AIM:To compare the visual perception(color and chromatic-achromatic contrast vision)of a small cohort of COVID-19 patients at the time of infection and after 6mo with that of a healthy population matched for sex and ***:A total of 25 patients(9 females,16 males,mean age:54±10y)with COVID-19 hospitalized in the COVID-19 Unit of the University clinical Hospital of Valladolid were recruited for this preliminary *** perception,as determined by monocular measurement of contrast sensitivity function(CSF)and color vision was assessed in each patient using the Optopad *** results obtained were then compared with those of a sample of 16 age-and sex-matched healthy controls(5 females,11 males,mean age:50±6y)in which the same measurement procedure was *** significant differences between groups were assessed using the Mann-Whitney U *** were repeated after a minimum follow-up period of 6mo and statistically significant differences between the two time points in each group were assessed using the Wilcoxon signed rank ***:Discrimination thresholds(color and chromatic-achromatic contrast vision)and their corresponding sensitivity,calculated as the inverse of the discrimination threshold,were *** of the data revealed higher contrast threshold results(i.e.,worse contrast sensitivity)in the COVID-19 group than in the control group for all spatial frequencies studied in the Optopad-CSF achromatic test and most of the spatial frequencies studied in the Optopad-CSF chromatic test for the red-green and blue-yellow *** addition,color threshold results in the COVID-19 group were also significantly higher(i.e.,worse color sensitivity)for almost all color mechanisms studied in the Optopad-Color *** 6mo,most of the differences found between the groups were maintained despite COVID-19 ***:The present results provide preliminary evidence that visual perception may be impaired in COVID-19

Background:None of the published studies involving cancer cachexia experimental models have included a measure of the severity of the syndrome like the scoring system previously developed for human *** aim of the present investigation was to define and validate a cachexia score usable in both rat and mouse tumor ***:In order to achieve this goal,we included in the study one rat model(Yoshida AH‐130ascites hepatoma)and two mouse models(Lewis lung carcinoma and Colon26 carcinoma).The Animal cachexia score(ACASCO)includes five components:(a)body and muscle weight loss,(b)inflammation and metabolic disturbances,(c)physical performance,(d)anorexia,and(e)quality of life measured using discomfort symptoms and behavioral ***:Using the ACASCO values,three cut‐off values were estimated by applying hierarchical cluster *** groups were originally described,one exactly below the observed mean,a second exactly over the mean,and two other groups adjusted to every cue(inferior and superior).The three cut‐off values were estimated through maximization of the classification *** was accomplished by using a similarity matrix based on the metric properties of the variables and assuming multinormal *** results show that the four groups were:no cachexia,mild cachexia,moderate cachexia and advanced ***:The results obtained allow us to conclude that the score could be very useful as an endpoint in pre‐clinical studies involving therapeutic strategies for cancer *** potential usefulness of ACASCO relates to the primary endpoint in pre‐clinical cancer cachexia drug evaluations.

Neuroinformatics is a fascinating research field that applies computational models and analytical tools to high dimensional experimental neuroscience data for a better understanding of how the brain functions or dysfunctions in brain diseases. Neuroinformaticians work in the intersection of neuroscience and informatics supporting the integration of various sub-disciplines（behavioural neuroscience, genetics, cognitive psychology, etc.） working on brain research. Neuroinformaticians are the pathway of information exchange between informaticians and clinicians for a better understanding of the outcome of computational models and the clinical interpretation of the analysis. Machine learning is one of the most significant computational developments in the last decade giving tools to neuroinformaticians and finally to radiologists and clinicians for an automatic and early diagnosis-prognosis of a brain disease. Random forest（RF） algorithm has been successfully applied to high-dimensional neuroimaging data for feature reduction and also has been applied to classify the clinical label of a subject using single or multi-modal neuroimaging datasets. Our aim was to review the studies where RF was applied to correctly predict the Alzheimer＇s disease（AD）, the conversion from mild cognitive impairment（MCI） and its robustness to overfitting, outliers and handling of non-linear data. Finally, we described our RF-based model that gave us the 1 ^（st） position in an international challenge for automated prediction of MCI from MRI data.

