Herbs-partitioned moxibustion alleviates aberrant intestinal epithelial cell apoptosis by upregulating A20 expression in a mouse model of Crohn's disease

BACKGROUND A20 inhibits intestinal epithelial cell apoptosis in Crohn's disease, and herbspartitioned moxibustion(HPM) has been demonstrated to be an effective treatment for Crohn's disease. However, the mechanism by which HPM reduces intestinal epithelial cell apoptosis in Crohn's disease has not been thoroughly elucidated to *** To elucidate whether HPM exerts its effects by upregulating A20 to affect intestinal epithelial cell apoptosis in a Crohn's disease mouse *** In this study, mice with A20 deletion in intestinal epithelial cells(A20 IEC-KO) were utilized to establish a Crohn's disease mouse model with 2,4,6-trinitrobenzenesulfonic acid(TNBS) administration, as well as wild-type mice. Mice were randomly divided into normal control(NC), model control(MC), mesalazine(MESA), and HPM groups. The morphology of the colonic mucosa was observed by hematoxylin-eosin staining, and serum endotoxin and apoptosis of epithelial cells were evaluated by enzyme-linked immunosorbent assay and terminal dUTP nick-end labeling assay accordingly. The protein expression levels of A20 and tumor necrosis factor receptor 1(TNFR1)-related signaling molecules were evaluated by Western blot, and co-expression of A20 and TNFR1-associated death domain(TRADD) and co-expression of A20 and receptor-interacting protein 1(RIP1) were observed by double immunofluorescence *** The intestinal epithelial barrier was noted to have an improvement in the HPM group of wild-type(WT) mice compared with that in A20 IEC-KO mice. Compared with A20 IEC-KO HPM mice, serum endotoxin levels and apoptosis percentages were decreased(P < 0.01), A20 expression levels were increased(P < 0.01), and expression of TNFR1, TRADDD, and RIP1 was decreased in the HPM group of WT mice(PTNFR1 < 0.05, PTRADD < 0.01, PRIP1 < 0.01). Protective effects of electroacupuncture on acetylsalicylic acid-induced acute gastritis in rats

AIM: To investigate the protective effects of electroacupuncture (EA) pretreatment on acetylsalicylic acid (ASA)-induced ulceration in rats. METHODS: We randomly divided 72 rats into three groups including control (administered with distilled water), ASA group (administered 100 mg/kg ASA) and EA group (administered EA + 100 mg/kg ASA). Each rat was fasted for 18 to 24 h before experimentation, and lesion scores, gastric acidity, cyclooxygenase (COX)-1 and -2 mRNA levels, and total nitric oxide (NO) concentration were measured. RESULTS: The lesion scores of the EA group were significantly lower than those of the ASA group. Gastric acidity of the ASA and EA groups was reduced compared to the control group. COX-1 and -2 mRNA levels were significantly increased in the EA group as compared to the control and ASA groups, and NO levels were also significantly increased in the EA group as compared to the ASA group. CONCLUSION: These results suggest that EA-mediated protection against ASA-induced ulceration in rats may occur via gastric defense components.

Moxibustion for Managing Type 2 Diabetes Mellitus:A Systematic Review

Acupuncture at Baihui and Dazhui reduces brain cell apoptosis in heroin readdicts

Acupuncture at Baihui （GV20） and Dazhui （GV14） reduces neuronal loss and attenuates ultra- structural damage in cerebral ischemic rats. However, whether acupuncture can treat addiction and prevent readdiction through changes to brain cell ultrastructure remains unknown. In this study, cell apoptosis was observed in the hippocampus and frontal lobe of heroin readdicted rats by electron microscopy. Immunohistochemical staining displayed a reduction in Bcl-2 ex- pression and an increase in Bax expression in the hippocampus and frontal lobe. After rats were given acupuncture at Baihui and Dazhui, the pathological damage in the hippocampus and frontal lobe was significantly reduced, Bcl-2 expression was upregulated and Bax expression was downregulated. Acupuncture exerted a similar effect with methadone, a commonly used drug for clinical treatment of drug addiction. Experimental findings suggest that acupuncture at Dazhui and Baihui can prevent brain cell apoptosis in heroin readdicted rats.

目的:观察电针(EA)预处理对脂多糖(LPS)所致急性肺损伤(ALI)大鼠肺泡巨噬细胞(AMs)M1极化的影响,探讨EA保护ALI的可能机制.方法:将40只Sprague-Dawley大鼠随机分为正常组、模型组和3个电针预处理组(包括尺泽组、足三里组和尺泽加足三里组),每组8只.以LPS[2 mg/(kg·bw)]滴注大鼠气管复制ALI大鼠模型.各电针预处理组均于模型制备前6 d于相应双侧穴位行电针预处理(1次/d,30 min/次).模型制备3 h后大鼠进行肺功能检测,取肺组织计算肺的湿重/干重比值(W/D),苏木素-伊红染色观察肺组织病理改变,并进行肺损伤评分;酶联免疫吸附法检测大鼠支气管肺泡灌洗液(BLAF)中肿瘤坏死因子(TNF)-α、白细胞介素(IL)-1β及髓过氧化物酶(MPO)的含量,荧光定量聚合酶链反应法和免疫印迹法分别检测肺泡M1型巨噬细胞标志物分化抗原簇86(CD86)、诱导型一氧化氮合酶(iNOS)及其信号通路Toll样受体(TLR)4、核因子-κB(NF-κB)p65 mRNA和蛋白表达.结果:LPS诱导3 h后大鼠肺功能显示第0.1秒用力呼气量(FEV0.1)、第0.3秒用力呼气量(FEV0.3)以及各自占用力肺活量(FVC)之比(FEV0.1/FVC;FEV0.3/FVC)均显著下降(P<0.01);肺组织W/D增高(P<0.01);肺损伤评分显著升高;BALF中TNF-α、IL-1β、MPO含量及AMs中CD86、iNOS、TLR4、NF-κB p65的mRNA及蛋白表达明显升高(P<0.01).经电针预处理后,FEV0.1、FEV0.3、FEV0.1/FVC及FEV0.3/FVC均显著上升(P<0.01);肺损伤评分降低(P<0.01);BALF中TNF-α、IL-1β、MPO含量及肺组织中CD86、iNOS、TLR4、NF-κB p65的mRNA及蛋白表达明显下降(P<0.05或P<0.01).尺泽加足三里组BALF中TNF-α、IL-1β、MPO含量及AMs中CD86、iNOS、TLR4、NF-κB p65的mRNA表达比其他两单穴组下降明显(P<0.01).结论:电针预处理尺泽和足三里对LPS所致大鼠ALI起到抑制炎症和减轻肺损伤的作用,且两穴合用优于单一穴位,其效应机制可能与电针下调TLR4/NF-κB信号通路从而抑制AMs向M1型极化有关.
