Porous scaffolds as succedaneum of natural bone were investigated and applied in medical *** this work, we carried out studies on mechanical properties of solid parts and porous scaffolds obtained by additive manufacturing(AM) *** is found that productions of AM process have a higher yield strength and higher microhardness compared to commercial *** surface was observed for layer-by-layer process of AM and sticking of powder *** machining accuracy is affected by both dimensions and ***, mechanical properties of porous scaffolds are influenced by machining accuracy and *** addition, the unit cell structures also impact the mechanical properties of porous scaffolds in terms of elastic modulus, yield strength and failure ***,considering the mechanical properties and biological properties, scaffolds with cube(CB) crystal cells are the best choice in our study.

Sucrose synthase(SUS,EC 2.***.1.13)is widely considered as a key enzyme involved in plant sucrose metabolism,and the gene family encoding different SUS isozymes has been identified and characterized in several plant ***,to date scant information about the SUS genes is available in Litchi chinensis ***,we identified five SUS genes in *** Lc SUSs shared high levels of similarity in both nucleotide and amino acid *** gene structure,phylogenetic relationships,and expression profiles were *** structure analysis indicated that the Lc SUSs have similar exon-intron *** analysis revealed that the five members could be classified into three groups(LcSUS1 and LcSUS2 in SUSⅡ,LcSUS4 and LcSUS5 in SUSⅢ,and LcSUS3 in SUSⅠ),demonstrating evolutionary conservation in the SUS family across litchi and other plant *** expression levels of Lc SUSs were investigated via real-time PCR in various tissues and different developmental stages of *** tissues and organs,Lc SUSs exhibited distinct but partially redundant expression profiles in litchi,being predominantly expressed in young leaves(sink).During aril development,the expression pattern of LcSUS1 was consistent with the trend of sugar accumulation,indicating it may play important roles in determination of sink strength in ***,transcript levels of LcSUS2,LcSUS4,and LcSUS5 varied between cultivars with different hexose/sucrose ratios,which may regulate the sugar composition in *** results provide insights into physiological functions of SUS genes in litchi,especially roles in regulating sugar accumulation in aril.

As one of the hallmarks of cancer,metabolic reprogramming leads to cancer progression,and targeting glycolytic enzymes could be useful strategies for cancer *** screening a small molecule library consisting of 1320 FDA-approved drugs,we found that penfluridol,an antipsychotic drug used to treat schizophrenia,could inhibit glycolysis and induce apoptosis in esophageal squamous cell carcinoma(ESCC).Gene profiling and Ingenuity Pathway Analysis suggested the important role of AMPK in action mechanism of *** using drug affinity responsive target stability(DARTS)technology and proteomics,we identified phosphofructokinase,liver type(PFKL),a key enzyme in glycolysis,as a direct target of *** could not exhibit its anticancer property in PFKL-deficient cancer cells,illustrating that PFKL is essential for the bioactivity of *** PFKL expression is correlated with advanced stages and poor survival of ESCC patients,and silencing of PFKL significantly suppressed tumor ***,direct binding of penfluridol and PFKL inhibits glucose consumption,lactate and ATP production,leads to nuclear translocation of FOXO3a and subsequent transcriptional activation of BIM in an AMPK-dependent *** together,PFKL is a potential prognostic biomarker and therapeutic target in ESCC,and penfluridol may be a new therapeutic option for management of this lethal disease.

BACKGROUND Both Gilbert's syndrome(GS)and hereditary spherocytosis(HS)are common genetic ***,comorbidity of GS with HS has always been considered a rare phenomenon,and it can impede accurate diagnoses in the presence of isolated unconjugated *** SUMMARY In a study on Levitt’s carbon monoxide(CO)breath test for the differential diagnosis of isolated hyperbilirubinemia,we found six GS patients with HS in 6 *** patients,including five males and one female,aged 25-58 years,were from four families and generally in good *** chronic fluctuating jaundice and/or hyperbilirubinemia had been diagnosed as simple constitutional jaundice for 6-30 *** function tests showed isolated unconjugated hyperbilirubinemia with serum total bilirubin ranging from 20.7-75.4μmol/*** hemoglobin was normal in five cases,and slightly decreased in one(11.5 g/dL).Overt hemolytic signs were absent,while erythrocyte lifespan determined by the newly developed Levitt’s CO breath test was significantly short(15-50 d),definitely demonstrating the presence of *** that their unconjugated hyperbilirubinemia compared inappropriately with hemolytic severity,as indicated by the hemoglobin level,further combined genetic tests for both UGT1A1 and hereditary erythrocyte deficiencies were *** tests confirmed,at last,the coexistence of GS with *** Comorbidity of GS and HS might not be uncommon in isolated unconjugated *** CO breath test would sensitively detect the hemolysis,the discordance between the hyperbilirubinemia and hemoglobin level could strongly indicate the coexistence of GS and HS.

