Regulating effect of glycyrrhetinic acid on bronchial asthma smooth muscle proliferation and apoptosis as well as inflammatory factor expression through ERK1/2 signaling pathway

Objective: To study the influence of glycyrrhetinic acid(GA) on bronchial asthma(BA)smooth muscle proliferation and apoptosis as well as inflammatory factor expression and its molecular ***: Male SD guinea pigs were selected and made into asthma models, bronchial asthma smooth muscle cells were cultured and divided into BA group, GA group and GA + LM group that were treated with serum-free RPMI1640 culture medium, serumfree RPMI1640 culture medium containing 50 ng/mL glycyrrhetinic acid, serum-free RPMI1640 culture medium containing 50 ng/mL glycyrrhetinic acid and 100 ng/mL LM22B-10 respectively; normal guinea pigs were collected and bronchial smooth muscle cells were cultured as control group. The cell proliferation activity as well as the expression of proliferation and apoptosis genes, inflammatory factors and p-ERK1/2 was ***: Proliferation activity value and m RNA expression of Bcl-2, TNF-α, IL-4, IL-6,YKL-40, protein expression of p-ERK1/2 of airway smooth muscle cell in BA group were significantly higher than those of control group while m RNA expression levels of Bax,caspase-9 as well as caspase-3 were significantly lower than that of control group(P < 0.05); proliferation activity value and m RNA expression of Bcl-2, TNF-α, IL-4, IL-6, YKL-40, protein expression of p-ERK1/2 of airway smooth muscle cell in GA group were significantly lower than those of BA group(P < 0.05) while the m RNA expression levels of Bax, caspase-9 as well as caspase-3 were significantly higher than those of BA group(P < 0.05); proliferation activity value and m RNA expression of Bcl-2, TNF-α, IL-4, IL-6, YKL-40 of airway smooth muscle cell in GA + LM group were significantly higher than those of GA group(P < 0.05) while m RNA expression levels of Bax, caspase-9 as well as caspase-3 were significantly lower that of GA group(P < 0.05).Conclusion: GA can inhibit the proliferation of bronchial smooth muscle cells and reduce the expression of inflammatory fa

[Evaluation of the toxicity of perfluorooctanoic acid toward human colorectal cancer cells using multi-dimensional approaches]

While human exposure to perfluorooctanoic acid (PFOA) can lead to ulcerative colitis, the molecular mechanisms responsible for PFOA-induced intestinal toxicity are unclear. Herein, we examined the toxicity of PFOA toward human colorectal cancer cells (HCT116) from three dimensions: the cytotoxic phenotype, cell respiration, and transcription levels of metabolism-related genes. Formazan was used to assess how PFOA exposure affects HCT116-cell relative viability, after which the mitochondrial respiratory activities of these cells were determined by analyzing extracellular flux. The quantitative real-time polymerase chain reaction (qPCR) method was used to detect metabolism-related gene expression levels. The cytotoxicity assay revealed that the HCT116 showed significantly inhibited relative activities compared to those of the control when exposed to 300 μmol/L PFOA for 48 h (development of PFOA-induced ulcerative colitis. Evaluation criteria for gas source rocks of marine carbonate in China

Hydrocarbon generating and expulsion simulation experiments are carried out using samples artifically matched between the acid-dissolved residue of relatively low-maturity limestone and the original sample. This work makes up for the insufficiency of source rock samples with high abundance of organic matters and low maturity in China. The organic carbon content of the 10 prepared samples varies between 0.15% and 0.74%. Pyrolysis data and simulation experiment results of hydrocarbon generating and expulsion, which were obtained by a high-temperature and high-pressure open system, indicate that the lower limit of organic carbon content for marine carbonate rock to generate and expel hydrocarbons is 0.23%—0.31%. In combination with the numerical analysis of organic carbon in marine carbonate rocks from Tarim Basin, Sichuan Basin, Ordos Basin and North China, as well as the contribution of these gas source rocks to the discovered gas pools, we think that the organic carbon criterion for carbonate gas source rocks should be 0.3%.

