A new cyclometalated iridium（Ⅲ） complex [Ir（2-pq）2（HPIP）]C1 （lrl, 2-pq=3-phenylisoquinoline, HPIP= 2-（2-hydroxyphenyl）imidazo[4,5-f] 1,10-phenanthroline） was synthesized and applied to image mitochondria in living cells. Irl displayed excellent ability to selectively accumulate in mitochondria of live cells with no requirement of replacement of the culture medium. Due to Irl exhibiting better photostability than the commercially available mitochondrial trackers, this complex could be applicable for the imaging and tracking of the mitochondrial mor- phological changes over long periods of time. In addition, in vitro cytotoxicity investigation revealed that it! showed negligible cytotoxicity at the concentrations employed. Based on these, Irl is suitable for organelle-selective imaging in living cells.

Fluorescent graphene oxide(GO) nanoparticles were obtained from the thermal carbonization of citrate *** on the synthesizing temperature,the size of GO varied from several to several hundred *** to the confinement from the size,green and blue emissions at around 504 and 450 nm were observed from the GO *** emissions could be dynamically quenched by titrating against copper(Ⅱ)(Cu^(2+)) ions,and the emission intensity was reduced exponentially as a function of Cu^(2+)*** quenching mechanism was ascribed to the bridging of the surface-coOH andOH groups by Cu^(2+),which restricted the vibration of edge atoms or clusters and reduced the number of luminophores of GO *** a result,the concentration of Cu^(2+)was detectable with the fluorescent intensity of GO.

Genetic Analysis and Molecular Mapping of a Rolling Leaf Mutation Gene in Rice

A rice mutant with rolling leaf, namely γ-rl, was obtained from M2 progenies of a native indica rice stable strain Qinghuazhan （QHZ） from mutagenesis of dry seeds by γ-rays. Genetic analysis using the F2 population from a cross between this mutant and QHZ indicated the mutation was controlled by a single recessive gene. In order to map the locus for this mutation, another F2 population with 601 rolling leaf plants was constructed from a cross between y-rl and a japonica cultivar 02428. After primary mapping with SSR （simple sequence repeats） markers, the mutated locus was located at the short arm of chromosome 3, flanked by RM6829 and RM3126. A number of SSR, InDel （insertion/deletion） and SNP （single nucleotide polymorphism） markers within this region were further developed for fine mapping. Finally, two markers, SNP121679 and InDe1422395, were identified to be flanked to this locus with genetic distances of 0.08 cM and 0.17 cM respectively, and two SNP markers, SNP75346 and SNPl10263, were found to be co-segregated with this locus. These results suggested that this locus was distinguished from all loci for the rolling leaf mutation in rice reported so far, and thus renamed rl10（t）. By searching the rice genome database with closely linked markers using BLAST programs, an e-physical map covering rl10（t） locus spanning about a 50 kb region was constructed. Expression analysis of the genes predicted in this region showed that a gene encoding putative flavin-containing monooxygenase （FMO） was silenced in γ-rl, thus this is the most likely candidate responsible for the rolling leaf mutation.

Human lysozyme （HL） inhibits Fusarium oxysporum （FocR4） growth in vitro. To obtain transgenic bananas （Musa spp.） that are resistant to Panama wilt （F. oxysporum）, we introduced an HL gene that is driven by a constitutive cauliflower mosaic virus 35S promoter into the banana via Agrobacteriummediated transformation. PCR confirmed that 51 transgenic plants were obtained. The development of Panama wilt symptoms were examined after the plants had been grown in pots. The non-transgenic plants developed typical fusarium symptoms 60 d after FocR4 inoculation, whereas 24 of 51 transgenic plants remained healthy. The transgenic banana plants that showed resistance to FocR4 in the pots were then planted in a field that was heavily infected with FocR4 for further investigation. Eleven of 24 plants devel- oped symptoms before bud emergence; another 11 plants showed symptoms after bud emergence and the remaining two plants, H-67 and H-144, remained healthy and were able to fruit. Northern blotting analysis demonstrated that H-67 and H-144, bearing the strongest resistance to Panama wilt, had the highest level of HL expression and that the expression of HL was well correlated with the FocR4 resistance of transgenic plants. We conclude that Agrobacterium-mediated transformation, with the assistance of particle bombardment, is a powerful approach for banana transformation and that a transgenic HL gene can cause resistance of the crop to FocR4 in the field.

Plant mitochondrial genes are often transcribed into complex sets of mRNA. To characterize the transcription initiation and promoter structure, the transcript termini of four mitochondrial genes, atpl, atp6, cob, rps7, in rice （Oryza sativa L.）, were determined by using a modified circularized RNA reverse transcription- polymerase chain reaction method. The results revealed that three genes （atp1, atp6, rps7） were transcribed from multiple initiation sites, indicating the presence of multiple promoters. Two transcription termination sites were detected in three genes （atp6, cob, rps7）, respectively. Analysis on the promoter architecture showed that the YRTA （Y=T or C, R=A or G） motifs that are widely present in the mitochondrlal promoters of other monocot and dicot plant species were detected only in two of the 12 analyzed promoters. Our data suggest that the promoter sequences in the rice mitochondrial genome are highly diverged in comparison to those in other plants, and the YRTA motif is not an essential element for the promoter activity.