Introduction:Machine learning (ML)‐based facial nerve injury (FNI) forecasting grounded on multicentric data has not been released up to *** distinct ML models,random forest (RF),K‐nearest neighbor,and artificial neural network (ANN),for the prediction of FNI were evaluated in this ***:A retrospective,longitudinal,multicentric study was performed,including patients who went through parotid gland surgery for benign tumors at three different university ***:Seven hundred and thirty‐six patients were *** most compelling aspects related to risk escalation of FNI were as follows:(1) location,in the mid‐portion of the gland,near to or above the main trunk of the facial nerve and at the top part,over the frontal or the orbital branch of the facial nerve;(2) tumor volume in the anteroposterior axis;(3) the necessity to simultaneously dissect more than one level;and (4) the requirement of an extended resection compared to a lesser extended *** contrast,in accordance with the ML analysis,the size of the tumor (>3 cm),as well as gender and age did not result in a determining favor in relation to the risk of ***:The findings of this research conclude that ML models such as RF and ANN may serve evidence‐based predictions from multicentric data regarding the risk of ***:Along with the advent of ML technology,an improvement of the information regarding the potential risks of FNI associated with patients before each procedure may be achieved with the implementation of clinical,radiological,histological,and/or cytological data.

Prognostic significance of lymphovascular invasion in colorectal cancer and its association with genomic alterations

BACKGROUND Lymphovascular invasion (LVI) is suggested to be an early and important step in tumor progression toward metastasis, but its prognostic value and genetic mechanisms in colorectal cancer (CRC) have not been well investigated. AIM To investigate the prognostic value of LVI in CRC and identify the associated genomic alterations. METHODS We performed a retrospective analysis of 1219 CRC patients and evaluated the prognostic value of LVI for overall survival by the Kaplan-Meier method and multivariate Cox regression analysis. We also performed an array-based comparative genomic hybridization analysis of 47 fresh CRC samples to examine the genomic alterations associated with LVI. A decision tree model was applied to identify special DNA copy number alterations (DCNAs) for differentiating between CRCs with and without LVI. Functional enrichment and protein-protein interaction network analyses were conducted to explore the potential molecular mechanisms of LVI. RESULTS LVI was detected in 150 (12.3%) of 1219 CRCs, and the presence was positively associated with higher histological grade and advanced tumor stage (both P group, the LVI group showed a 1.77-fold (95% confidence interval: 1.40-2.25, P data, 184 DCNAs (105 gains and 79 losses) were identified to be significantly related to LVI (P < 0.05), and the majority were located at 22q, 17q, 10q, and 6q. We further constructed a decision tree classifier including seven special DCNAs, which could distinguish CRCs with LVI from those without it at an accuracy of 95.7%. Functional enrichment and proteinprotein interaction network analyses revealed that the genomic alterations related to LVI were correlated with inflammation, epithelial-mesenchymal transition, angiogenesis, and matrix remodeling. 在实际应用数据中,我们所收集到的纵向数据可能受到测量上下限的影响,出现双侧删失的情形.此外,纵向数据的观测时间可能会携带响应变量部分的信息量.因此,在统计分析中,结合观测时间信息来分析双删失纵向数据十分必要.本文采用半参数回归模型来分析此类观测时间带信息的双删失纵向数据,通过构造鞅差估计方程来估计模型中的回归系数.同时,也指出参数估计量的渐近分布含有未知冗余参数.为进一步研究所得估计量的统计推断,本文应用随机加权方法计算估计量的渐近分布,并说明在给定样本下,随机加权估计的条件渐近分布与参数估计的渐近分布相同.为了计算参数的估计值,受交替乘子算法思想的启迪,本文建立一种新的参数计算算法,并通过数据模拟验证所提随机加权方法的可行性.最后,利用该模型分析临床试验中晚期大肠癌患者的生活质量数据,指出观测次数和治疗方法对患者生活质量有显著的影响.
