Low expression of CDK5RAP3 and DDRGK1 indicates a poor prognosis in patients with gastric cancer

AIM To investigate the effects of different levels of expression of CDK5RAP3 and DDRGK1 on long-term survival of patients undergoing radical *** The expression of CDK5RAP3 and DDRGK1 was detected by immunohistochemistry in 135 patients who received standard gastrectomy were enrolled in the study. Western Blot was used to detect the expression of CDK5RAP3 and DDRGK1 in gastric cancer and its adjacent tissues and cell lines. The correlations between the expression of CDK5RAP3 and DDRGK1 and clinicopathological factors were analyzed, and the value of each parameter to the prognosis of the patients was compared. Receiver operating characteristic analysis was used to compare the accuracy of the prediction of clinical outcome by the *** CDK5RAP3 and DDRGK1 expression was downregulated in the gastric cancer compared to its respective adjacent non-tumor tissues. The expression of CDK5RAP3 was closely related to the age of the patients(P = 0.035) and the T stage of the tumor(P = 0.017). The expression of DDRGK1 was correlated with the sex of the patients(P = 0.080), the degree of tumor differentiation(P = 0.036), the histological type(P = 0.036) and the N stage of the tumor(P = 0.014). Low expression CDK5RAP3 or DDRGK1 is a poor prognostic factor for gastric cancer patients. Prognostic analysis showed that the co-expression of CDK5RAP3 and DDRGK1 was an independent prognostic factor correlating with the overall survival of gastric cancer patients. Combined expression analysis of CDK5RAP3 and DDRGK1 may provide a more accurate prognostic value for overall survival.

BACKGROUND Because of the powerful abilities of self-learning and handling complex biological information,artificial neural network(ANN)models have been widely applied to disease diagnosis,imaging analysis,and prognosis ***,there has been no trained preoperative ANN(preope-ANN)model to preoperatively predict the prognosis of patients with gastric cancer(GC).AIM To establish a neural network model that can predict long-term survival of GC patients before surgery to evaluate the tumor condition before the *** The clinicopathological data of 1608 GC patients treated from January 2011 to April 2015 at the Department of Gastric Surgery,fujian Medical University Union Hospital were analyzed *** patients were randomly divided into a training set(70%)for establishing a preope-ANN model and a testing set(30%).The prognostic evaluation ability of the preope-ANN model was compared with that of the American Joint Commission on Cancer(8th edition)clinical TNM(cTNM)and pathological TNM(pTNM)staging through the receiver operating characteristic curve,Akaike information criterion index,Harrell's C index,and likelihood ratio *** We used the variables that were statistically significant factors for the 3-year overall survival as input-layer variables to develop a preope-ANN in the training *** survival curves within each score of the preope-ANN had good discrimination(P<0.05).Comparing the preope-ANN model,cTNM,and pTNM in both the training and testing sets,the preope-ANN model was superior to cTNM in predictive discrimination(C index),predictive homogeneity(likelihood ratio chi-square),and prediction accuracy(area under the curve).The prediction efficiency of the preope-ANN model is similar to that of *** The preope-ANN model can accurately predict the long-term survival of GC patients,and its predictive efficiency is not inferior to that of pTNM stage.

Rapid biosorption and reduction removal of Cr(Ⅵ) from aqueous solution by dried seaweeds

Four types of common seaweeds(Laminaria japonica,Undaria pinnatifida,Porphyra haitanensis,and Gracilaria lemaneiformis) were examined to remove Cr(Ⅵ) ions from aqueous *** experimental parameters that affected the biosorption process including pH,biomass dosage,contact time and temperature were investigated via batch *** surface characteristics of seaweeds before and after Cr(Ⅵ) adsorption were studied with scanning electron microscopy and Fourier transform infrared *** results show that an initial solution with the pH of 1.0 is most favorable for Cr(Ⅵ) *** adsorption is observed in the initial stage and adsorption equilibrium state is reached within 1 *** adsorption efficiency by Porphyra haitanensis is the maximum among four types of seaweed powders,followed by Laminaria japonica and Undaria pinnatifida with biosorption efficiency up to 90%.The removal rate of Gracilaria lemaneiformis is less than 60%.The kinetic data obtained using the seaweeds are found to follow pseudo-second order kinetic *** sorption data adequately correlate with the Langmuir *** indicates that amino and carboxyl groups play an important role in the process of Cr(Ⅵ) adsorption and a large percentage of Cr(Ⅵ) ions are reduced by reductive groups on the surface of seaweeds.

