 （0.467%）〉 No.10 （0.399%）〉*** （0.386%）〉No.9 （0.378%）〉No.8 （0.350%）〉No.2 （0.342）〉No.1 （0.325%）.
