Optimization of Enzymatic Extraction Process of Polysaccharides from Pseudostellaria heterophylla Fibrous Roots by Response Surface Methodology and Its Pilot Application

[Objectives]To study and optimize the process co.ditions of enzymatic hydrolysis technology for extracting polysaccharides from Pseudostellaria heterophylla fibrous roots and its application in workshop pilot tests.[Methods]*** fibrous roots were taken as the matrix material,and Box Behnken design was used to analyze the extraction time,co.posite enzyme addition amount,and liquid-solid ratio for response surface optimization experiments,and then applied to the pilot extraction of *** fibrous roots.[Results]Response surface analysis showed that all factors had a significant impact on the experimental *** optimal extraction process co.ditions for polysaccharides from *** fibrous roots were extraction time of 2.7 h,co.pound enzyme addition of 2.5%,and liquid-solid ratio of *** yield of polysaccharides from *** fibrous roots was 4.83%.The water extraction process of *** fibrous roots extraction pilot was used as the co.trol group for response surface optimization of the pilot *** optimization results showed that the extraction time was 3 h,the amount of co.posite enzyme added was 2.5%,and the liquid-solid ratio was *** polysaccharide yield was 4.75%,an increase of 4.63%co.pared to the co.trol group.[co.clusions]This paper co.ld provide feasibility for the innovation of enzy-matic hydrolysis technology for *** fibrous roots and its workshop pilot practice application,as well as a reference for the industrial application of its medicinal resources.

Screening of Anti-inflammatory Active Ingredients from Gancao Qinlian Extract and Study of Its Efficacy

[Objectives]To establish the chromatographic fingerprint of Gancao Qinlian Extracts(GQE)and reveal the possible material basis of the anti-inflammatory effect of GQE by the co.relation analysis between the fingerprint chromatographic peaks of different co.ponents of GQE and its anti-inflammatory activity.[Methods]Ultra-performance liquid chromatography(UPLC)was used to detect the different ingredients of GQE to establish its chromatographic fingerprint and analyze the differences among the three medicine co.ponents;LPS stimulated RAW264.7 cells to co.struct an inflammatory cell *** NO secretion of cells was detected by the Griess *** was used to detect the changes in TNF-αand IL-10 ***-qPCR tested the mRNA expression levels of TNF-αand *** relational analysis was carried out by co.bining fingerprint chromatographic peak data and anti-inflammatory activity data.[Results]The GQE fingerprint was established,34 fingerprint characteristic peaks were calibrated,and 33 related chromatographic peaks were screened *** co.responding chromatographic peaks in the three co.ponents were obtained,and the co.tent of the co.ponents was calculated;the anti-inflammatory results showed that the co.tent of NO,TNF-α,and the expression of TNF-αmRNA in the high and medium-dose groups of GQE were significantly lower than those in the blank group(Pco.tent and TNF-αmRNA expression in the high-dose group of GQE I was co.siderably lower than those in the blank group(Pco.relation degree of the three co.ponents was GQE II>GQE>GQE I,and the characteristic fingerprint peaks 12,15,22,23,28,31,33 may be closely related to the anti-inflammatory effect.[co.clusions]The best co.ponent of the anti-inflammatory effect in GQE is w

It is well-established that high carbonization temperature will trigger the enzyme-like activity of carbon-based ***,the catalytic mechanism is still ambiguous,which hinders the further rational design of nanomaterials as enzyme ***,N,S-rich carbonized wool nanosheets(CWs)were synthesized at different pyrolysis *** expected,only CWs treated with high-temperature possess intrinsic oxidase-and peroxidase-like ***,density functional theory(DFT)calculations demonstrate that graphitic nitrogen and the co.existence of nitrogen and sulfur in the carbon matrix serve as the active sites for the enzyme-like *** importantly,co.bining theoretical calculations and experimental observations,the high-temperature triggered catalytic mechanism can be ascribed to the fact that an appropriate high-temperature maximizes the graphitization degree to a certain extent,at which most of the catalytic active sites are well retained rather than ***,co.pling with excellent photothermal co.version efficiency and catalytic performance,CWs can be applied to photothermal-catalytic cancer therapy under near-infrared region(NIR)light *** believe this work will co.tribute to understanding the catalytic mechanism of carbon-based nanozymes and promote the development of new biomedical and pharmaceutical applications.

Pien Tze Huang(PZH),a class-1 nationally protected traditional Chinese medicine(TCM),has been used to treat liver diseases such as hepatitis;however,the effect of PZH on the progression of sepsis is ***,we reported that PZH attenuated lipopolysaccharide(LPS)-induced sepsis in mice and reduced LPS-induced production of proinflammatory cytokines in macrophages by inhibiting the activation of mitogen-activated protein kinase(MAPK)and nuclear factor-kappa B(NF-κB)***,PZH stimulated signal transducer and activator of transcription 3(STAT3)phosphorylation to induce the expression of A20,which co.ld inhibit the activation of NF-κB and MAPK *** of the bile acid(BA)receptor G protein-co.pled bile acid receptor 1(TGR5)in macrophages abolished the effects of PZH on STAT3 phosphorylation and A20 induction,as well as the LPS-induced inflammatory response,suggesting that BAs in PZH may mediate its anti-inflammatory effects by activating ***,deprivation of BAs in PZH by cholestyramine resin reduced the effects of PZH on the expression of phosphorylated-STAT3 and A20,the activation of NF-κB and MAPK signalling,and the production of proinflammatory cytokines,whereas the addition of BAs to cholestyramine resin-treated PZH partially restored the inhibitory effects on the production of proinflammatory ***,our study identifies BAs as the effective co.ponents in PZH that activate TGR5-STAT3-A20 signalling to ameliorate LPS-induced sepsis.

Novel ZIF-8/ZnS hollow polyhedral heterostructures derived from ZIF-8 with enhanced photocatalytic activity for degradation of aflatoxin B1

Myco.oxins,formed in lots of foods during the production,processing and transportation processes,are the seco.dary metabolites of Fusarium genus,Penicillium and *** B1(AFB1) is co.sidered as one of the most dangerous myco.oxins for human and animals due to its strong carcinogenicity and hepatotoxic ***,degradation of AFB1co.pletely or reduction of AFB1to a low degree co.tent has caused much *** this paper,novel ZIF-8/ZnS hollow polyhedral heterostructures were successfully synthesized by sulphurizing ZIF-8 via solvothermal *** synthesized samples were used as catalysts for AFB1degradation under UV light *** as-synthesized samples were analyzed by XRD,SEM,BET,UV-vis,ESR and so *** results show that parts of ZIF-8 can be sulphurized by TAA to synthesize ZnS and the ZIF-8 particles were etched to form the hollow *** sulphurizing degree of ZIF-8 has great influence on the photocatalytic activity of ZIF-8/*** with ZIF-8/ZnS(50) and ZIF-8/ZnS(150),ZIF-8/ZnS(100) exhibited the best photocatalytic activity and co.ld degrade 98.6 % of *** that,the reactive species of ZIF-8/ZnS(100) were co.firmed to be ·OH and ·O2-.This strategy of co.pling MOFs with semico.ductor provides a facile method to synthesize for catalyst with enhanced photocatalytic activity for degradation of food pollutants produced by aflatoxins.
