Mechanisms of NLRP3 inflammasome-mediated hepatic stellate cell activation: Therapeutic potential for liver fibrosis

The liver injury leads to an inflammatory response, which causes the activation of hepatic stellate cells (HSCs) that further secrete ECM proteins and play an important role in liver fibrosis. Moreover, the inflammatory response is a driving force for fibrogenesis, which is triggered by many types of injuries. Exaggerated inflammatory immune responses are mediated by cytoplasmic protein complexes known as inflammasomes, which are involved in many chronic liver diseases. Inflammasomes are pattern recognition receptors (PRRs) that can sense any microbial motifs known as pathogen-associated molecular patterns (PAMPs), and host- or environmental-derived stress signals known as damage-associated molecular patterns (DAMPs). The inflammasomes cause caspase-mediated proteolytic cleavage of pro-IL-1β and pro-IL-18 into active IL-1β and IL-18. In this review, we provide a comprehensive summary of the important roles of NLRP3 inflammasome in the pathogenesis of liver fibrosis with an emphasis on several direct and indirect pathways responsible for the NLRP3 inflammasome-mediated HSCs activation and fibrogenesis. In addition, we discuss the general pharmacological and genetics strategies for the inhibition of NLRP3 inflammasome activation and its downstream signaling with examples of emerging pharmacotherapeutics, targeting the NLRP3 inflammasome signaling as well as a possible way to develop effective and safer NLRP3 inflammasome inhibitors.

Characterization of a new smog chamber for evaluating SAPRC gas-phase chemical mechanism

A new state-of-the-art indoor smog chamber facility(CAPS-ZJU) has been constructed and characterized at Zhejiang University,which is designed for chemical mechanism evaluation under well-controlled conditions.A series of characterization experiments were performed to validate the well-established experimental protocols,including temperature variation pattern,light spectrum and equivalent intensity(JNO2),injection and mixing performance,as well as gases and particle wall *** addition,based on some characterization experiments,the auxiliary wall mechanism has been setup and *** chamber experiments were performed across a broad range of experimental scenarios,and we demonstrated the ability to utilize these chamber data for evaluating SAPRC chemical *** was found that the SAPRC-11 can well predict the 03 formation and NO oxidation for almost all propene runs,with 6 hr △(O3-NO) model error of-3%±7%,while the final O3 was underestimated by-20% for isoprene *** for toluene and p-xylene experiments,it was confirmed that SAPRC-11 has significant improvement on aromatic chemistry than earlier version of SAPRC-07,although the aromatic decay rate was still underestimated to some *** model sensitivity test has been carried out,and the most sensitive parameters identified are the initial concentrations of reactants and the light intensity as well as HONO offgasing rate and 03 wall loss *** of which demonstrated that CAPS-ZJU smog chamber could derive high quality experimental data,and could provide insights on chamber studies and chemical mechanism development.

Foodborne pathogens continue to pose a potential food safety hazard in ready-to-eat (RTE) meat. Chlorine is commonly used to sanitize processing equipment where Escherichia coli O157:H7 (Ec) may survive and contaminate food products. The objective of this study was to characterize the survival behavior of Ec with different stresses on RTE meats. A multi-strain cocktail of Ec was pre-treated with freezing shock for 15 - 20 h and/or chlorine (0, 25, and 50 ppm) for one hour, and then inoculated onto RTE meat surfaces to obtain about 3.0 log CFU/g. Samples were stored at three abuse temperatures (12℃, 18℃, and 24℃) and Ec was enumerated during the storage. The freezing shock impact was studied using the Ec cocktail stored in a freezer overnight followed by chlorine exposure for one hour. The lag phase and growth rate of Ec were estimated using DMFit (Combase, Baranyi’s model). Results indicated that Ec growth was suppressed by chlorine treatment. Freezing shock was found to have little impact in terms of lag time and growth rate. The lag phase of Ec after exposure to 0 ppm of chlorine (50.3 h) was shorter than that of Ec treated with 25 ppm (54.6 h) and 50 ppm (164.1 h) at 12℃. However, the lag phase decreased with an increase in temperature, e.g. at 25 ppm, lag times were 54.6, 51.1 and 48.9 h for 12℃, 18℃ and 24℃, respectively. Lag times increased with an increase in chlorine concentration. At 24℃, lag times were 15.8, 48.9, and 52.4 h for 0, 25, and 50 ppm, respectively. The growth rate increased with an increase in temperature for 0 and 25 ppm chlorine levels, but decreased at 50 ppm level. Growth rate and lag phase as a function of temperature and chlorine concentration can be described by polynomial models and modified Ratkowsky-type and Zwietering-type models. Results of this study will contribute to risk assessment of RTE meats.