Nomograms for pre-and postoperative prediction of long-term survival among proximal gastric cancer patients: A large-scale,single-center retrospective study

BACKGROUND The incidence of proximal gastric cancer(GC)is increasing,and methods for the prediction of the long-term survival of proximal GC patients have not been well *** To develop nomograms for the prediction of long-term survival among proximal GC *** Between January 2007 and June 2013,we prospectively collected and retrospectively analyzed the medical records of 746 patients with proximal GC,who were divided into a training set(n=560,75%)and a validation set(n=186,25%).A Cox regression analysis was used to identify the preoperative and postoperative risk factors for overall survival(OS).RESULTS Among the 746 patients examined,the 3-and 5-year OS rates were 66.1%and 58.4%,*** the training set,preoperative T stage(cT),N stage(cN),CA19-9,tumor size,ASA core,and 3-to 6-mo weight loss were incorporated into the preoperative nomogram to predict the *** addition to these variables,lymphatic vascular infiltration(LVI),postoperative tumor size,T stage,N stage,blood transfusions,and complications were incorporated into the postoperative *** calibration curves used to determine the OS probability fit *** the training set,the preoperative nomogram achieved a C-index of 0.751[95%confidence interval(CI):0.732-0.770]in predicting OS and accurately stratified the patients into four prognostic subgroups(5-year OS rates:86.8%,73.0%,43.72%,and 20.9%,P<0.001).The postoperative nomogram had a C-index of 0.758 in predicting OS and accurately stratified the patients into four prognostic subgroups(5-year OS rates:82.6%,74.3%,45.9%,and 18.9%,P<0.001).CONCLUSION The nomograms accurately predicted the pre-and postoperative long-term survival of proximal GC patients.

A new nanostructured amino-functionalized magnetic bacterial cellulose/activated carbon（BC/AC）composite bioadsorbent（AMBCAC）was prepared for removal of Pb^2＋ and methyl orange（MO）from aqueous *** results demonstrated that the equilibrium adsorption capacity（qe）for Pb^2＋ obviously increases by 2.14 times after introduction of amino groups,the optimum p H for Pb^2＋and MO adsorption was 5.0 and 3.0,respectively,and the qeof AMBCAC was 161.78 mg g^-1 for Pb^2＋ and 83.26 mg g^-1 for MO under the optimal conditions in this *** kinetics and adsorption isotherm data of the sorption process were well fitted by pseudo-second-order kinetic model and Langmuir isotherm *** thermodynamic results（the Gibbs free energy change G〈0,the enthalpy change H〉0,the entropy change S〉0）implied that the adsorption process of Pb^2＋ and MO was feasible,endothermic and spontaneous in *** results support that the AMBCAC composite developed in this work can provide a cheap and efficient way for easy removal of both Pb^2＋ and MO as a promising adsorbent candidate for wastewater treatment.

Thermostable SOD is a promising enzyme in biotechnological applications. In the present study, ***3 was isolated from a deep-sea hydrothermal field in the East Pacific. A thermo-stable superoxide dismutase （SOD） from this strain was purified to homogeneity by steps of fractional am-monium sulfate precipitation, DEAE-Sepharose chromatography, and Phenyl-Sepharose chromatography. SOD was purified 13.4 fold to homogeneity with a specific activity of 3 354 U/mg and 11.1% recovery. SOD fromGeobacillussp. EPT3 was of the Mn-SOD type, judged by the insensitivity of the enzyme to both KCN and H2O2. SOD was determined to be a homodimer with monomeric molecular mass of 26.0 kDa. It had high thermostability at 50＆#176;C and 60＆#176;C. At tested conditions,SOD was relatively stable in the presence of some inhibitors and denaturants, such asβ-mercaptoethanol （β-ME）, dithiothreitol （DTT）, phenylmethylsulfonyl fluoride （PMSF）, urea, and guanidine ***. EPT3 SOD showed striking stability across a wide pH range from 5.0 to 11.0. It could withstand denaturants of extremely acidic and alkaline conditions, which makes it useful in the industrial applications.

