Rheumatoid arthritis(RA)is an incurable systemic autoimmune *** progression leads to joint deformity and associated loss of function,which significantly impacts the quality of life for sufferers and adds to losses in the labor *** the past few decades,RA has attracted increased attention from researchers,the abnormal signaling pathways in RA are a very important research field in the diagnosis and treatment of RA,which provides important evidence for understanding this complex disease and developing novel RA-linked intervention *** current review intends to provide a comprehensive overview of RA,including a general introduction to the disease,historical events,epidemiology,risk factors,and pathological process,highlight the primary research progress of the disease and various signaling pathways and molecular mechanisms,including genetic factors,epigenetic factors,summarize the most recent developments in identifying novel signaling pathways in RA and new inhibitors for treating *** interventions including approved drugs,clinical drugs,pre-clinical drugs,and cutting-edge therapeutic *** developments will hopefully drive progress in new strategically targeted therapies and hope to provide novel ideas for RA treatment options in the future.

Previous evidence suggests that emotion dysregulation may have different biological correlates between adults and children/adolescents. Although the role of genetic factors has been extensively studied in adult-onset emotion dysregulation, the genetic basis for pediatriconset emotion dysregulation remains elusive. The current review article presents a summary of previous studies that have suggested a few genetic variants associated with pediatric emotion dysregulation. Among these candidate loci, many prior studies have been focused on serotonin transporter promoter gene polymorphism 5-HTTLPR. Certain alleles of the 5-HTTLPR gene polymorphism have been found to be associated with traits associated with emotion dysregulation, such as aggression, affect reactivity, and insecure attachment. Additionally, genetic variants involving dopamine and neurophysiological biomarkers like the COMT Val158Met(rs460) and dopamine receptor D2/ ankyrin repeat and kinase domain containing one polymorphisms may play a role in emotion dysregulation. Inconsistent findings have been noted, possibly due to the heterogeneity in study designs and characteristics of different populations. Further research on the role of genetic predetermination of emotion dysregulation in children and adolescents is warranted.

Background: Dengue is the fastest spreading arboviral disease, posing great challenges on global public health. A reproduceable and comparable global genotyping framework for contextualizing spatiotemporal epidemiological data of dengue virus (DENV) is essential for research studies and collaborative ***: Targeting DENV-1 spreading prominently in recent decades, by reconciling all qualified complete E gene sequences of 5003 DENV-1 strains with epidemiological information from 78 epidemic countries/areas ranging from 1944 to 2018, we established and characterized a unified global high-resolution genotyping framework using phylogenetics, population genetics, phylogeography, and ***: The defined framework was discriminated with three hierarchical layers of genotype, subgenotype and clade with respective mean pairwise distances 2-6%, 0.8-2%, and ≤ 0.8%. The global epidemic patterns of DENV-1 showed strong geographic constraints representing stratified spatial-genetic epidemic pairs of Continent-Genotype, Region-Subgenotype and Nation-Clade, thereby identifying 12 epidemic regions which prospectively facilitates the region-based coordination. The increasing cross-transmission trends were also demonstrated. The traditional endemic countries such as Thailand, Vietnam and Indonesia displayed as persisting dominant source centers, while the emerging epidemic countries such as China, Australia, and the USA, where dengue outbreaks were frequently triggered by importation, showed a growing trend of DENV-1 diffusion. The probably hidden epidemics were found especially in Africa and India. Then, our framework can be utilized in an accurate stratified coordinated surveillance based on the defined viral population compositions. Thereby it is prospectively valuable for further hampering the ongoing transition process of epidemic to endemic, addressing the issue of inadequate monitoring, and warning us to be concerned about the cross-national, cross

目的探讨白细胞介素-4(IL-4)基因多态性与儿童哮喘患病风险的关系。方法采用以人群基础的病例对照研究(293例哮喘儿童和668例对照儿童)方法 ,进行IL-4C-590T基因多态与儿童哮喘风险关联分析,采用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)进行基因分型。结果未发现IL-4C-590T多态性与儿童哮喘显著关联,但按照性别分层后,在女童中CC基因型携带者哮喘患病危险性显著降低,OR值为0.10(95%CI:0.01～0.77)。IL-4-590CC和CT基因型与饲养鸽子或其他鸟类两因素共同存在时,有明显的正向交互作用,产生交互作用的ORint值为2.562(P=0.008),哮喘患病风险中可以归因于两因素交互作用的比例(AP)占63%,交互作用指数(S)达到1.392。IL-4-590CC和CT基因型与被动吸烟也存在正向交互作用,ORint值为1.836(P=0.064),达到临界显著性。结论在女童中发现IL-4C-590T多态性与儿童哮喘患病风险显著关联,并且该基因变异与饲养鸽子或其他鸟类、被动吸烟存在正向交互作用。