Comparison of updated birth weight,length and head circumference charts by gestational age in China with the INTERGROWTH-21 st NCSS charts:a population-based study

Formation Laws of Inorganic Gas Pools in the Northern Jiangsu Basin

In the Northern Jiangsu basin there are high pure CO2 gas pools, low condensed oil-containing CO2 gas pools, high condensed oil-containing CO2 gas pools and He-containing natural gas pools, with the δ13Cco2 (PDB) values ranging from ?2.87%o to ?6.50%o, 3He/4He 3.71 × 10?6 to 6.42 × 10?6, R/Ra 2.64 to 4.5, 40Ar/36Ar 705 to 734, belonging to typical mantle source inorganic gas pools which are related to young magmatic activity. The gas layers occur in two major reservoir-caprock systems, the terrestrial Meso-Cenozoic clastic rock system and the marine Meso-Palaeozoic carbonate rock-clastic rock system. Controlled by the difference in the scale of traps in the two reservoir-caprock systems, large and medium-scale inorganic gas pools are formed in the marine Meso-Palaeozoic Group and only small ones are formed in the terrestrial Meso-Cenozoic strata. Inorganic gas pools in this basin are distributed along the two deep lithospheric faults on the west and south boundaries of the basin. Gas pools are developed at the intersected part of the ENE-trending faults that control the half graben and the E-W tenso-shear faults, mainly distributed near the Es1, Ny1 and Ny2-Q basalt eruption centres.

Energy consumption and emission mitigation prediction based on data center traffic and PUE for global data centers

With the rapid development of technologies such as big data and cloud computing,data communication and data computing in the form of exponential growth have led to a large amount of energy consumption in data ***,data centers will become the world’s largest users of energy consumption,with the ratio rising from 3%in 2017 to 4.5%in *** to its unique climate and energy-saving advantages,the high-latitude area in the Pan-Arctic region has gradually become a hotspot for data center site selection in recent *** order to predict and analyze the future energy consumption and carbon emissions of global data centers,this paper presents a new method based on global data center traffic and power usage effectiveness(PUE)for energy consumption ***,global data center traffic growth is predicted based on the Cisco’s ***,the dynamic global average PUE and the high latitude PUE based on Romonet simulation model are obtained,and then global data center energy consumption with two different scenarios,the decentralized scenario and the centralized scenario,is analyzed quantitatively via the polynomial fitting *** simulation results show that,in 2030,the global data center energy consumption and carbon emissions are reduced by about 301 billion kWh and 720 million tons CO2 in the centralized scenario compared with that of the decentralized scenario,which confirms that the establishment of data centers in the Pan-Arctic region in the future can effectively relief the climate change and energy *** study provides support for global energy consumption prediction,and guidance for the layout of future global data centers from the perspective of energy ***,it provides support of the feasibility of the integration of energy and information networks under the Global Energy Interconnection conception.

Prussian blue nanosphere-embedded in situ hydrogel for photothermal therapy by peritumoral administration

To establish an injectable hydrogel containing Prussian blue(PB) nanospheres for photothermal therapy against cancer, PB nanospheres were prepared by one-pot synthesis and the thermosensitive Pluronic F127 was used as the hydrogel matrix. The PB nanospheres and the hydrogel were characterized by shape, particle size, serum stability, photothermal performance upon repeated 808 nm laser irradiation, as well as the rheological features. The effect of the PB nanospheres and the hydrogel were evaluated qualitatively and quantitatively in 4T1 mouse breast cancer cells. The retention,photothermal efficacy, therapeutic effects and systemic toxicity of the hydrogel were assessed in a tumor-bearing mouse model. The PB nanospheres had a diameter of about 150 nm and exhibited satisfactory serum stability, photo-heat convert ability and repeated laser exposure stability. The hydrogel encapsulation did not negatively influence the above features of the photothermal agent. The nanospherecontaining hydrogel showed a phase transition at body temperature and, as a result, a long retention time in vivo. The photothermal agent-embedded hydrogel displayed promising photothermal therapeutic effects in the tumor-bearing mouse model with little-to-no systemic toxicity after peritumoral administration.
