Novel substrates consisted of different fresh agro-industrial residues,their corresponding and commercial spent mushroom substrates(*** deriving from laboratory-scale experiments and SMS deriving from industrial-scale experiments by Green Zin S.A.-SMS GZ)and Pleurotus waste(PW;stipes/mishappen mushrooms)were used in blends for a new cultivation cycle of Pleurotus ostreatus and *** mushrooms in *** impact on the biochemical properties(intra-cellular polysaccharides-IPSs,protein,lipid,total phenolic compounds-TPCs,individual carbohydrates composition of the IPSs)in the first-and second-flush whole mushrooms,pilei and stipes,as well as the fatty acids composition,the antioxidant activity(in the first-flush mushroom parts)and glucan content of stipes were *** species produced satisfactory IPSs quantities in all substrates(28.69-46.38%,w/w)and significant protein amounts(18.37-26.80%,w/w).The further SMS addition(80%,w/w instead of 40%,w/w)in the cultivation substrates affected positively the mushroom IPSs values,whereas the highest protein content was detected in mushroom’s parts cultivated on substrates consisted of fresh agro-industrial ***’s lipid content was affected differently by the various substrate combinations,with SMS presence resulting in mushrooms with a lower fat content than those produced in substrates with PW *** substrates with PW and those with coffee residue were the most favorable for TPCs *** production flushes,the nutritional value of mushrooms was comparable between them,only a slight increase in TPCs of second-flush carposomes was *** was the predominant monosaccharide of the produced IPSs,combined with a significant production of total and β-*** and PW addition had a positive impact on antioxidant activity,too.A higher quantity of lipids,TPCs and significant antioxidant activity were detected in all Pleurotus pilei than stipes,whereas the latter were rich

Enhanced protective effect of selenium-biofortified peptide RYNA(Se)MNDYT compared with its native peptide RYNAMNDYT in LPS-injured murine gut microbiota

Selenium (Se)-containing peptides may be a valuable bioactive compound to promote gut microbiota-targeted therapeutic methods for intestinal disease and hepatopathy. However， limited information is available on the utilization of selenopeptides by gut microbiota， especially Se’s function. For this purpose， the present study aimed to investigate the protective effect of selenopeptide (RYNA(Se)MNDYT， Se-P2， purity of ≥95%) and its original peptide (RYNAMNDYT， P2， purity of ≥95%) in vivo by the microbiota-metabolite axis and further analyze the potential contribution of Se biofortification to Se-P2 bioactivity. The results showed that Se-P2 exhibits a higher protective effect on lipopolysaccharide (LPS)-induced inflammation than P2， including pathology of the colon and liver， which suggested that the bioactivity of P2 was promoted by the organic combination of Se. Notably， gut microbiota composition tended to be a healthy structure by Se-P2 pretreatment in LPS-injured mice， which had a positive effect on LPS-induced gut microbiota dysbacteriosis. Additionally， only Se-P2 promoted an increase in the relative abundance of Lactobacillus， Alistipes， and Roseburia and a decrease in the relative abundance of Akkermansia， Erysipelatoclostridium， and Bacteroides in LPS-injured mice. The changes in gut microbiota were obviously correlated with the changes in metabolites and affected the metabolic pathways of valine， leucine， isoleucine， phenylalanine， tyrosine， and tryptophan biosynthesis and phenylalanine metabolism. This may be one of the key reasons for Se-P2 to exert bioactivity through the microbiota-metabolite axis. Furthermore， Se-biofortification in selenium-enriched Cordyceps militaris affected the parental proteins of Se-P2 to modulate mitogen-activated protein kinase， GPI anchored protein， and carbohydrate metabolism， translation， folding， sorting and degradation， which may contribute to the bioactivity of Se-P2. Effects of imidacloprid and thiamethoxam as seed treatments on the early seedling characteristics and aphid-resistance of oilseed rape

Seed treatments with the neonicotinoid insecticides imidacloprid and thiamethoxam were evaluated to determine whether the chemicals at effective concentrations for aphid control would influence the germination and early growth of oilseed rape,Brassica *** with imidacloprid or thiamethoxam did not affect the cumulative germination rate,but significantly inhibited establishment potential by suppressing root system development in the cotyledon ***,these alterations in seedling development in the thiamethoxam-treated seeds appeared not to be detrimental as leaves developed;in contrast,for the seedlings with imidacloprid as seed treatment agent,a significantly decreased shoot/root ratio was still evident at the late two-leaf *** two leaves developed,chlorophyll content per leaf in the thiamethoxam treatment was significantly higher than that of the control,while chlorophyll content per leaf in the imidacloprid treatment remained close to that in the *** other parameters,i.e.,height,leaf area,weight of stem,leaf or root,and other growth indexes,between the treatments and the control showed no significant ***,it was found that storage time of the treated seeds had a significant effect on cumulative germination *** 30 d before planting significantly reduced germination relative to that of the *** of the plants treated with neonicotinoids were shown to have significant anti-aphid characteristics that persisted until the end of the trial.

The Angolan strain of Marburg virus （MARV/Ang） can cause lethal disease in humans with a case fatality rate of up to 90%, but infection of immunocompetent rodents do not result in any observable symptoms. Our previous work includes the development and characterization of a MARV/Ang variant that can cause lethal disease in mice （MARV/Ang-MA）, with the aim of using this tool to screen for promising prophylactic and therapeutic candidates. An intermediate animal model is needed to confirm any findings from mice studies before testing in the gold-standard non-human primate （NHP） model. In this study, we serially passaged the clinical isolate of MARV/Ang in the livers and spleens of guinea pigs until a variant emerged that causes 100% lethality in guinea pigs （MARV/Ang- GA）. Animals infected with MARV/Ang-GA showed signs of filovirus infection including lymphocytopenia, thrombocytopenia, and high viremia leading to spread to major organs, including the liver, spleen, lungs, and kidneys. The MARV/Ang-GA guinea pigs died between 7-9 days after infection, and the LD50 was calculated to be 1.1x10-1 TCID50 （median tissue culture infective dose）. Mutations in MARV/Ang-GA were identified and compared to sequences of known rodent-adapted MARV/Ang variants, which may benefit future studies characterizing important host adaptation sites in the MARV/Ang viral genome.
