Reveal the pharmacodynamic substances and mechanism of an edible medicinal plant Rhodiola crenulate in DSS-induced colitis through plasma pharmacochemistry and metabolomics

Rhodiola crenulate is the edible medicinal herbal medicine widely used for altitude sickness in China.Interestingly,our previous work has found that R.crenulate extract(RCE)could significantly improve the pathology associated with dextran sulfate sodium-induced colitis.Thus,the current research aims to reveal the pharmacodynamic material basis of RCE,as well as its mechanism against colitis.The chemical characterization of RCE was performed by UHPLC-HR-MS,through which a total of 88 constituents were identified.Meanwhile,our results also found 29 constituents absorbed into blood and 8 metabolized absorbable compounds.The decreased flavonoids prototype and the elevated sulfated products of phenols were observed under pathophysiological conditions of colitis.The metabolomics study revealed that colitis caused the alternation of fatty acid metabolism,steroid hormone biosynthesis and bile acid metabolism.Correspondingly,RCE could prevent colitis by improving fatty acid metabolism and secondary bile acid metabolism.

(±)-Cyclopeltains A and B(1 and 2),two pairs of unusual alkylated adenine derivatives,were isolated from the insect Cyclopelta parva and structurally characterized.1 and 2 both possess a 6/5/7/5 ring system arising from the alkylation of N^(7) and N^(10) by a C6 unit and the formation of a furan ring.A plausible biosynthetic pathway for 1 and 2 was proposed.Biological evaluation of the enantiomers of 1 and 2 revealed that(−)-1 and(+)-2 could inhibit the production of TNF-αand IL-6 and the expression of iNOS,COX-2,and p-NF-κB.

Identification and characterization of chemical constituents in Mahuang Guizhi Decoction and their metabolites in rat plasma and brain by UPLC-Q-TOF/MS

Objective:Mahuang Guizhi Decoction(MGD),an essential herbal pair in traditional Chinese medicine,is able to release cold,fever and asthma,mainly containing alkaloids,flavonoids,phenylpropanoids and amino acids.However,the absorption and distribution of these four category compounds in vivo still remained unclearly.Methods:In our research,we utilized UPLC-Q-TOF-MS technique to identify the constituents within MGD,as well as the prototypes of MGD and their metabolites absorbed in plasma and brain.We further profiled the drug-time curve of prototypes and metabolites of MGD both in plasma and brain.Results:Our results showed that 105 constituents were characterized in MGD.Thirty of them could be absorbed into blood,and ten of them could be distributed into brain.We also discovered eight new bio-transformed metabolites in blood,and a half of which could pass through the blood-brain barrier.In addition,all components detected in vivo could be absorbed and distributed immediately.Conclusion:These findings provide an approachable method to analyze the potential bio-active compounds in MGD and their in vivo behaviors,which could promote the efficacious material basis study of MGD and the security of clinical utilization.

Boswelliai ns A-E(1-5),five new cembra ne-type diterpenoids,were isolated from the resin of Boswellia papyifera.Their structures,determined by using spectroscopic,computational and chemical methods,demonstrate that 2-5 are novel 4,5-seco-,4,5-seco-,9,10-seco-,and 6,7-seco-cembrane diterpenoids,and that 1,4 and 5 bear aldehyde moieties.An evaluation of the wound healing properties of these natural products along with a derivative of 2 shows that compounds 2 and 3 significantly stimulate proliferation,mobility and tuber formation of human umbilical vein endothelial cells(HUVECs)at the concentration of 20μmol/L.

Among the numerous fruit species of Baccaurea ramiflora Lour.,‘LR’(white flesh)and‘BR’(pink flesh)are two kinds of local strains with high edibility.In order to study the metabolic causes of taste differences,we performed the non-targeted metabonomics analysis of‘LR’and‘BR’using LC-MS/MS.541 metabolites were totally identified,and 45 kinds of metabolites(carbohydrates,fatty acids,flavonoids and terpenoids,etc.)were different between the two strains.The results indicate L-sorbose,D-(+)-glucose,citric acid,L-phenylalanine and oleamide,α-eleostearic acid were the main primary metabolites.The significant difference existed in pathways of unsaturated fatty acids between the studied two strains by pathway enrichment analysis.The results demonstrate that the different in composition,as well as the abundance of primary and secondary metabolites may be the potential causes of taste differences,which provides a new insight into the possible metabolic factors setting off the changing taste of B.ramiflora.

Rhodiola crenulate is the edible medicinal herbal medicine widely used for altitude sickness in China. 41种亚热带植物大量元素和微量元素含量对干湿季节环境变化的不同反应

季节性气候变化引起的植物叶片元素含量差异反映了植物的潜在生长策略。但季节性气候变化介导的元素分布模式尚不清楚,因此限制了对植物生长状态的评估和对全球气候变化下植物生长动态的预测。我们在湿季和干季时间段采集了41种亚热带常绿阔叶植物(包括:31种树木和10种灌木)叶片,通过电感耦合等离子体质谱仪(ICP-MS)测定了叶片中的大量和微量元素含量。研究结果显示,41种植物叶片大量元素含量在湿季显著大于干季,微量元素含量在湿季显著低于干季。湿季环境下乔木层植物叶片大量元素含量最高,干季环境下乔木层植物和湿季环境下灌木层植物次之,干季环境下灌木层植物最小;干季环境下乔木层植物和灌木层植物叶片微量元素含量最高,湿季环境下灌木层植物次之,最少的是湿季环境下乔木层植物。这些研究结果说明,不同垂直结构植物叶片大量元素和微量元素含量受季节气候变化的影响,湿季到干季的转变不利于乔木层植物的生长,而对灌木层植物生长的影响较小。这些发现对于预测亚热带常绿阔叶林不同垂直结构植物如何适应湿季和干季环境变化以及未来全球气候变化提供了宝贵的证据。
