Exercise has long been known for its active role in improving physical fitness and sustaining *** moderate-intensity exercise improves all aspects of human health and is widely accepted as a preventative and therapeutic strategy for various *** is well-documented that exercise maintains and restores homeostasis at the organismal,tissue,cellular,and molecular levels to stimulate positive physiological adaptations that consequently protect against various pathological *** we mainly summarize how moderate-intensity exercise affects the major hallmarks of health,including the integrity of barriers,containment of local perturbations,recycling and turnover,integration of circuitries,rhythmic oscillations,homeostatic resilience,hormetic regulation,as well as repair and ***,we summarize the current understanding of the mechanisms responsible for beneficial adaptations in response to *** review aimed at providing a comprehensive summary of the vital biological mechanisms through which moderate-intensity exercise maintains health and opens a window for its application in other health *** hope that continuing investigation in this field will further increase our understanding of the processes involved in the positive role of moderate-intensity exercise and thus get us closer to the identification of new therapeutics that improve quality of life.

中医以整体观来认识人的健康与疾病防治,在中国的应用已有数千年历史,传向国际亦有百余年。但是中医与现代医学在疾病理论机理上有很多差别。在现代医学中,免疫学是最复杂的系统之一,是针对病原体的整体防御系统,通过系统平衡调节参与疾病的发生与进展。特别值得一提的是,中医与免疫学在整体观、防御机制、平衡调节,昼夜时辰等方面有着共通之处。本综述重点关注中医和免疫学之间的相同与不同,用现代免疫学的数据阐明传统中医理论的生物学机理。我们相信不断进步的多组学技术,可以深入诠释中医理论机理,用现代免疫学数据为中医的治疗原理和应用疗效提供更多的生物学证据。

Parkin Somatic Mutations Link Melanoma and Parkinson's Disease

Epidemiological studies suggest a direct link between melanoma and Parkinson＇s disease（PD）; however, the underlying molecular basis is unknown. Since mutations in Parkin are the major driver of early-onset PD and Parkin was recently reported to play a role in cancer development, we hypothesized that Parkin links melanoma and PD. By analyzing whole exome/genome sequencing of Parkin from 246 melanoma patients, we identified five non-synonymous mutations, three synonymous mutations, and one splice region variant in Parkin in3.6% of the samples. In vitro analysis showed that wild-type Parkin plays a tumor suppressive role in melanoma development resulting in cell-cycle arrest, reduction of metabolic activity, and apoptosis. Using a mass spectrometry-based analysis, we identified potential Parkin substrates in melanoma and generated a functional protein association network. The activity of mutated Parkin was assessed by protein structure modeling and examination of Parkin E3 ligase activity. The Parkin-E28 K mutation impairs Parkin ubiquitination activity and abolishes its tumor suppressive effect. Taken together, our analysis of genomic sequence and in vitro data indicate that Parkin is a potential link between melanoma and Parkinson＇s disease. Our findings suggest new approaches for early diagnosis and treatment against both diseases.

Main text In the past decades,there have been several studies con-cerning the efficacy of sequencing of chemotherapy on disease control and survival in locoregionally advanced(LA)nasopharyngeal carcinoma(NPC).The addition of concurrent cisplatin to radiotherapy has demonstrated survival improvements that are attributable to both dis-tant metastasis and locoregional *** to the latter,the advent of intensity-modulated radiotherapy has resulted in superior tumor control given the bet-ter dosimetry compared to conventional techniques[1].However,distant recurrence still occurs in 20-30%patients and accounts for the main cause of *** address this,several groups have explored the advantages of adding neoadjuvant chemotherapy(NACT)or adju-vant chemotherapy(ACT)to the backbone of concurrent chemoradiotherapy(CCRT).

Dear Editor,Chromosomal translocations result from the interchange of geneticmaterial between non-homologous *** translocations are formed by erroneous repair of double-stranded breaks(DSBs)via non-homologous end joining(NHEJ)[1].Some genotoxic drugs produce DSBs and thus present a major risk factor for the development of oncogenic chromosomal *** risk factors that interfere with translocationprone DSB repair,once DSBs are already formed,are obscure,and potential effects of drugs on translocation formation during this step have never been explored.
