Human bocavirus(HBoV)1 is considered an important pathogen that mainly affects infants aged 6–24 months,but preventing viral transmission in resource-limited regions through rapid and affordable on-site diagnosis of individuals with early infection of HBoV1 remains somewhat ***,we present a novel faster,lower cost,reliable method for the detection of HBoV1,which integrates a recombinase polymerase amplification(RPA)assay with the CRISPR/Cas12a system,designated the RPA-Cas12a-fluorescence *** RPA-Cas12a-fluorescence system can specifically detect target gene levels as low as 0.5 copies of HBoV1 plasmid DNA per microliter within 40 min at 37℃without the need for sophisticated *** method also demonstrates excellent specificity without cross-reactivity to non-target ***,the method was appraised using 28 clinical samples,and displayed high accuracy with positive and negative predictive agreement of 90.9%and 100%,***,our proposed rapid and sensitive HBoV1 detection method,the RPA-Cas12a-fluorescence assay,shows promising potential for early on-site diagnosis of HBoV1 infection in the fields of public health and health *** established RPA-Cas12a-fluorescence assay is rapid and reliable method for human bocavirus 1 *** RPA-Cas12a-fluorescence assay can be completed within 40 min with robust specificity and sensitivity of 0.5 copies/μl.

AIM:To provide the direct evidence for the crucial role of trimethylamine N-oxide(TMAO)in vascular permeability and endothelial cell dysfunction under diabetic ***:The role of TMAO on the in vitro biological effect of human retinal microvascular endothelial cells(HRMEC)under high glucose conditions was tested by a cell counting kit,wound healing,a transwell and a tube formation *** inflammation-related gene expression affected by TMAO was tested by real-time polymerase chain reaction(RT-PCR).The expression of the cell junction was measured by Western blotting(WB)and immunofluorescence *** addition,two groups of rat models,diabetic and non-diabetic,were fed with normal or 0.1%TMAO for 16wk,and their plasma levels of TMAO,vascular endothelial growth factor(VEGF),interleukin(IL)-6 and tumor necrosis factor(TNF)-αwere *** vascular permeability of rat retinas was measured using FITC-Dextran,and the expression of zonula occludens(ZO)-1 and claudin-5 in rat retinas was detected by WB or immunofluorescence ***:TMAO administration significantly increased the cell proliferation,migration,and tube formation of primary HRMEC either in normal or high-glucose ***-PCR showed elevated inflammation-related gene expression of HRMEC under TMAO stimulation,while WB or immunofluorescence staining indicated decreased cell junction ZO-1 and occludin expression after high-glucose and TMAO *** rats showed higher plasma levels of TMAO as well as retinal vascular leakage,which were even higher in TMAO-feeding diabetic ***,TMAO administration increased the rat plasma levels of VEGF,IL-6 and TNF-αwhile decreasing the retinal expression levels of ZO-1 and ***:TMAO enhances the proliferation,migration,and tube formation of HRMEC,as well as destroys their vascular integrity and tight *** also regulates the expression of VEGF,IL-6,and TNF-α.

We investigate the single-electron loss processes of light charged ions （Li^1＋,2＋, C^2＋,3＋,5＋, and O^2＋,3＋） in collisions with helium. To better understand the experimental results, we propose a theoretical model to calculate the cross section of projectile electron loss. In this model, an ionization radius of the incident ion was defined under the classical over-barrier model, and we developed ＂strings＂ to explain the processes of projectile electron loss, which is similar with the molecular over-barrier model. Theoretical calculations are in good agreement with the experimental results for the cross section of single-electron loss and the ratio of double-to-single ionization of helium associated with one-electron loss.

Triclosan(TCS)has garnered significant attention due to its widespread use and associated endocrine-disrupting ***,its impact on the neuroendocrine system and underlying mechanisms remain poorly ***,we established correlations between TCS exposure and serum sex hormone levels in participants of the National Health and Nutrition Examination Survey(NHANES).Additionally,we investigated TCS’s influence on the neuroendocrine system using adult zebrafish exposed to environmentally relevant concentrations of TCS(0.361–48.2μg/L)for 21 *** of reproductive and neurotoxicity included histopathological examination and behavioral ***,proteomics analyses,and biochemical detection were employed to elucidate mechanisms underlying TCS-induced neuroendocrine *** correlations were found between TCS exposure and estradiol,testosterone,and sex hormone-binding globulin levels in NHANES *** addition,TCS exposure inhibited ovary development and spermatogenesis in *** and proteomics analysis revealed gender-specific key signaling and metabolism-related pathways implicated in TCS-induced reproductive ***,TCS exposure induced nervous system impairment,as evidenced by histological changes and altered motor behavior,possibly associated with oxidative *** analysis further highlighted the potential connection between endocrine system disruption and nervous system impairment following TCS ***,this study provided evidence supporting TCS-induced endocrine disruption and offered insights into its underlying mechanisms.

Background:Evaluating the impact of digestive system diseases is vital for devising effective prevention ***,comprehensive reports on the burden of digestive system diseases in China are *** study aimed to provide an overview of the burden and trends of digestive system diseases from 1990 to 2019 in China and its ***:This cross-sectional study utilized the Global disease Burden Study 2019 to estimate the incidence,mortality rate,disability-adjusted life years(DALYs),years of life disability,years of life lost,and changes in the burden of digestive diseases across Chinese provinces from 1990 to *** analysis of disease burden primarily examines the characteristics of sub-disease distribution,time trends,age distribution,and sex ***,we compared provincial age-standardized DALYs for digestive diseases with the expected rates based on the socio-demographic index(SDI).Results:In 2019,there were 499.2 million cases of digestive system diseases in China,resulting in 1,557,310 *** cancer,colon and rectal cancer,and esophageal cancer are the top three diseases associated with mortality and DALY related to digestive system ***,cirrhosis and other chronic liver diseases,gastroesophageal reflux disease,and gallbladder and biliary diseases are the top three kinds of diseases with the highest prevalence among digestive system *** risk of gastric cancer sharply increases among men after the age of 40 years,leading to a significant disparity in burden between men and *** the SDI increased,the DALYs associated with digestive system diseases in China and its provinces showed a downward ***:Our study highlights the inverse correlation between DALYs associated with digestive system diseases and the SDI.

Hepatocellular carcinoma(HCC),a prevalent solid carcinoma of significant concern,is an aggressive and often fatal disease with increasing global incidence rates and poor therapeutic *** etiology and pathological progression of non-alcoholic steatohepatitis(NASH)-related HCC is multifactorial and ***,no single animal model can accurately mimic the full NASH-related HCC pathological progression,posing considerable challenges to transition and mechanistic ***,a novel conditional inducible wild-type human HRAS overexpressed mouse model(HRAS-HCC)was established,demonstrating 100%morbidity and mortality within approximately one month under normal dietary and lifestyle *** symptoms of HCC such as ascites,thrombus,internal hemorrhage,jaundice,and lung metastasis were successfully replicated in ***-depth pathological features of NASH-related HCC were demonstrated by pathological staining,biochemical analyses,and typical marker gene *** murine anti-PD-1 and sorafenib treatment effectively prolonged mouse survival,further confirming the accuracy and reliability of the *** on protein-protein interaction(PPI)network and RNA sequencing analyses,we speculated that overexpression of HRAS may initiate the THBS1-COL4A3 axis to induce NASH with severe fibrosis,with subsequent progression to ***,our study successfully duplicated natural sequential progression in a single murine model over a very short period,providing an accurate and reliable preclinical tool for therapeutic evaluations targeting the NASH to HCC continuum.