As a member of cyclic nucleotide phosphodiesterase(PDE)enzyme family,PDE10A is in charge of the degradation of cyclic adenosine(cAMP)and guanosine monophosphates(cGMP).While PDE10A is primarily expressed in the medium spiny neurons of the striatum,it has been implicated in a variety of neurological ***,inhibition of PDE10A has proven to be of potential use for the treatment of central nervous system(CNS)pathologies caused by dysfunction of the basal ganglia–of which the striatum constitutes the largest component.A PDE10A-targeted positron emission tomography(PET)radioligand would enable a better assessment of the pathophysiologic role of PDE10A,as well as confirm the relationship between target occupancy and administrated dose of a given drug candidate,thus accelerating the development of effective PDE10A *** this study,we designed and synthesized a novel ^(18)F-aryl PDE10A PET radioligand,codenamed[^(18)F]P10A-1910([^(18)F]9),in high radiochemical yield and molar activity via spirocyclic iodonium ylide-mediated radiofluorination.[^(18)F]9 possessed good in vitro binding affinity(IC_(50)=2.1 nmol/L)and selectivity towards ***,[^(18)F]9 exhibited reasonable lipophilicity(logD=3.50)and brain permeability(P_(app)>10×10^(−6) cm/s in MDCK-MDR1 cells).PET imaging studies of[^(18)F]9 revealed high striatal uptake and excellent in vivo specificity with reversible tracer *** studies in rodents revealed an improved plasma and brain stability of[^(18)F]9 when compared to the current reference standard for PDE10A-targeted PET,[^(18)F]***,dose–response experiments with a series of escalating doses of PDE10A inhibitor 1 in rhesus monkey brains confirmed the utility of[^(18)F]9 for evaluating target occupancy in vivo in higher *** conclusion,our results indicated that[^(18)F]9 is a promising PDE10A PET radioligand for clinical translation.

The type V-I CRISPR-Cas system is becoming increasingly more attractive for genome ***,natural nucleases of this system often exhibit low efficiency,limiting their ***,we used structure-guided rational design and protein engineering to optimize an uncharacterized Cas12i nuclease,*** a result,we developed Cas-SF01,a Cas12i3 variant that exhibits significantly improved gene editing activity in mammalian ***-SF01 shows comparable or superior editing performance compared to SpCas9 and other Cas12 *** to natural Cas12i3,Cas-SF01 has an expanded PAM range and effectively recognizes NTTN and noncanonical NATN and TTVN *** addition,we identified an amino acid substitution,D876R,that markedly reduced the off-target effect while maintaining high on-target activity,leading to the development of CasSF01^(HiFi)(high-fidelity Cas-SF01).Finally,we show that Cas-SF01 has high gene editing activities in mice and *** results suggest that CasSF01 can serve as a robust gene editing platform with high efficiency and specificity for genome editing applications in various organisms.

Objective A strain of Aspergillus niger(A. niger), capable of releasing bound phenolic acids from wheat bran, was isolated. This strain was identified by gene sequence identification. The antioxidant and anti-inflammatory capacity of ferulic acid released from wheat bran by this A. niger strain(FA-WB) were evaluated. Methods Molecular identification techniques based on PCR analysis of specific genomic sequences were conducted; antioxidant ability was examined using oxygen radical absorbance capacity(ORAC), cellular antioxidant activity(CAA) assays, and erythrocyte hemolysis assays. RAW264.7 cells were used as a model to detect anti-inflammatory activity. Results The filamentous fungal isolate was identified to be A. niger. ORAC and CAA assay showed that FA-WB had better antioxidant activity than that of the ferulic acid standard. The erythrocyte hemolysis assay results suggested that FA-WB could attenuate AAPH-induced oxidative stress through inhibition of reactive oxy gen species(ROS) generation. FA-WB could significantly restore the AAPH-induced increase in intracellular antioxidant enzyme activities to normal levels as well as inhibit the intracellular malondialdehyde formation. TNF-?, IL-6, and NO levels indicated that FA-WB can inhibit the inflammation induced by lipopolysaccharide(LPS). conclusion Ferulic acid released from wheat bran by a new strain of A. niger had good anti-inflammatory activity and better antioxidant ability than standard ferulic acid.

Background: The root of Tripterygium wilfordii(Tripterygii radix), a natural powerful traditional Chinese medicine(TCM) for various diseases treatment, has been used for centuries in the Asian countries as anti-rheumatoid arthritis(RA) agent, antioxidant agent, and anti-inflammatory agent. Its combination with other herbs in treating RA has been explored. The anti-RA effect of T. wilfordii for cancer treatment has been supported by some evidence. Aims and Objectives: To investigate the anticancer mechanism of T. wilfordii, bioinformatics databases were used to identify its active ingredients. Materials and Methods: Target proteins associated with cancer were determined using a network pharmacology analysis platform, and 25 key active compounds and 55 key targets of T. wilfordii were identified in our study. A common potential mechanism of T. wilfordii involvement in cancer was disclosed by in-depth network analysis of diseases, functions, and ***, the analysis results of the TCM-disease target protein interaction network revealed 5 potential targets;subsequently, a total of 30targets(these 5 targets, as well as 25 previously identified compounds) were subjected to molecular docking. Results: Our results showed that the therapeutic effect of T. wilfordii in cancer is characterized by multiple components, targets, and pathways. The regulation of signaling pathways such as Kaposi sarcoma-associated herpes virus infection, colorectal cancer, small-cell lung cancer, and prostate cancer may be the important pharmacodynamic basis of anticancer therapy. conclusion: Triptonoditerpenic acid inhibited proliferation and induced apoptosis in SW480 cells. The mechanism may be related to the downregulation of Bcl-2 expression, upregulation of Bax mRNA expression, and expression inhibition of PTGS2.