Previous studies have indicated an association of fat mass and obesity-associated(FTO)with nonalcoholic fatty liver disease(NAFLD),the most common chronic liver disease *** study aimed to decipher the complex role of FTO in hepatic lipid *** found that a decrease in N^(6)-methyladenosine(m^(6)A)RNA methylation in the liver of mice fed with a high-fat diet(HFD)was accompanied by an increase in FTO *** of FTO in the liver promoted triglyceride accumulation by upregulating the expression of lipogenic *** studies revealed that FTO could stabilize the mRNAs of sterol regulatory element binding transcription factor 1(SREBF1)and carbohydrate responsive element binding protein(ChREBP),two master lipogenic transcription factors,by demethylating m^(6)A *** ofeither SREBF1 or ChREBP attenuated the lipogenic effect of FTO,suggesting that they are bona fide effectors for FTO in regulating *** could stimulate FTO transcription through a mechanism involving the action of intranuclear insulin receptor beta,while knockdown of FTO abrogated the lipogenic effect of *** ofFTo by entacapone decreased the expression of SREBF1,ChREBP,and downstream lipogenic genes,ameliorating liver steatosis in HFD-fed ***,our study established a critical role of FTO in both the insulin-regulated hepatic lipogenesis and the pathogenesis of NAFLD and provided a potential strategyfor treating NAFLD.

As the gap between a shortage of organs and the im-mense demand for liver grafts persists, every available donor liver needs to be optimized for utility, urgency and equity. To overcome this challenge, decision modelling might allow us to gather evidence from previous studies as well as compare the costs and consequences of alternative options. For public health policy and clinical intervention assessment, it is a potentially powerful tool. The most commonly used types of decision analytical models include decision trees, the Markov model, microsimulation, discrete event simulation and the system dynamic model. Analytic models could support decision makers in the field of liver transplantation when facing specifc problems by synthesizing evidence, comprising all relevant options, generalizing results to other contexts, extending the time horizon and exploring the uncertainty. For modeling studies of economic evaluation for transplantation, understanding the current nature of the disease is crucial, as well as the selection of appropriate modelling techniques. The quality and availability of data is another key element for the selection and development of decision analytical models. In addition, good practice guidelines should be complied, which is important for standardization and comparability between economic outputs.

Evaluating role of bone marrow-derived stem cells in dry age-related macular degeneration using multifocal electroretinogram and fundus autofluorescence imaging

AIM：To evaluate the role of bone marrow-derived stem cells in the treatment of advanced dry age-related macular degeneration（AMD）using multifocal electroretinogram（mf-ERG）and fundus autofluorescence ***：Thirty patients（60 eyes）with bilateral central geographic atrophy（GA）were *** eye of each patient received autologous bone marrow-derived hematopoietic stem cells（BM-HSCs）（group 1）and the fellow eye with better visual acuity served as control（group2）.The effect of stem cell therapy was determined in terms of visual acuity,amplitude and implicit time in mf-ERG and size of GA on fundus autofluorescence *** tests were performed at presentation and first,third and sixth month follow *** events（if any）were also ***：At 6mo follow-up there was no statistically significant improvement in median log MAR best corrected visual acuity（BCVA）in either ***-ERG revealed significant improvement in amplitude and implicit time in the intervention group.A significant decrease was also noted in greatest linear dimension（GLD）of GA in the eyes receiving stem cells[6.78±2.60 mm at baseline to 6.56±2.59 mm at 6mo（P=0.021）].However,no such improvement was noted in the control ***：Electrophysiological and anatomical improvement in the intervention group sheds light on the therapeutic role of *** studies are required to determine the stage of disease at which the maximal benefit can be achieved and to standardize the dose andfrequency of stem cell injection.