Efficacy and safety of computed tomography-guided microwave ablation with fine needle-assisted puncture positioning technique for hepatocellular carcinoma

BACKGROUND In microwave ablation(MWA), although computed tomography(CT) scanning can overcome gas interference, it cannot achieve real-time localization. Therefore, the puncture technique is more important in CT-guided *** To compare the fine needle-assisted puncture(FNP) positioning technique and the conventional puncture(CP) technique for the safety and efficacy of CT-guided MWA in treating hepatocellular carcinoma(HCC).METHODS This retrospective study included 124 patients with 166 tumor nodules from February 2018 and June 2021. Seventy patients received CT-guided MWA under the FNP technique(FNP group), and 54 patients received MWA under the CP technique(CP group). Intergroup comparisons were made regarding local tumor progression(LTP), recurrence-free survival(RFS), overall survival(OS), and complications. The influencing variables of LTP and RFS were analyzed through univariate and multivariate *** The 1-, 2-, and 3-year cumulative incidences of LTP in the FNP group were significantly lower than those in the CP group(7.4%, 12.7%, 21.3% vs 13.7%, 32.9%, 36.4%;P = 0.038). The 1-, 2-, and 3-year RFS rates in the FNP group were significantly higher than those in the CP group(80.6%, 73.3%, 64.0% vs 83.3%,39.4%, and 32.5%, respectively;P = 0.008). The FNP technique independently predicted LTP and RFS. Minor complications in the FNP group were lower than those in the CP group(P < 0.001). The difference in median OS was insignificant between the FNP and CP groups(P = 0.229).CONCLUSION The FNP technique used in CT-guided MWA may improve outcomes in terms of LTP, RFS, and procedure-related complications for HCC.

To investigate the therapeutic effect of dendritic cells pulsed with lysates of heat-treated CT26 colon carcinoma cells. Bone marrow-derived DCs were pulsed with lysates of heat-treated tumor cells and were used to immunize BALB/c mice with established colon carcinoma. Cytotoxic T lymphocyte (CTL) response was detected. The therapeutic effect induced by DCs was observed by tumor weight and survival time. DCs pulsed with lysates of heat-treated tumor cells markedly induced specific cytotoxic activity of CTLs. tumor growth in the immunized BALB/c mice was significantly inhibited and the survival time of the tumor-bearing mice was prolonged. DCs pulsed with lysates of heat-treated tumor cells have an observable therapeutic effect on established colon carcinoma-bearing mice.

Subunit Characteristics of Pig Pancreas Ferritin Revealed by MALDI-TOF MS and RP-HPLC

Pig pancreas ferritin（PPF） was purified by ultra-centrifugation, ion-exchange chromatography, and native gradient polyacrylamide gel electrophoresis（PAGENG）. Sodium dodecyl sulfate（SDS）-PAGE indicates that PPF consists of two subunit types, namely, H（21000） and L（19000） subunits, and its core shows an average element composition of 1698 Fe3＋ and 179 phosphate molecules within the hollow shell, giving a 9.5：1 ratio of Fe3＋ to phosphate. An off line approach combining reversed-phase high-performance liquid chromatography（RP-HPLC） with matrix-assisted laser desorption ionization time of flight mass spectrometry（MALDI-TOF MS） made the decomposition of PPF shell into H and L subunits for the analysis of mass spectrometry（MS）, giving molecular weights of both H（21014.4） and L（18319.9） subunits. Both subunit types were further identified by an approach combining peptide mass fingerprint（PMF） with database search. A ratio of 1H to 2L subunits in PPF was determined by SDS-PAGE, RP-HPLC, and MALDI-TOF MS, respectively. It is well known that the non-covalent interaction of L-L or H-L subunits is stronger than that of H-H subunits in PPF, which may be further used to explain the unclear physiological function between H and L subunits in PPF.