Anisakiasis caused by anisakid nematodes is an emerging infectious disease that can cause a wide range of clinical syndromes and are difficult to diagnose and treat in *** spite of their significance as pathogens,the systematics,genetics,epidemiology and biology of these parasites remain poorly *** the present study,we sequenced the complete mitochondrial(mt) genome of Pseudoterranova azarasi(sensu stricto,s.s.),which is one of the most important zoonotic anisakid *** circular mt genome is 13,954 bp in size and encodes of 36 genes,including 12 protein-coding,2 ribosomal RNA and 22 transfer RNA *** mt gene order of *** *** the same as those of Ascaris spp.(Ascarididae),Toxocara spp.(Toxocaridae) and Anisakis simplex(Anisakidae),but distinct from those of Ascaridia spp.(Ascaridiidae) and Cucullanus robustus(Cucullanidae).Phylogenetic analyses based on concatenated amino acid sequences of 12 protein-coding genes by Bayesian inference(BI) showed that Pseudoterranova were more closely related to Anisakis than they were to Contracaecum with strong a posterior probability *** mt genome provides a novel genetic markers for exploring cryptic/sibling species and host affiliations,and should have implications for the diagnosis,prevention and control of anisakiasis in humans.

Gnathostoma doloresi is one of the neglected pathogens causing gnathostomiasis. Although this zoonotic parasite leads to significant socio-economic concerns globally, little is known of its genetics and systematics. In the present study, we sequenced and characterized the complete mitochondrial(mt) genomes of G. doloresi isolates from China and Japan. The lengths of the mt genomes of the Gnathostoma doloresi China and Japan isolates are 13,809 bp and 13,812 bp, respectively. Both mt genomes encode 36 genes, including 12 protein-coding genes(PCGs), 2 ribosomal RNA genes and 22 transferRNA genes. The gene order, transcription direction and genome content are identical with its congener G. spinigerum. Phylogenetic analyses based on concatenated amino acid sequences of 12 PCGs by Bayesian inference(BI) indicated that G. doloresi are closely related to G. spinigerum. Our data provide an invaluable resource for studying the molecular epidemiology, phylogenetics and population genetics of Gnathostoma spp., and should have implications for further studies of the diagnosis, prevention and control of gnathostomiasis in humans and animals.

Background: According to the T-helper (Th)1/Th2 paradigm, it has been argued that Th1-dominated diseases such as psoriasis and Th2-dominated diseases such as asthma and hay fever should be mutually exclusive, as the immune regulatory cells, Th1 and Th2, cross-regulate each other. An increase in asthma and hay fever has occurred in Sweden over the past four to five decades, but the time trend for psoriasis is not clear. Objectives: To assess the prevalence of psoriasis in young Swedish men over a period of three decades and the association between psoriasis and allergic disorders. Methods: Register study based on data from the Swedish Military Service Conscription Register and the Total Population and the Population and Housing Censuses. Psoriasis, asthma (with and without allergic rhinitis) and allergic rhinitis at conscription were studied in 1 226 193 male conscripts in successive cohorts born between 1952 and 1977. Results: The prevalence of psoriasis was about 0.5%and it remained stable over three decades. Conscripts with psoriasis and their siblings were less likely to have eczema. The reduced risk of eczema in conscripts with psoriasis was unchanged over time. A reduced risk of allergic rhinitis was also demonstrated in conscripts with psoriasis and their siblings but only in the most recent birth cohort born between 1970 and 1977. Conclusions: A strong genetic influence may explain the unchanged prevalence of psoriasis in Swedish conscripts. Agenetic predisposition for psoriasis may confer partial protection from eczema and allergic rhinitis. The inverse relationship between allergic rhinitis and psoriasis appeared to be a recent phenomenon.

AIM:To investigate the correlation between the Helicobacter pylori(***)infection and host genetic background of healthy populations in ***:In March 2007,epidemiological studies were undertaken on the general population of a city in Indonesia(Mataram,Lombok).The participants included 107 men and 187 women,whose ages ranged from6 to 74 years old,with an average age of 34.0(±14.4)(±SD).The *** of subject by UBT method determination,and through the polymerase chain reaction with confronting two-pair primers(PCR-CTPP)method parsing the single nucleotide polymorphism of interleukin(IL)-8,IL-4,IL-1β,CD14,tumor necrosis factor(TNF-a)and tyrosine-protein phosphates non-receptor type 11(PTPN11)*** experimental data were analyzed by the statistical software ***:The *** infection rates in the healthy Indonesian population studied were 8.4%for men and12.8%for women;no obvious differences were noted for *** infection rates by sex or *** genotypes of IL-4,TC and CC genotypes of TNF-a,and GA genotypes of PTPN11,were higher in *** CC and TC genotype of TNF-a T-1031C loci featured higher expressions in the healthy Indonesian population Indonesia studied of(OR=1.99;95%CI:0.67-5.89)and(OR=1.66;95%CI:0.73-3.76),respectively.C allele of IL-1βT-31C gene locus was at a higher risk(OR=1.11;95%CI:0.70-1.73)of *** infection,but no statistical significance was found in our ***:We reveal that the association between the TNF-a and IL-1βgenotypes may be the susceptibility of *** in the studied population.