The existing recycling and regeneration technologies have problems,such as poor regeneration effect and low added value of products for lithium(Li)-ion battery cathode materials with a low state of *** this work,a targeted Li replenishment repair technology is proposed to improve the discharge-specific capacity and cycling stability of the repaired LiCoO_(2) cathode *** with the spent cathode material with>50%Li deficiency,the Li/Co molar ratio of the regenerated LiCoO_(2) cathode is>0.9,which completely removes the Co_(3)O_(4) impurity phase formed by the decomposition of LixCoO_(2) in the failed cathode material after *** repaired LiCoO_(2) cathode mater-ials exhibit better cycling stability,lower electrochemical impedance,and faster Li^(+)diffusion than the commercial materials at both 1 and 10 ***,Li_(1.05)CoO_(2) cathodes have higher Li replenishment efficiency and cycling *** energy consumption and greenhouse gas emissions of LiCoO_(2) cathodes produced by this repair method are significantly reduced compared to those using pyrometallurgical and hydro-metallurgical recycling processes.

The latent infection by herpes virus type 1(HSV-1)may be lifelong in trigeminal ganglia and a suspected cause of Alzheimer's Disease(AD)and Amyotrophic lateral sclerosis(ALS).Whether and how N6-methyladenosine(m6A)modification of viral RNAs affects virus infection are poorly ***,we report that HSV-1 infection enhanced the expression of m6A writers(METTL3,METTL14)and readers(YTHDF1/2/3)at the early infection stage and decreased their expression later on,while suppressed the erasers'(FTO,ALBKH5)expression immediately upon infection to facilitate viral *** m6A modification by 3-deazaadenosine(DAA)significantly decreased viral replication and reduced viral reproduction over 1000 *** interestingly,depleting the writers and readers by siRNAs inhibited virus replication and reproduction;whereas depleting the erasers promoted viral replication and *** YTHDF3 strikingly decreased viral replication by up to 90%,leading to reduction of up to 10-fold viral replication and over 100-fold virus reproduction,*** of m6A initiator METTL3(by 60%–70%)by siRNA correlatedly decreased viral replication 60%–70%,and reduced virus yield over ***,ectopic expression of METTL3 largely increased virus ***3 knockdown suppressed the HSV-1 intermediate early and early genes(ICP0,ICP8 and UL23)and late genes(VP16,UL44,UL49 and ICP47);while ectopic expression of METTL3 upregulated these gene *** from our study shed the lights on the importance for m6A modification to initiate HSV-1 early *** components of m6A modification machinery,particularly m6A initiator METTL3 and reader YTHDF3,would be potential important targets for combating HSV-1 infections.

Endometrial cancer(EC)is the second most prevalent female reproductive system tumor after cervical cancer and its incidence is rising globally.1 Recently,N6-methyladenosine(m^(6)A)has emerged as an important regulator of a variety of physiological and pathological processes,especially in promoting EC progression.2,3 However,the potential roles of m^(6)A modification in EC and its immune landscape remain ***,the correlation between m^(6)A modification patterns and tumor microenvironment(TME)cell-infiltrating characteristics was comprehensively evaluated based on 21 m^(6)A regulators of 1301 endometrial cancer *** identified four m6Aclasses characterized by distinct TME cell *** used principal component analysis algorithms to construct m6Ascore to quantify m^(6)A modification patterns of endometrial cancer *** results showed that lower m6Ascore may be closely related to immune-inflamed phenotype and displayed immune-activated characteristics and better prognosis.

OBJECTIVE: To assess the effectiveness of violateoil from Blumea Balsamifera(L.) DC. leaves(BB oil)on wound healing in ***: Undiluted BB oil and its diluted solutions with olive oil to 1/5 and 1/10 to yield BB oil1/5and BB oil1/10 were applied to the wounded skin before wound healing conditions were assessed by healing rate, histopathology, and contents of collagen, hydroxyproline, and Neuropeptide Substance P(SP). All above results were compared with the efficacies of the control, pure olive oil, basic fibroblast growth factor(BFGF), and cream of Jing Wan Hong(JWH).RESULTS: BB oil1/5and BB oil1/10 improved wound contraction and closure. Histopathology study further confirmed a desirable histological organization of wound tissues. BB oil1/5and BB oil1/10 reduced the number of inflammatory cells, increased wound-healing rates, and significantly increased the hydroxyproline content. Both BB oil1/5and BB oil1/10 improved formation of collagen, and reduced the frequency of fibroblasts. Moreover, BB oil1/5and BB oil1/10 markedly promoted SP expression. However, undiluted BB oil may induce skin thickening and hardening, inhibite collagen synthesis and delay complete skin wound ***: The BB oil1/5and BB oil1/10 promoted capillary regeneration, blood circulation, collagen deposition, granular tissue formation, epithelial deposition, and wound contraction. The mechanism underlying the action might be related to induction of SP secretion, and the proliferation and differentiation of mesenchymal cells.
